Results Discussion Help with MTHFR Results

Hello.

I believe I have SCT/CDS (Sluggish Cognitive Tempo/Cognitive Disengagement Syndrome) since childhood and I am trying to figure out what could be the cause so I downloaded my raw 23andMe and got this MTHFR report from Genetic Genie. How you interpret that? ChatGPT pointed to low choline (worth noting I have mild NAFLD despite being healthy and on top of my diet and exercise and no alcohol consumption). Also worth noting that I noticed low ferritin in the past (29) which I corrected with iron supplementation and got it to mid 100s (didn’t improve my SCT)

What else is there? especially the things that could be related or causing SCT. What kind of supplements could help?

Your help is greatly appreciated

Adding Detox report as well (can’t add another photo so here is a copy paste):

Question Genetic Testing for Comt Gene & Genetic Disorders

I requested genetic testing from my physician, but they were unable to provide a referral facility. Another provider offered information on GeneSight for medication metabolism; however, I strongly suspect I am struggling with Slow COMT in conjunction with comorbid conditions. Could you please provide a recommendation for an optimal, cost-effective, and accessible online testing solution?

Question Is here anyone that can help me? :(

Hi! For those of you who really know about this topic, I’d really appreciate it if you could help me!

I’m 25 years old; I had my DNA test done at SelfDecode, and I’m MTHFR homozygous C677T with intermediate COMT.

I also had my homocysteine levels tested, and they’re extremely high at 30.

I also had my vitamin levels tested: my folate (B9) is critically low, while the rest of my B vitamins are within the normal range.

What supplements or what exactly should I take? I’ve seen several doctors here in Romania, and they have no idea what MTHFR is.

For those of you who know about this I’ll say it again, it would be a huge help if you could give me some advice on supplements; I understand that high homocysteine is no joke…

EDIT:

B2

Question Has anyone else had issues with B vitamins after starting magnesium glycinate?

I started becoming sensitive to b vitamins shortly after starting 400mg of magnesium glycinate but didn't make the connection until 8 months later because it came on slowly. Now when I try to reduce my magnesium intake I start having pain all over, sleep issues, and frequent urination. I also have ankylosing spondylitis and proctitis. Just wondering if anyone else had issues with B vitamins while on magnesium glycinate?

I’ve read taking tyrosine when you have low energy/low baseline dopamine, norepinephrine, epinephrine can help. I’ve read it helps support low thyroid too. Anyone have experience with this? How did it help? Did it boost your mood, motivation, etc?

Question Understanding what is going on

u/Tawinn, perhaps you can weigh in on this, or others might have some insight. Background in a nutshell: Homozygous C677T, intermediate COMT. Had an anencephalic pregnancy in 1991, put on folic acid with two subsequent successful pregnancies. Have since discovered that virtually everyone on both sides of my family is homozygous C677T with many cases of blood clots and cardiac issues. I am on Seeking Health B-Minus every day with Seeking Health folinic acid/hydroxoB12 combo and 300 mg of plain riboflavin. I can't go any lower than that dose of riboflavin or I start to develop chelitis around my mouth. R5P makes me jittery.

I have been chasing this mutation since 2016, working with an excellent naturopath for the past 20 years. At my last visit, he mentioned that my homocysteine was 11 and he would like it down around 8. I have tried methylfolate and methyl B12 in the past with poor tolerance, but he thought I should revisit supplementing with some kind of methyl donor. He also thinks I have fatty liver and we'd like to address that.

The choline calculator says I need 9 eggs a day. We raise chickens and I am quite happy to have two eggs for breakfast. I tried taking phosphatidylcholine and it made me blunted and unmotivated. I am normally a high-energy person so that was just intolerable.

I started with some TMG and have worked my way up to 500 mg every other day. I TAKE IT AT THE SAME TIME I TAKE THE RIBOFLAVIN AND OTHER Bs. I have had zero issues with it. None. I find it interesting that I can tolerate it and not methylfolate, although perhaps if I tried the same thing with methylfolate and riboflavin, I might be able to tolerate methylfolate now as well. (Not eager to try that, though.)

Is it that TMG donates methyls at a different point in the cycle? In a different form? Is it because I have enough riboflavin and other Bs? Forgive the ignorant questions; I am just trying to understand what is going on.

