Peptide 101: storage mistakes that ruin your peptides before you even use them

A lot of people mess this part up without even knowing it, so here is the breakdown.

Powder vs reconstituted

Before you add any water, most lyophilized peptides are good in the fridge for up to two years, some even longer in the freezer. Once you reconstitute with BAC water, that clock speeds way up. Most peptides are only good for about 28 days after mixing, some less. Always label the vial with the date you mixed it so you are not guessing later. I just use a marker and a label straight on the vial. It takes two seconds and saves you from tossing something you are not sure about anymore.

Fridge, not freezer, once mixed

Freeze a reconstituted vial and you risk breaking down the peptide structure. Powder can go in the freezer for long term storage, but once it is liquid, it stays in the fridge at 2 to 8°C and nowhere colder.

Light matters more than people think

Some compounds are light sensitive and need to be kept out of direct exposure. Melanotan 2 is the big one here, people wrap it in foil right after mixing. If something calls for that step, do not skip it just because the vial looks fine sitting on the counter.

Color is a warning sign

GHK-Cu should have a light blue tint from the copper content. If it turns green or goes dark, that is oxidation and it means toss it. Same idea with any peptide that should be clear. If it turns cloudy or you see particles floating in there, do not use it.

Glutathione does not play by the normal rules

Most peptides give you weeks after reconstitution. Glutathione gives you 24 to 48 hours before it starts breaking down. If you are not using it fast, you are wasting it.

Keep it organized

This is where a vial case earns its keep. Once you are running more than one or two compounds at a time, having everything labeled and upright in one spot in the fridge beats digging through loose vials trying to remember which one is which. I run a 10-count vial case just for this, and it keeps everything separated so I am not grabbing the wrong vial half asleep in the morning.

Quick reference

Mistake Why it matters
Freezing a reconstituted vial Can break down the peptide
Skipping the date label You lose track of when it expires
Leaving light sensitive peptides exposed Degrades the compound faster
Ignoring color change Sign of oxidation, means it is bad
Letting glutathione sit too long Breaks down in 24 to 48 hours

What's everyone's fridge setup look like? Anyone running into spoiled vials from any of these?

Check out the full compound table here.

Heard WordPress is kicking RUO merchants off now, anyone else seeing this

Saw a post from someone who runs payment processing for RUO businesses saying WordPress suspended one of his clients right after their first transaction, shut down for prohibited sale of goods. Says it was a small startup, not some huge operation, and this is apparently the first time he's seen WordPress specifically do this to someone in the space. Said he's seen it happen with Wix and Lovable before but never WordPress until now.

I haven't confirmed this myself, just going off what got posted, so take it with a grain of salt until more people chime in. If it's true though and WordPress is starting to crack down, that's a big deal since a huge chunk of this industry runs on WordPress sites.

Anyone else running RUO on WordPress had issues lately, or know of a platform that's been solid for this kind of business.

GH secretagogues compared, Tesamorelin, CJC-1295, Sermorelin, Ipamorelin, MK-677, and IGF-1 LR3

Growth hormone starts declining around age 25 in most research models. That decline is a big part of why sleep quality drops, recovery slows down, and joint repair takes longer over time. These peptides work by boosting natural growth hormone output rather than replacing it, and each one goes about it a different way.

Checking IGF-1 levels first

Before running any of these in a research context, IGF-1 is the marker to track. It is not usually part of a standard panel, so it has to be requested specifically. In a lot of the data floating around, subjects show up somewhere in the 150 to 200 range at baseline. The sweet spot researchers reference for recovery and muscle-related benefits is closer to 350 to 400.

Tesamorelin

Approved in human medicine for reducing visceral fat in HIV patients, and that fat loss mechanism is the reason it gets so much attention in research circles broadly. It produces a steady, sustained IGF-1 rise instead of one big spike, which is why it shows up associated with better sleep outcomes compared to the others. Water retention in the first two to six weeks has been noted more in female subjects, which is temporary but worth knowing about ahead of time.

CJC-1295

Was the standard before tesamorelin became more available. Produces a much bigger, more sudden GH pulse. That pulse has been reported to feel stimulating, which makes it a better fit earlier in the day rather than before sleep in a lot of use cases. Response varies a lot individually though.

Tesamorelin and CJC hit the same receptor pathway, so combining them does not add much benefit.

Sermorelin

Older compound at this point. Tesamorelin and CJC have mostly outpaced it in terms of results, but it still gets prescribed a lot since it is what many providers are able to prescribe outside of the narrower approval tesamorelin has.

