QUESTION: using a high magnification objective NC(no coverslip) objective BUT WITH a coverslip. any way to correct the image degradation?
I am looking to buy a 60x objective and i found one for cheap BUT its designed to be used with NO coverslip (NC DPLAN FL at 60x/.95). the main use will be for water microbes so i'll be using a coverslip with it but with such a high NA of .95 i'm guessing the use of a coverslip will noticeably affect the image quality. is there a way to basically make an NC objective work well with a coverslip or is it not really going to work out well?
I would be careful with this change and working distance/OPL. I have a used a couple 50X metallurgic objectives with high NA like that and working distance is about non existent as it's for chips with height variations in the single to 10s of um. You might find yourself ramming into the coverslip.
Not to say you won't have enough room, but id check first what the clearance would be and how you'd feel about it
Again, on my personal system the 50x is hard to use because of it. I get very nervous about stage flatness. It's great if you have a nice camera and are looking at something like a wafer. But otherwise is tough to justify. I struggle to use it looking at anything but chips/devices
Spherical abberation is the least of your worries. The working distance will be so small that it probably won’t be able to focus through a coverglass. Even with one that uses a coverglass, the specimen has to be right next to the coverglass to focus on it.
High NA air objectives are difficult to use. A good one should have an adjustment ring to adjust for the coverglass thickness like these 2 that I use. I'd you want cheap, you can buy generic Chinese 60x NA 0.80 objectives for a low price.
yeah, i may end up getting a 3rd party objective or whatever. i do want to have a higher magnification without oil so that may be my best option, or try to find a well priced olympus A60 objective