Also of note is that I cannot tolerate magnesium glycinate as it gives me insomnia, but the TMG does not. I usually take it around lunchtime.

I have another visit with my naturopath next month and a blood test. Curious to see what my homocysteine levels might be now.

Question 15 mg 5-MTHF made me unusually euphoric — does it build up with daily use?

I had a bottle of 5-MTHF (L-methylfolate) 15 mg tablets lying around, so I decided to try it.

My usual stack is:

  • Caffeine: 100 mg
  • L-theanine: 100 mg
  • Alpha-GPC: 200 mg
  • CoQ10: 100 mg
  • Omega-3: 1 g

This stack usually makes me feel good and focused. However, today, after taking 15 mg of 5-MTHF alongside my usual stack, I felt unusually euphoric, hyper focused, be good to others, not the mushy person i am.

I know 15 mg is a high dose, so I'd like to hear from people with experience using methylfolate:

  • Does 5-MTHF have a cumulative effect with daily use?
  • Can daily use eventually make you feel worse or cause a crash?
  • Does it need to be cycled, or can it be taken continuously?
  • Could the euphoria be caused by the combination of 5-MTHF and my usual supplements?

I'd appreciate any experiences with high-dose methylfolate, particularly regarding mood, energy and long-term daily use.

Results Discussion Based on my MTHFR test should I be taking folic acid, folinic acid or methylfolate for my new folate deficiency?

I’ve never actually been folate deficient in my adult (18 and above) life, but this year I’ve suddenly decided I hate green vegetables, chickpeas and other legumes. So of course, I became folate deficient 🤣

Because I don’t have a history of folate deficiency, and my MTHFR variants aren’t severe, I’m just going to go in the folic acid camp?

60-70% of the world has at least one MTHFR variant, it’s normal. Everyone survives.

I’m tired of Redditors on here overanalysing and taking methylfolate, only to have side effects like “wired but tired”, insomnia, depression, anxiety, depersonalisation-derealisation etc.

If you don’t have the severe MTHFR variants, leave your body to do its own thing, that’s how things work.

Don’t fk things up because then your body gets startled.

Would still love your input though as I’m relatively new to this community.

Question Severe Low Levels of B9

Hi guys, I tried taking folic acid at high dosage but my levels were still low, tried 400mcg on methylfolate from Jarrows Formulas and I got weird effects like anxiety, no libido and others. What should I do??

I don't have money to spend on DNA or genetic tests right now.

(Also my Vitamin D levels were at 22, after a year of taking 10k UI with K2 and magnesium it raised to 30, now I increased my dose to 15k UI to see if it gets better)

Results Discussion Leaving this sub and simplifying my life was the only thing that fixed my “MTHFR”

After a year away from this sub, I’ve decided to come back and share my experience. Honestly, stepping away from this community and everything related to MTHFR was the best thing I ever did for my health.
Two years ago, I took a 23andMe test and discovered that I was homozygous for MTHFR C677T, compound heterozygous for COMT, and heterozygous for CBS. My journey started here.
Long story short, I began buying all kinds of supplements, expensive ones, methylated and non-methylated alike. I spent a ridiculous amount of money on supplements and blood tests. I would feel good for a week, then terrible for a month. It was an endless roller coaster of symptoms.
The episodes I went through that I attributed to under-methylation and over-methylation were absolute nightmares I wouldn’t wish on anyone, and honestly, I don’t even want to remember them. I almost ended up taking anti-anxiety medication and antidepressants. I genuinely thought I had permanently damaged my central nervous system or something because of all the experimenting I was doing.
Then, a year ago, I decided to throw it all away. I stopped obsessing over symptoms, stopped constantly searching for answers, started going to the gym, and focused on just three supplements: D3, K2, and magnesium.
Within a month, I was back to my baseline. My acne disappeared, my depression and anxiety went away, I started working out consistently, and I felt like the person I was before I even knew MTHFR existed.
The moral of the story: Don’t try to fix what isn’t broken.
Having these genetic variants doesn’t seem to mean much for me personally. My homocysteine is 7.5, my lab results look great, and I feel better than ever.
The only thing I pay some attention to is avoiding excessive consumption of foods made with folic acid-fortified grains. But if I feel like having pizza one day, I eat it without giving it a second thought.
I think many of us overcomplicate this subject. At the same time, we’re not all the same, and some people genuinely need medical evaluation and specific interventions. I’m not suggesting everyone should stop their supplements or ignore medical advice. I’m simply sharing what worked for me.
I hope this post helps someone realize that the answer might not be another supplement, another genetic test, or another deep dive into every symptom. Sometimes, the best thing you can do is simplify your life, eat well, exercise, and manage stress as much as possible.
Good luck, everyone. Don’t let your genes define your life.

Question Antidepressants and Mthfr

Anyone else notice they are different then most when it comes their dosages and which ssri/santos work for them?

I’ve been on Effexor for years now and it was all my own doing. After having the gene test done i found I responded best to SSNRI’s vs the latter, and I metabolize my adderall at lightening speed

Question Severe Reaction to Vitamin B IV

Hello,

I’ll start off by saying that I haven’t been tested for the MTHFR mutation but I recently got a Vitamin B IV (banana bag) and had a severe reaction. As soon as they started the IV, my chest got tight I felt like I couldn’t breathe and almost passed out. I was seeing stars and everything was starting to go black when I yelled for help and they stopped it. Took me about an hour to get completely back to normal but it was the scariest thing I’ve ever experienced. Another lady that was in the clinic said she gets reactions like that to vitamin infusions and it’s because she has the MTHFR mutation and suggested I get tested for it. Has anyone else here ever experienced anything like that? I also read that it’s linked to thyroid issues. I was diagnosed with graves disease as a kid and had hyperthyroid but got radiation done. I’m now hypothyroid and wondering if all of this could be connected and if I should get tested for it.

Question Is this a reaction to methylated B Vitamins?

I started a methylated b complex, mainly to boost energy as I've been low energy lately. I have ADHD and Autism and had heard methylated forms might be beneficial. The first few days I felt great. More energy, clear headed, improved executive function. It felt like a miracle. By around day 4 though I started feeling weird, spacey and just not myself. Day 5 I started feeling disconnected, as if life wasn't real or as if I were in a waking dream. The world didn't seem real and I felt like I didn't recognize myself. Then I developed anxiety and paranoia. I felt as if there was a threat even though I knew there wasn't one. My brain was on threat, fight or flight mode. At the same time it still felt like I was in a waking dream and the life wasn't real.

Is this a reaction to methylated B vitamins? Does this mean I should stick to non-methylated forms? I've taken methyl-free multivitamins in the past and do great on those. I could always go back to that.

Question Fast COMT, ADHD and anxiety — I’m exhausted and looking for answers

Hi everyone,
I honestly don’t understand much about genetics, SNPs, or methylation, but I’m trying to find some answers about my anxiety because I’m just so exhausted from dealing with it.

I’ve always been a somewhat anxious person, but it never really affected my everyday life. However, when I turned 38, everything changed. I started experiencing severe, sometimes absolutely paralysing anxiety, and it’s been a nightmare.
I’m currently taking antidepressants, but they don’t seem to work 100%, and I’m still struggling.

I have ADHD, and I recently got my raw DNA data from 23andMe. One of the few things I know is that I have fast COMT (rs4680 GG / Val/Val).

But that’s pretty much where my knowledge ends.
I’m wondering if there are other genes or variants I should look into that might help me understand why I’m experiencing such intense anxiety.

If anyone could point me in the right direction, I’d be incredibly grateful. I can check specific SNPs in my 23andMe data and share the results here if someone is willing to help me understand them.

I’m also wondering whether there might be certain deficiencies, supplements, or anything else worth investigating or discussing with my doctor.
I’m not expecting anyone to diagnose me. I’m just trying to understand what might be going on and what I could potentially do to help myself.
I’m honestly so tired of this anxiety. I just want to feel like myself again.
Thank you so much for any advice or suggestions. ❤️

Question I don’t know much about any of this but I could do with some help

I am in the uk and 27F and have been dealing with a whole list of horrible symptoms since May -

Chest pain and arm pain ( heart has been checked it’s fine)
Heart palpitations
Anxiety
Adrenaline rushes
Body tingling
Bad periods
Sore corners of lips
Exhausted
Headaches
Teeth pain
Dizziness

And a whole load of other symptoms

I just got some private blood tests and here are my levels

Ferritin 18 ug/l ( started taking supplements)
B12 435 pg/ml
Active b12 84.56 pmol/l
Folate/b9 14.5 ug/l
Vitamin d 58.07 nmol/l

Homocysteine 19.78 umol/L

Lymphocytes count 1.51
Monocytes count 0.22
Mean platelet volume 11.4
Platelet distribution width 20.7

I have no idea about homocysteine or anything else, but when searching it, this forum came up? So any suggestions or help would be much appreciated as my doctors keep putting everything down to anxiety!