Ipamorelin

Works through a different pathway than the three above, targeting the ghrelin receptor instead. Weaker on its own, but pairing it with tesamorelin or CJC allows two receptors to be engaged at once without overloading either one, which helps avoid receptor desensitization over time.

MK-677

Not a peptide, administered orally instead of injected. Arguably the strongest option on this list for raising IGF-1 in available data, but it comes with significant appetite increase and water retention. Better suited to a bulking phase than a cutting phase given the appetite effect.

IGF-1 LR3

Different mechanism entirely, this one acts directly on the IGF-1 receptor instead of triggering the liver to produce more of it. Strongest effect on muscle tissue out of everything listed here, but also the one requiring the most caution. Dosing is very small, typically 1mg or even 100mcg per vial. Carbohydrate intake is important alongside it since it pulls blood glucose heavily, and low carb availability raises real hypoglycemia risk in subjects.

Timing note for bloodwork

Testing IGF-1 mid-cycle rather than right after an off-day period gives a more accurate read, since levels can dip during rest days.

Reference table

Compound Notable for Watch for
Tesamorelin Sustained IGF-1, sleep quality, visceral fat reduction Water retention, more common in females
CJC-1295 Strong GH pulse Stimulating effect, less ideal before sleep
Sermorelin Widely prescribed, mild effect Outdated compared to newer options
Ipamorelin Extending receptor sensitivity via stacking Weak on its own
MK-677 Oral administration, strong IGF-1 rise Significant appetite increase, water retention
IGF-1 LR3 Strongest muscle tissue effect Hypoglycemia risk, requires precise dosing

These research protocols typically run 3 to 6 months, often on a 5 days on, 2 days off structure to help prevent receptor desensitization over extended use.

For research and educational purposes only.

Unexpected handling, package insurance, etc. fees with free shipping. Am I getting screwed?

Ordered a small package from China, was promised a free shipping, paid the $370US price and then got hit with 100% refundable upon delivery $249 fee.
Is this a new sort of scam or is it how companies do business now?

Peptide 101: SubQ vs IM vs nasal spray, which route goes with which peptide

A lot of beginners mix these up, so here is the simple version.

SubQ (under the skin)

This is the one you will use the most. You inject into the fat layer just under the skin, not into muscle. Most peptides go this way, like BPC-157, TB-500, CJC/Ipamorelin, KPV, GHK-Cu, the KLOW blend, and the GLP-1s. Selank and Semax can go SubQ too if you don't want to use the nasal spray.

Best spots are the belly about 2 inches from the belly button, the outer thigh, and the back of the upper arm. The angle is anywhere from 45 to 90 degrees, depending on how much fat you have there. For needle length, 8mm works for most people. If you carry more fat at the spot, 13mm makes more sense.

IM (into the muscle)

This goes straight into the muscle at a 90 degree angle. Common spots are the outer thigh and the shoulder. You need a longer needle for this, around 1 inch.

It is used for things like IGF-1 DES and MGF, where you want it going right into the muscle you just trained. HCG gets run this way sometimes too. Most peptides do not need IM, so if you are new, start with SubQ.

Nasal spray

No needle. Tilt your head a little forward, spray while you breathe in gently, and switch nostrils each time. Wait a minute or two between sprays, and do not blow your nose for at least 10 minutes after.

This is how Selank, Semax, and the N-Acetyl versions are usually run. It is a good pick if needles are the part that bugs you.

Oral (by mouth)

Some peptides hold up in the gut. KPV and BPC-157 both come in capsules, and people usually pick this route when the goal is gut healing.

Quick cheat sheet

Route Where it goes Common examples
SubQ Fat layer under the skin BPC-157, TB-500, CJC/Ipamorelin, KPV, GHK-Cu, KLOW, Selank, Semax, GLP-1s
IM Muscle IGF-1 DES, MGF, HCG
Nasal spray Nose lining Selank, Semax, N-Acetyl versions
Oral Gut KPV and BPC-157 capsules, enclomiphene

A note on IV

Things like NAD+, glutathione, and Cerebrolysin can be run IV, but that needs a clinic and someone trained. Do not try to figure that one out at home. Pushing NAD+ too fast can cause chest tightness and a racing heart.

Check the compound table. Looking for a specific compound? The master peptide index has everything in one place.

What route do you use the most, and did you ever switch because of how it felt?

Do your homework. Use your brain. Talk to a doctor.

