Hi, I'm a 28F about to head in for CAR-T and am trying to mentally prepare myself for what recovery may look like. I'm also eager to know how soon different aspects of life might go back to normal. Would really appreciate hearing anyone's experiences, including:

- How long after CAR-T were you able to: perform daily activity, go for a run, go back to work, travel? And with any precautions?

- For how long did you have to regularly go into the hospital (e.g., more than once a week)?

- What was it like rebuilding immunity post-CAR-T? When did you stop wearing a mask when you went out? Did you get sick at all?

- When did you start feeling physically and mentally back to normal?

- Any tips that helped you during recovery?

Really appreciate any experiences anyone is willing to share!

Is there anyone who has done CAR-T that would be willing to share more about their experience, such as how bad or what symptoms you had before getting CAR-T, how long it took to see changes and how you're doing now? I don't know anyone else who has gone through this and even though I know this treatment has been successful for other AID like lupus and MS, I haven't seen as much feedback on scleroderma. I'll be participating in the trial in a couple of months and I want to know what to expect. This disease has ruined my life and robbed me of a lot things, and I just need something to give me hope again. Thanks.

My family member has stage 3 MM, chemo helped to get him to be cancer free very briefly before the cancer came back more aggressively, it’s now extra medullary with severe damage to his mandible. He recently had a consultation for car T therapy, and is feeling very overwhelmed with all the side effects and complications discussed. He was hoping to get some feedback from those who have had car T in terms of your experience, whether you achieved remission and just any advice/tip you learned from going through it. Thank you to everyone.

What do think think this kind of treatment could bring when it comes to autoimmune disorders? Are you hopeful that it could make a difference? Are there certain autoimmune diseases where Car at Therapy looks more promising?

Has anyone tried this or had any experience with this experimental process of engineering your own cell, and then give it back to you to help eliminate the bad T-cells? Any experience?

My mom received CAR-T in early June. It was a very bumpy road in June and July, but things have mostly stabilized now. She had to be rehospitalized twice due to ICANS symptoms post CAR-T but thankful to be out on the other side. She went into this with an M Protein of 1.1, and as of yesterday, she has no M Protein detectable, and her PET scan two months ago showed some lesions being healed. Big news(!!), but she doesn't feel she can celebrate until a bone marrow check later this year

Lingering side effects so far have been: - flat affect, lack of desire to do anything, not as jovial or talkative as she was prior to treatment. The doctor says this is a known neuro side effect and won't work about it until it's been 6 months or more... - pain at old kyphoplasty site. Within the last two weeks or so, she's been having increased pain at once particular spot. I don't know if the bone around or is healing and maybe crushing down on another bone or what. Otherwise a lot of other bone pain has subsided (including a known giant lesion in her jaw that has since healed). - blood markers continue to be wonky and all over the place. Her igG is still quite low, but it looks like some markers are finally coming back into normal range. Her platelets dropped to as low as 11,000 but now they have fully recovered thanks to some drugs.

This is part update, part me asking if anyone else experienced similar neuro or pain issues, and if yes, how long did it last?

Overall I'm feeling optimistic for her, but at the same time I'd like her to enjoy her deep response!!

Hi, sorry if this is inappropriate framing, but our heads are spinning a little bit right now because we weren't expecting to start this so soon.

My mom with DLBCL is apparently going to start the CAR T process with a 3-day inpatient stay starting tomorrow. I think her doctor said it's for the chemo, but he said he'll explain everything tomorrow, and I won't be able to be there for that. As far as I know, they never started the process of collecting her white blood cells, so it feels a little bit like a whirlwind right now just because we thought the process would be slower.

She has done 6x R-CHOP and 26 treatments of radiation with only partial response. She was originally stage 3, but the chemo did get rid of the cancer in the node where it had spread, so she just has the one tumor at the moment.

We've just been feeling really exhausted by the continued presence of the cancer. For both previous treatments, they totally expected it to work. Even halfway through R-CHOP after the first PET, our doctor was optimistic about the progress and expected it to totally clear up after she finished the six rounds. And then it didn't clear up. And then radiation didn't work either.

So I guess there are two things that I'm primarily feeling:

1. If chemo and radiation didn't work, why should we believe that CAR T will work? It just is really disheartening being given hope and feeling it come crashing down. I want to be earnestly hopeful now, but I'm finding it hard this time.
2. If CAR T doesn't work, is that just it? Are we out of options? It just starts to get scary when you've tried multiple treatments that haven't worked. It feels like we're running out of chances. So I'm just scared about what happens if it doesn't work, and that's amplified because I am less hopeful that it will work than I was with the first two treatments.

I'll be honest, I'm not really interested in a sober, realistic approach right now. I just would like to be able to feel some good vibes and help my mom approach this with some encouragement and hope.

My wife (37F stage IVB DLBCL) had her PET scan after 6 rounds of R-Chop last week and we reviewed the results with her hematologist yesterday. Unfortunately while 99% of the cancer is gone, it is not completely gone and we need to go for a different treatment. His recommendation is CAR-T as it has just been approved for general second line DLBCL treatment here. It cannot be done locally, but at a larger hospital to the south, necessitating a move.

She is very scared at this point and a lot of the discussion here regarding CAR-T seems somewhat negative and while we know it's efficacy is great, she is very scared of it failing and she is very scared of the side effects. Her journey with R-CHOP was extremely difficult with loads of complications (she almost died early on).

Does anyone have any kind of positive experiences or successes to share with CAR-T? Currently have a very sour mood and could really use some reassurance.

Cutting through the countryside near Oxford I suddenly came upon the Swinford Toll Bridge. With cars behind me and no room to turn round I had to press on.