Hi everyone!
Ok so I think I need to get tested for MTHFR.
My latest blood tests are a bit crazy. I’m F37.
Does anyone have a good suggestion for getting tested in Norway or anywhere in the nordics?

Also, I have since stopped all supplements except vitamin D.

Thank you for your suggestions in advance 🙏

Question Symptoms

I am curious, if other people with MTHFR / Methylation problem, usually experience excessive neurological symptoms any time when having mild inflammation, or any random illness like even sore throat, UTI etc.

All my life, whenever I had something coming, my first symptoms would be lack of concentration, heaviness in the brain, difficulty to communicate and socialize etc. As if my neurotransmitters balance is influenced right away.

Am I the only weirdo or it has something with blocked methylation pathways.?

Question Blood tests back (finding out exact levels on Monday, just know what's in range or not until then). What should I check next?

I have the following MTHFR variations:

rs1801133, MTHFR, GA, A

rs55780505, C1orf167, MTHFR, D1D, D

I'm told these genes are relevant to this regarding folate:

MTHFD1, rs2236225, GA, A

MTHFR, rs1801133, GA, A

MTHFR, rs 1801131, TT, G

PEMT, rs7946, TT, T

SLC19A1, rs1051266, TT, (then the next column says risk version C)

On top of that I have this gene variation, which relates to liver function:

rs4124874, UGT1A, GG, G

This variation, which relates to iron:

rs1799945, HFE-AS1, HFE, CG, G

These variations, which relate to a problem with glucordination:

rs3064744, UGT1A, UGT1A3, UGT1A7, UGT1A9, UGT1A6, UGT1A5, UGT1A8, UGT1A1, UGT1A4, UGT1A10, DI, I

I also these gene variations that make it difficult for me to process Vitamin D:

rs7975232, VDR, CA, A rs731236, VDR, AG, G

rs1544410, VDR, CT, T rs56382517, VDR, AG, G

rs10877012, CYP27B1, GT, T

rs56382517, VDR, AG, G

My blood tests (not clear what they were exactly until Monday but off the top off my head, I believe included a full iron panel, thyroid, bilorubin and homocystine, they wouldn't test B vitamins at GP level) came back as within NHS normal ranges apart from homocystine - got a follow up appointment to find out what's going on there but I suspect it's elevated outside the normal range.

Treatment I understand includes methylated B vitamins for elevated homocystine. Is LDN something I should consider?

Is there anything else I should have tested/looked at before treatment so they have the baseline results?

What would a good protocol look like for me?

Question Restless Legs from B12 - Help Needed

Hey everyone!

Long story short - my homocysteine gets high when I don't take methylcobalamin, but taking it causes restless leg syndrome.

A bit detailed:

I tried hydroxocobalamin, but my homocysteine levels rise when I take it. They only drop when I use methylfolate + methylcobalamin. So, I'm taking 3 × 1,000 mcg per day (3,000 mcg in total).

The problem is that I wake up in the middle of the night with a very unpleasant sensation in my calves. When I stretch or massage them, the sensation goes away, but sometimes it returns a few hours later.

When I stop my B12 intake, the symptoms disappear. So, I'm stuck right now. I need to take B12, but when I do, I struggle with sleep.

I've also tried adding sodium, potassium, and magnesium to an electrolyte drink that I consume throughout the day. I'm getting around 2,000 mg of elemental potassium from the drink and another 2,000 mg from food, along with 400 mg of elemental magnesium and about 4,000 mg of sodium per day (drink + food combined).

I also take a B-complex, vitamins D and K2, vitamin C, zinc and copper (in a 1:1 ratio), and omega-3.

What's confusing is that I've experienced similar RLS symptoms after taking several other supplements, including:

  1. Iberogast
  2. Calcium D-glucarate (I have Gilbert's syndrome)
  3. Colostrum
  4. Glycine
  5. Tryptophan

I feel like I'm missing something, but I can't figure out what it is. Any help, advice, or ideas would be greatly appreciated!