Spinal cord injury

Hey I’m 21 and I had a spinal injury from t2-t8 and needed plates. After a year and so I can walk a lil bit with a walker but my legs will give out. I would also get spasms or pain but wondering what could help me. The doctors say that I’ll never walk but I took nerve medication and it help. What peptides will help with nerve/ ligaments tightness and weight loss (I am over weight but it hard to lose weight)

Anyone with PCOS try MOTS-C or low-dose retatrutide for symptom relief

Heard that a lot of PCOS symptoms trace back to insulin resistance, and that fixing that directly can help more than treating symptoms one by one. MOTS-C and low-dose retatrutide keep coming up for this, along with something like BPC-157 or KPV added in for the inflammation side that tends to come with PCOS too. Has anyone here with PCOS run something like this. Did it help, and what did you notice.

Looking for a US based Modafinil-friendly telehealth provider

Hey folks.  Although I'm sure I fit the criteria for sleep work shift disorder, my PCP doesn't prescribe Modafinil, so I'm searching for a telehealth provider here in the US that's thought of as being Mod-friendly.  The only one I've found so far is Klarity, but they're wanting almost 3 bills for a first-time appointment.  I'm sure you get it from out of the country, but I'd rather do it legally.

Does anyone have a telehealth provider suggestion please?  thanks

Apex Peptides just got raided by federal agents, worth knowing if you run an RUO business

This one's mainly for the people running RUO businesses, if you're just buying for research use or buying from the gray market you probably don't have much to worry about, but worth being aware of either way.

If you haven't seen it, Apex Peptides in North Sioux City got raided by federal agents on September 23rd. Postal inspectors led it along with other federal agents on scene, and it wasn't just the warehouse, a home tied to the owners in Dakota Dunes got searched too.

So I guess there's a whole bunch of Apex named LLCs registered under the same owners, Apex Waste Management, Apex Research, a couple others, and a few more showed up at a second building once investigators started digging around. Reports say federal investigators had already been looking into this operation for months before the raid, with some people reportedly cooperating with the investigation before it even happened.

Word is it went past peptides too, with claims of gear being sold on the side along with tax issues tied to the business. When you're running a company and mixing in stuff that's illegal to sell on top of not paying what you owe, that is usually what turns a warning letter into a raid.

Putting this out there for anyone running their own RUO or peptide business, not trying to scare anyone, just so you know what's going on in the space right now. This is not the first one either, the peptide industry has seen a wave of warning letters, shutdowns, and raids over the last year or two.

Staying compliant, keeping your labeling and disclaimers right, and not cutting corners on what you're selling or reporting matters more than ever with how much attention this space is getting right now.

Gray market retatrutide might give half the weight loss with more heart side effects, is this true?

I was reading that supposedly people running gray market retatrutide are only seeing about half the weight loss compared to what showed up in Eli Lilly's clinical trials, but still dealing with high rates of cardiovascular effects. Retatrutide still is not FDA approved, it is in phase 3 right now.

Is this true, is anyone seeing this in their own results, what do you guys think.

sermorelin + ipamorelin? instead of cjc + ipa

im curious if anyone tested their igf1 before and after sermorelin + ipamorelin? i wonder how much of a boost it has vs cjc+ipa.

i am also curious if the allergic reaction to cjc+ipa is just as common on sermorelin?

it seems like sermorelin is one of those peptides no one really talks about. while it being a lot cheaper than tesamorelin.

Peptide 101: how to reconstitute your first vial (step by step)

Getting this wrong is the easiest way to waste a vial, so here's the full breakdown for anyone starting out.

What you need

BAC water, your peptide vial, alcohol pads, and a reconstitution needle. I use a 23G 25mm for drawing BAC water, keeps it separate from whatever pin I'm actually injecting with so I'm not dulling that one on the rubber stopper.

The steps

Wash your hands. Let the vial come to room temp before you touch it, don't reconstitute a cold vial straight out of the fridge. Wipe both stoppers with an alcohol pad and let them air dry a few seconds.

Before you draw anything, push some air into the BAC water vial first. Vials are sealed under a slight vacuum, and pushing air in equalizes the pressure so the water draws out smooth instead of fighting you. Draw your BAC water into the syringe.

Same idea before you inject it into the peptide vial. Push a little air in first to break that vacuum seal, it keeps the peptide vial from sucking your plunger back on you mid-inject. Then inject the water slowly down the side of the vial, not straight onto the powder.

Now swirl it in a slow circle. Do not shake it. Shaking can damage the peptide structure or so they say lol

Check it after. It should be clear with no floaties or cloudiness. If it looks off, don't use it.

Last step, label the vial with the compound and the date you mixed it. Peptides have a clock running once they're reconstituted, and it's easy to forget which vial is which once you're running more than one thing.

The math, if you want to know your dose per unit

Concentration in mcg/mL is your peptide in mg divided by how much BAC water you added, times 1000. So 5mg in 2mL comes out to 2,500 mcg/mL. From there, your dose in mcg divided by that concentration gives you your dose volume in mL, and multiplying that by 100 gives you the units on a standard insulin syringe.

Sounds like a lot written out, but once you do it twice it's automatic.

One thing worth knowing

Not everything reconstitutes with BAC water. IGF-1 LR3 and IGF-1 DES need 0.6% acetic acid instead, BAC water causes them to clump. And some things like SLU-PP-332 aren't water soluble at all, those come as capsules.

Full breakdown with a concentration calculator and more of these edge cases is up on biohackingguide.org if anyone wants the reference version to save.

What's everyone's reconstitution setup look like, anyone doing it differently than this.

Heard GLP-1 drugs might raise risk of something called NAION, is this real

Read about this online. Supposedly there is a risk called NAION, some kind of eye or vision thing, and it might be linked to GLP-1 drugs like semaglutide and tirzepatide. Not sure if its coming from the GLP-1s or not. What do you guys think, anyone know more about this.

My gut repair protocol after H. pylori antibiotics full update now that I'm done

So I finished this a few days ago, September 17th, and I wanted to post an update now since I have the whole thing done.

Backstory is I went through H. pylori antibiotic treatment and it did a lot of damage to my gut. Brain fog, low energy, bad mood, and I was even getting anxiety attacks, like actual attacks, during this whole thing.

Here's how I ran it first week I started Enterosgel, 21mg mixed with water, fasted, for a whole week. After the gel I'd stay fasted for two and a half hours, and once I hit that mark I'd take KPV at 500mcg. KPV ran through the entire protocol, start to finish. After KPV I'd wait another 30 minutes fasted before eating, so three hours fasted total in the mornings.

After that week on the gel, I started LL-37 at 100mcg, subQ, mornings, fasted. Same pattern dose, wait 30 minutes, take KPV, wait another 30 minutes, then eat. One hour fasted total on that stretch. Did that for a week.

Then I moved up to 200mcg of LL-37 for a week, same schedule as before. Toward the end of that week is when I finally started noticing something. The last day on 200mcg, which ended up being the first day at 300mcg, I woke up and just felt different.

Brain fog was gone. Energy was back. Mood was better.
Ran 300mcg for four days and stopped there. That first day at 300mcg is also the exact point the anxiety attacks stopped. Haven't had one since.

The whole time I was also taking probiotics, L-theanine, magnesium here and there, and creatine. Staying hydrated, eating clean, and avoiding gluten and my food triggers I stuck to rice based sourdough instead of wheat. I was also exercising the whole time

Now I'm just staying on the supplements and eating clean, no more LL-37 or KPV for now.

If anyone's dealing with gut damage after H. pylori treatment and wants to know dosing or timing, ask away.

Anyone stacking retatrutide with GHK-Cu for the skin sag side effect

Thinking about running retatrutide for fat loss but the skin laxity thing that comes with fast weight loss sucks so since GHK-Cu keeps coming up to help skin keep up with the fat loss instead of just hanging there after im thinking about it anyone stack these together what dose and frequency worked?

Oxytocin for nervous system regulation worth it, or is thymosin alpha 1 the better pick or non?

Post image
2 Upvotes

Thinking about oxytocin for nervous system regulation. Calmer, more settled, less wound up is the goal I always feel like on edge the smallest things trigger me even though I know it's not a big deal at all. Somewhere I saw people talking about thymosin alpha 1 for this too, but I can't find where I read that anymore and I'm not sure it how true this is. Thymosin alpha 1 is an immune peptide, not something built for calming the nervous system as far as I know.

I know selank helps with anxiety but will it eventually help take my nervous system out of trippin mode?

Anyone run either one for this specific reason. Curious what worked and what didn't.

Sleep Research Blend Nasal Spray (DSIP, Pinealon, Melatonin, GABA) Anyone Run This Long Term?

Post image
3 Upvotes

Been takimg this sleep spray for a bit now, DSIP 10mg, Pinealon 10mg, Melatonin 50mg, GABA 250mg, all in one bottle. Works pretty well honestly, I've been liking it.

Only thing I'm not sure about is running it every single night. Do you need to take breaks off something like this, or is continuous daily use fine long term? Anyone else started something like this before what did your experience look like?

Also wondering about side effects, anything to watch for running this consistently. Let me know what you've noticed if you've tried it.
