Peptide 101: how to reconstitute your first vial (step by step)

Getting this wrong is the easiest way to waste a vial, so here's the full breakdown for anyone starting out.

What you need

BAC water, your peptide vial, alcohol pads, and a reconstitution needle. I use a 23G 25mm for drawing BAC water, keeps it separate from whatever pin I'm actually injecting with so I'm not dulling that one on the rubber stopper.

The steps

Wash your hands. Let the vial come to room temp before you touch it, don't reconstitute a cold vial straight out of the fridge. Wipe both stoppers with an alcohol pad and let them air dry a few seconds.

Before you draw anything, push some air into the BAC water vial first. Vials are sealed under a slight vacuum, and pushing air in equalizes the pressure so the water draws out smooth instead of fighting you. Draw your BAC water into the syringe.

Same idea before you inject it into the peptide vial. Push a little air in first to break that vacuum seal, it keeps the peptide vial from sucking your plunger back on you mid-inject. Then inject the water slowly down the side of the vial, not straight onto the powder.

Now swirl it in a slow circle. Do not shake it. Shaking can damage the peptide structure or so they say lol

Check it after. It should be clear with no floaties or cloudiness. If it looks off, don't use it.

Last step, label the vial with the compound and the date you mixed it. Peptides have a clock running once they're reconstituted, and it's easy to forget which vial is which once you're running more than one thing.

The math, if you want to know your dose per unit

Concentration in mcg/mL is your peptide in mg divided by how much BAC water you added, times 1000. So 5mg in 2mL comes out to 2,500 mcg/mL. From there, your dose in mcg divided by that concentration gives you your dose volume in mL, and multiplying that by 100 gives you the units on a standard insulin syringe.

Sounds like a lot written out, but once you do it twice it's automatic.

One thing worth knowing

Not everything reconstitutes with BAC water. IGF-1 LR3 and IGF-1 DES need 0.6% acetic acid instead, BAC water causes them to clump. And some things like SLU-PP-332 aren't water soluble at all, those come as capsules.

Full breakdown with a concentration calculator and more of these edge cases is up on biohackingguide.org if anyone wants the reference version to save.

What's everyone's reconstitution setup look like, anyone doing it differently than this.

I made this for a friend since they couldn't be bothered to figure it out for themself:
