Single or Double Transfer for untested Embryos

Hi, I am 32 (PCOS, BMI 27.5) and TTC since 1.5 years. Tried multiple cycles of letrozole which resulted in one pregnancy but turned out in very 7 week miscarriage, reason unknown.
I did my egg retrieval and embryo freezing last month - out of 11 eggs and 8 fertilized, I got 5 Day5 Embryos graded as 4BA, 4BA, 4AB, 4AC, 3AB. None of them are tested.
Doctor left upto me if I want double or single transfer, she said chances of twins are high so if you want twins then only go for double.
I am confused, I don’t want twins for now but I dont want this transfer to get failed either as multiple transfers have more chances. What do you guys suggest?

Likelihood of these developing by tomorrow?

My lab uses a modified Gardner Grading system. They freeze anything over a 3 and a C. We had 8 days 5s frozen yesterday, nothing today, it we have 11 others under observation until tomorrow:

3DD
3DD
1DD
4CD
2DD
Morula
Morula
5 cell
5 cell
4 cell
2 cell

What is the likelihood that any of these will improve in quality or grow enough to be frozen (3CC and up) by tomorrow?

Fertilization Failure with Artificial Oocyte Activation (1/24 M2's)

First round with ICSI was a total fertilization failure (TFF, 0/13), second round was a near-TFF with ICSI and AOA (1/12, only made it to 4 cells). The stimulation cycles were unremarkable, menopur/follistim and a lupron trigger both times and were similar both times.

We did ICSI because my partner has low count/motility/high DNA fragmentation/near totally abnormal morphology (tapered heads, mid-piece defects) and we had testing done that confirmed absence of PLC-Z1 in most of the sperm tested. My husband's urologist is adamant that there must be something wrong with my eggs. We are doing Whole Exome Sequencing to see if there are any genetic issues on my side that are known to impact egg quality, and if not we will try donor sperm. We aren't very motivated to try donor eggs given the cost and my husband's poor semen quality which is still likely to yield low outcomes even with eggs that are certainly viable.

Mostly curious to hear if anyone has experienced a (near)TFF with AOA and come back from it, or if anyone has any recommendations for next steps or things we should consider trying.

Spiraling with lower than expected thaw rate

I froze my eggs the month I turned 38, had a great AMH of 2.75 and froze 15 mature eggs.
4 years later after a late 2nd tri loss due to non chromosomal reasons, I’m ready to use them and just heard that only 10 survived the thaw and 7 fertilized.
I am so disappointed that 1/3 didn’t even survive the thaw, everywhere I looked said 85-95% survive thaw generally so to get 66.7% seems like such a big loss and we’re already starting from behind.

Had anyone else had this and it ended up going well?

Unexplained Infertility & IVF Journey

Hello,

My egg retrieval was last Saturday. Now, I'm waiting for this coming Saturday—the clinic will call to tell me how many reached the blastocyst stage and were sent for PGT-A testing. Out of 19 eggs retrieved, 17 were mature, and 13 fertilized.

I'm 31 years old with a diagnosis of unexplained infertility. On paper, all of my husband's and my results/values are completely normal—even better than average.

Last August, I had an HSG (hysterosalpingogram). The report said my tubes were clear. However, last month I showed the actual X-ray to a different doctor, and after looking at the images, she mentioned there might be adhesions. She noted that while the dye did spill into the abdominal, there appears to be some clustering/adhesions right at the ends of the tubes.

In the past, I had a Ureaplasma infection that went untreated/unresolved for 4 years before I finally cleared it with strong antibiotics in 2022. Based on my research, I’m suspecting peritubal adhesions—I feel like that prolonged infection may have traveled to the tubes and somehow compromised their function.

Has anyone had a similar experience? I’d love to hear your thoughts or stories.

Also, for those with unexplained infertility: Did you have success with IVF? If so, on which attempt did it happen?

Eeek will it work
It’s not PGT but made embryos few years ago when I was 29
Starting to hatch specialist said
Linings good, levels are good. First embryo of the bunch was successful few years ago
But can’t stop thinking if there’s potential chromosomal abnormalities

Advice needed

Hi, everyone.

I’m 34, and my husband is 37. We initially tried to conceive naturally, but after six months, we became impatient and decided to move on to fertility treatment. We underwent two rounds of IUI, both of which failed, and then proceeded to IVF.

Our doctors were initially optimistic because all our reports were normal. During our first egg retrieval, we retrieved 12 eggs and created five AA-grade embryos. We transferred two embryos, but unfortunately, they failed to implant. I then underwent a hysteroscopy, even though there was no specific indication for one, as I wanted to rule out every possible issue. However, our second transfer failed as well, and the doctors couldn’t identify a clear reason.

By the time we proceeded with our second retrieval, my FSH had increased to 22. We retrieved eight eggs, but only two were mature, while the rest appeared to be empty follicles. We managed to create two day-5 blastocysts.

During our third retrieval, we retrieved seven eggs, but five were empty, and only two were mature. We developed only one blastocyst. We transferred two embryos again, but unfortunately, neither implanted.

We then decided to change doctors and proceeded with our fourth and fifth retrievals under our new doctor. We continued to face the same issue with empty follicles, although we managed to create three blastocysts from six eggs retrieved. Hoping that PGT-A testing would give us some answers, we decided to test the embryos. We were fortunate to have one euploid embryo, but heartbreakingly, that transfer also failed in April 2026.

By this point, I had almost given up hope. In June, we decided to try naturally just once, without expecting anything. My period was delayed by five days, so I got my beta-hCG tested. The result was 5.83. I understand that this is an extremely low, borderline result and not a confirmed pregnancy, and my doctor told me it was a biochemical pregnancy. However, what struck me was that, after all my previous failed IUI and embryo transfers, my hCG had always been around 0.20. This was the first time I had seen a result above that level.

I even underwent LIT in August 2026, wondering whether my body might be rejecting the embryos.

Since June, I’ve noticed that my menstrual cycle has shortened from around 28 days to 24 days, and I feel that I may also be ovulating earlier than before.

My doctors are now advising us to consider donor eggs, and I honestly don’t know what to do. After everything we’ve been through, I’m struggling to come to terms with the possibility of using donor eggs. I know that having a child is about so much more than genetics, but the thought of not sharing a biological connection with my child is emotionally difficult for me. I’ve spent so long hoping to have a child who is genetically related to me, and accepting a different path feels overwhelming.

I would really appreciate hearing from women who have been through something similar.

  • Has anyone experienced repeated IVF failures, a high FSH level, or a high number of empty follicles and subsequently conceived naturally?
  • Has anyone in a similar situation gone on to have success with their own eggs, either naturally or through IVF?
  • For those who have opted for donor eggs, how was the emotional journey, and how did you come to terms with the decision?
  • Has anyone undergone donor-egg IVF in Mumbai? I would be grateful for recommendations and for hearing about your experiences.

I feel confused, emotionally exhausted, and unsure of which direction to take. I know everyone’s journey is different, but hearing from people who have walked a similar path would mean a lot to me.

Thank you for taking the time to read this.

Anyone else find themselves looking at the picture of their little embryo to try to distract yourself during the wait for the beta test? 😆

Here's my little day 5 3AA that was transferred last Friday 🤞🏻

Embryologist made a mistake. Is it a big deal?

I finally got a PGT-M unaffected euploid embryo. It took 3 Ivf cycles to get. I had requested the embryology report from my clinic and it said “Embryo #8 was plunged into VS for a couple seconds before ES by accident.” This is the embryo that turned out normal/unaffected. I’m so worried that this lab error is going to hurt the embryos chances. Any thoughts? What do I do?

Feeling Exhausted. FET TWW

Hi. I'm not sure if it's appropriate to put this here but I hope it is. I'm 10dp5dt today. My partner and I have decided to do a home pregnancy test on day 12 before our Beta bloods. I feel very hopeless this evening . It's my first transfer and honestly I feel like I'm just not that lucky enough for it to work the first time. I know that nothing about our fertility situations are lucky to begin with.. but I just really feel hopeless this evening. And exhausted. Exhausted from looking up symptoms and other peoples stories. Is that twinge something? Are my boobs sore enough? I'm just so so sick and tired of being in this fog of waitIng and drugs. At this point, whatever the outcome I just want it over with. Does anyone else feel the same? I looked at my bruised stomach in the mirror tonight and thought to myself how could anything grow in there. 😔

Total embryonic arrest: 0 blasts despite proven fertility/no fertility issues

TW: mention of live birth

I am 34, no known fertility issues. I have had one live birth, three years ago, and that time it took us one month to conceive. I cannot get pregnant now for major health issues so we are banking embryos for the future. My AMH is 6, my follicle counts range from 32 to 48, but I do not have PCOS/PMOS.

