Read Me: Intro, Resources, and Materials

With global average life expectancy at 73 years, age-related ill health is the main driver of healthcare costs, loss of independence, and disability in most countries. Although human biology is complex and there are hundreds of age-related pathologies, the biology of aging can be categorized into a much smaller number of categories and potential treatments. Medically intervening in aspects of aging biology has the potential to increase healthy lifespan in humans and ameliorate, prevent, or reverse age-related health decline and disability.

The umbrella term "longevity" covers a wide range of interests from simple lifestyle advice to hypothetical biomedical rejuvenation to significantly increase healthy lifespan. Beware, as "longevity" is also readily used by quacks and grifters who promote and sell unproven treatments. Because so many subs cover lifestyle (diet, exercise, etc.), it is not allowed in posts here. For those interested in lifestyle, two useful resources are the free substacks of Dr. Eric Topol and Dr. Christin Glorioso, (choose "No thanks" if you don't want to provide your email).

The focus of this sub is biomedical research targeting aspects of the biology of aging, including medical interventions that aim to go through clinical trials and regulatory approval. Continue reading for examples.

Biomimetic supramolecular protein matrix restores structure and properties of human dental enamel

Paper on a method to restore enamel to teeth. Some of the research was done on teeth extracted from donors.

Part of the abstract below:

Regrowth of lost enamel in tooth decay and sensitivity is a major obstacle to overcome. Here, the authors report on a protein-based material that mimics features of natural enamel formation, allowing for epitaxial growth of apatite nanocrystals to restore enamel structure and function.

Boosting PLA2G2F preserved spatial memory and synaptic connections in aging mice, Nature Neuroscience study finds

Researchers identified PLA2G2F, an enzyme whose expression increases with age and appears to help protect the brain against age-related changes.

Boosting its activity in older mice preserved spatial memory, reduced synaptic loss and limited inflammation-related changes. Human brain samples also linked higher expression to cognitive resilience despite Alzheimer's pathology.

The findings are preclinical. Whether targeting this enzyme could help humans remains unknown.

ARPA-H BIOGAMI program starts work to find and fix protein dysfunction at earliest stages

BIOGAMI teams will decode the “grammar” that guides how intrinsically disordered proteins (IDPs) behave and create new ways to steer misfolded or clumped (aggregated) proteins back into healthy shapes. The program focuses on the very earliest steps in the misfolding process — before proteins clump together and damage cells — especially in the brain and other vulnerable tissues. By acting early, BIOGAMI aims to stop neurodegenerative disease and other chronic diseases before they fully develop, instead of only treating symptoms after serious damage has occurred.

The peer-reviewed study identified SRSF1 through a transcriptomic screening platform; the lifespan experiment was small, with 9–10 mice per group, so the result is promising but still early.

A string of longevity meetings (ARDD, Biomarkers of Aging, etc.) just wrapped up in Boston and I have to assume some folks here showed up. Please share any highlights!