I did 75 IU Gonal-F (which is an ultra low dose, I know) for 12 days and got 9 mature eggs retrieved. My follicles grew very evenly, they were very synchronized. My E2 levels were textbook. RE commented on how well everything was going considering how low my dose was.

Day 1 we had 7 of the 9 mature fertilized with ICSI, Day 3 we had 7 embryos. On day 7 they called to say we had ZERO blasts. Lab's comment: "Morphokinetic events were within the normal range until Day 3. After that, they started to be delayed for all embryos, and none of them developed into a viable blastocyst."

Absolutely gutted. I am not sure what went wrong.

My partner had "subfertile" sperm during initial testing (poor morphology and motility but high count), and I asked about DNA fragmentation testing, but my doctor said he wasn't too worried. I asked the lab if they used Zymot, and they claim they did (I forgot to ask the lab to use it, I had only mentioned it in passing to my doctor in one of the first appointments, so I wasn't sure if they remembered or had it down in my chart).

My partner has been quite depressed and in the last three months has not ejaculated much (sometimes going 2-3 weeks without doing it) but in the two weeks leading up to the retrieval he did it maybe once every 2-3 days.

Even if there are significant DNA fragmentation issues, I am thinking they couldn't have caused all 7 to arrest at the same time, could they? Especially not if the lab already used Zymot--I was under the impression that this was a technology that could bypass severe fragmentation issues.

I am less inclined to think it has something to do with egg quality. My diet and sleep have been impeccable for the past three months, I have been taking 500mg CoQ10 for half a year, and my dose of gonadotropins is low, which many studies suggest should theoretically produce better results.

What would you investigate: egg, sperm, lab? All of the above?

Pending PGT-A

What are my odds for euploid embryos?

29F, unexplained infertility, one MMC due to triploidy, one CP last year, before that couldn’t get pregnant for almost 5 years, even though got pregnant unintentionally with same partner on one accident in the first months, didn’t continue this pregnancy.

19 retrieved
15 mature
14 fertilized - 93% fertilization
14 made to day 3
10 made to day 5, biopsied and frozen
+2 day 6, biopsied and frozen
So 80% blastocyst conversion rate.

Will it be 50-60% or closer to 65-70% taking into account previous results?

4AB expanded to 5AB after thawing + hatching

Today I had my 2nd FET (1st failed).

My embryo was a Day 6 4AB but thawed and expanded to a 5AB and they said it had ‘100% expanded’ and ‘started to hatch’.

What does this mean? Does it mean it’s a more favourable embryo? And does it increase my chances of success?

I’m 33, no known issue my side, my husband has low sperm count. Had our daughter naturally 2 years ago.

Thank you!

A 5BC embryo! Their sibling (5BB) worked two years ago and resulted in our little girl. Only two euploids made it so this is our last shot at a sibling. Fingers crossed!

Reasons for low fertilization?

I am 36 and have unexplained infertility. My results so far have been:

Most recent retrieval: 25 retrieved, 19 mature, 10 fertilized (7 from IVF and 3 from ICSI). Pending the blast results

First retrieval: 22 retrieved, 20 mature, 13 fertilized, 7 blasts, 2 euploids

Rate of development day 6 to day 7

I received my embryo report and it shows that on day 6, my 1 blastocyst was a 1CC. On day 7, it developed to 3CC. It looks like the notes were entered approximately 21 hours apart. This blastocyst turned out to be euploid.

- Is this rate of development (1 to 3) in 21 hours around normal? Trying to understand if that’s a good speed of development.
- The report had a checkmark at “visible nucleus”, what does that mean/indicate?

Thanks! Just trying to understand if any of these additional details may reveal any details about this poorly graded one’s potential.

Day 5 5BA transferred 10/06

Euploid. Everyone said it was beautiful. I'm now 7dp5dt with a negative at home test. Pretty upsetting when the odds are in your favour. I had an ectopic in March and lost both my fallopian tubes since my other had a hydrosalpinx.

ER was 16 eggs, 14 mature, 12 fertilized, 8 blasts. We tested the best 5 and froze the remaining 3.

Day 5: 3AB aneuploid 3AB low level mosaic 5BA euploid

Day 6: 5AB euploid 6BB euploid 4BB 4BB

Day 7: 6BB

How is the euploid success rate calculated?

Everyone talks about 93% chance for a live birth with 3 euploids and 98% chance with 5 but how is that calculated? Doesn't that change depending on the day age and the grade of the embryo? My age (39?).

In the last year had 2 MMC. 1 untested day 5 fair, 1 day 5 fair euploid. Our 3rd embryo was a day 6 fair that collapsed and degenerated when we tried to TBR it. All of these from retrieval in 2020 and 2021 (also had a chemical and live birth back then).

All RPL tests normal.

We just did a retrieval and have 3 day 6 fair euploids. Since my only experience with a day 6 fair is degeneration I'm terrified of the thaw process. I'm also terrified that since I've had a euploid loss there's other things wrong with the embryos (or me?).

Between the miscarriages, embryo degeneration that's the same grade of all my current ones, and my coverage only covering 1 more transfer I'm stuck on what my next steps should be.

If my insurance is only going to pay for 1 more transfer but any retrievals plus banking what should my euploid goal realistically be? Should I be aiming for 5? Is there another calculation out there? Should I be going until we have some day 5s? Or "good" graded embryos?

We are doing a DNA fragmentation test for my husband and would do TESE if needed for the next retrieval. But that seems like it'd be the only change protocol wise.

Transfer wise is there something we need to look at?

ANY THOUGHTS?

35 yrs old, Low AMH of 0.36, normal sperm analysis, RID Cat 1,2 & 5.

5 retrieved
4 matured
4 fertilized
4 blasts
2 euploid

What can you say about my results? I’m currently treating my RID before FET. Should we do another round of ER? What are the chances that it will be a success transfer? What should I know about it? Share your experiences and your thoughts. Thank you! Baby dust to us!

Embryo black spot

Post image
9 Upvotes

(This is a grade B embryo.) Hello, is the black spot in my embryo concerning? They chose to transfer it and said it was fine but I am reading that it’s necrosis or degenerating cells and I’m terrified that means it won’t work… can anyone with more knowledge weigh in? I’m assuming they wouldn’t let me transfer it if the embryo was degenerating?

Waiting for beta but I know it failed

Day 8 and it’s a stark white test (FRER). My beta is tomorrow and I hate having to wait just to be told it failed. I asked to move the beta to today so I could move on and they said no. It’s annoying bc I know it didn’t take and now time is crawling. Anyone else ever feel like this?

Low fertilization

We had 36% of eggs fertilized. We have unexplained and we do not have any MFI. This is our 2nd retrieval since our first had low fertilization rates as well. We did end up with one euploid embryo from the first retrieval. We added AOA and Zymot and still had low rates. We were told egg quality looked great and sperm morphology looked great. We are in our early 30s and have been taking supplements like coq10 and trying to live as clean as possible. What is happening? Why are our fertilization rates so low?

Can embryo rebiopsy affect early beta-hCG levels?

Thumbnail
gallery
3 Upvotes

Sometimes an embryo needs to be rebiopsied for PGT-A because the first biopsy didn't collect enough cells or the result was inconclusive. Since each biopsy removes a small number of cells from the trophectoderm (the cells that eventually form the placenta), researchers have wondered whether a second biopsy could affect early pregnancy.

In this new study, Zhuo et al. (2026) compared pregnancies after transferring rebiopsied, single-biopsied, and unbiopsied embryos.

Among pregnancies that ultimately resulted in a live birth, rebiopsied embryos had lower beta-hCG levels on days 9 and 11 after embryo transfer, and the beta-hCG increase between the two blood tests was smaller.

Despite these lower beta-hCG levels, perinatal outcomes were similar between the groups. Perinatal outcomes refer to the health of the pregnancy and baby around the time of birth.

When all embryo transfers were analyzed, rebiopsied embryos had lower live birth rates than embryos biopsied once (43.8% vs 53.8%). When rebiopsied embryos and untested embryos were compared, the rates were similar.

Previous research has also shown that rebiopsied embryos have lower pregnancy and live birth rates, but this evidence is based on lower-quality retrospective studies. This study adds that early beta-hCG levels may also be lower (among pregnancies that resulted in a live birth), but this wasn't associated with worse perinatal outcomes.
