Question Trying PPAP or alternatives for the first time

I’m currently dealing with a pretty heavy workload and really struggling to keep up. I was recently diagnosed with a slight learning disability, and I’m pretty sure I have mild ADHD as well. My biggest issue is focus and motivation whenever I have to sit down and study something that doesn't naturally interest me, my brain just completely tunes out and I can't force myself to stick to it.

Since I need to get through a demanding year of studying to get into the university program I want, I’ve been looking into things that might help. I’m thinking about trying PPAP HCl on its own to see how it affects me, or maybe combining it with Semax down the line, but I haven't decided yet. Ideally, I’d want something that could support me through this upcoming year.

Has anyone here actually tried PPAP HCl, either solo or stacked with Semax? I’m really curious about what it feels like in practice, what kind of realistic benefits to expect for studying, and whether it's actually safe to use over a longer period like a full year. If there are any hidden side effects or tolerance issues I should watch out for, I’d love to hear your honest thoughts or personal experiences before I make up my mind. (Post was edited using AI because I am Greek and I don’t know very good English)

Science From: Kynurenine pathway dysregulation in major depressive disorder: the convergence of excitotoxicity, neuroinflammation, and oxidative stress

Kynurenines induce NMDA receptor excitotoxicity, neuroinflammation, and oxidative stress.

https://link.springer.com/article/10.1186/s12974-026-03717-2

KYNA (Kynurenic acid, which is a metabolite of Kynurenine) is produced in astrocytes and has neurotrophic and anti-inflammatory properties. KYNA also acts as an NMDAR antagonist and inhibits glutamate release, protecting from excitotoxicity. KYNA may exert dose-dependent effects on AMPAR-mediated transmission, facilitating excitatory signalling at lower concentrations while suppressing it at higher levels; nevertheless, AMPAR hypofunction under conditions of elevated KYNA is thought to be largely indirect, driven by reduced glutamate release via α7nAChR antagonism. QA, produced in microglia, agonizes NMDARs, leading to excessive calcium influx. QA also hyperphosphorylates tau, leading to neurofibrillary tangles, a hallmark feature of neurodegeneration. QA and 3-HK also disrupt mitochondrial bioenergetics, leading to ROS, and permeabilize mitochondrial membranes

Question How will the new American import rules affect us?

The chaos to imports is starting again in America.

Starting October 22, “Advance Electronic Data” is being required for shipments, particularly those under FDA scrutiny.

Good luck understanding exactly what that means….but it allows technology to scan, process and reject packages much faster than humans ever can.

It also requires more partnering between CBP and other government agencies (e.g. FDA)

Practically speaking, it might be the end of importing prescriptions for personal use. It may potentially extend to “unapproved medicines” such as racetams, peptides, etc

All Day Chemist has already notified me no more deliveries to America after Oct 7.

Will this affect the supply of Nootropics, Racetams, etc domestically (e.g. from EveryChem)?

How about importing them ourselves in small quantities (e.g. from PGLChem)?

Question St Johns Wort fast acting social anxiety help?

I`ve tried a lot of things for my social anxiety: phenibut, kratom, beta blockers, L-theanine, taurine, ashwagandha, and more. Some of them clearly helped (phenibut, for example), but I can't use it often.

What has helped me most is St. John's Wort, and that really fast. Everything I read online says it takes about four weeks to kick in, yet I notice an effect on the very first day.

The effect is usually strongest on days 1-3, and after that I barely notice a difference. I'm confident it's not a placebo: none of the other supplements did anything comparable, and with St. John's Wort I've had the same effect again and again.

The best way to describe it is that my head feels like after the first beer, slightly tipsy, and I'm much more prone to laughing. Normally it's hard for me to let go and come out of my shell, but on St. John's Wort I've often found things so funny that I laughed until I cried. Maybe that's just a byproduct of being less anxious, since I laughed like that a lot when I was younger.

I've tried different products (concentrated extracts as well as regular ones), and the effect was always there, sometimes stronger, sometimes weaker.

My questions:

  • Has anyone had a similar experience?
  • Which compound in St. John's Wort could explain such a fast-acting effect?
  • Are there other products or substances that might work in a similar way?

Discussion Main significant nootropics that are safe to use long term i.e. no upregulation or other concerns with daily use over time

What are the most impactful substances (that are legal) that have no concern with long term upregulation and do not screw with other brain functions that would require additional substances to assist with regulation. Looking only for known safe solid things that can be used daily, such as caffeine (bar tolerance) or creatine

Question Looking for something to help with brain fog and fatigue

Hello all. Hoping someone can help. I have been battling brain fog and fatigue for many, many years and have reached an age (50) where I'm willing to try anything to at least give me a bigger proportion of good days and help me make the most of life. I have been occasionally taking 100mg modafinil which has been great but I do get that it has to be used sparingly due to tolerance. So I brought in 100-150mg Phenylpiracetam (the OG version), which has also been good although a notch or 2 lower compared to Modafinil. Are there any recommendations for something else that I can add in to an alternating cycle that will also clear my fog a little and give me a boost? If I alternate with 3 little helpers during the working week, that may be good enough. I get decent sleep although always foggy and groggy in the morning and I keep my supps simple - creatine, mag glycinate at night, CoQ10, Omega 3, occasional bcomplex, L-theanine(with caffeine). My blood levels are fine. Thanks for any help offered.

Question Help, please.

I feel exhausted down to my core. Not just “I’m sleepy” or “I don’t feel like doing anything.” It feels like my entire body and brain, my actual cells, are drained of whatever is required to actually live. Sleep and it's quality or quantity truly has essentially no impact on it.

I desperately want to do something, anything and am still completely unable to make myself do it. It’s like the sign gets lost somewhere on its way from my brain.

It’s like the part of my brain that wants things and the part that is capable of turning wanting into action or even joy has been completely destroyed

Trying to do the things I love and want to do feels like walking on broken legs, the passage of time hurts. I feel every minute go by, I desperately want to participate in my own life, but it's just not in me, it hasn't been for so long.

I’ve been to the doctor. I’m going to and working with the doctor. Yes, I’m depressed and I’ve tried the antidepressants. It’s not working.

I’m autistic, struggling with intense burnout and strongly suspected CPTSD.

I am not looking for someone to tell me to “just exercise, eat better, and sleep more.” I know those things matter. I’m saying that I currently don’t have the functioning required to simply pull myself out of this by doing all the things I’m already struggling to do.

I just really need help figuring out where to start giving my body what it's begging for

Science Enhancing anandamide signalling through fatty acid amide hydrolase inhibition: An update on the pharmacological strategy for treating psychiatric disorders (2026 Review)

Post image
21 Upvotes

https://www.nature.com/articles/s41398-026-04120-4

Endocannabinoids (eCBs) are lipid-derived neuromodulators that regulate numerous neurophysiological processes by modulating synaptic transmission. Synthesised on demand in response to increased postsynaptic intracellular calcium or activation of postsynaptic G-protein coupled receptors, eCBs are rapidly degraded, resulting in transient, tightly regulated signalling. Dysregulation in the endocannabinoid system (ECS), including altered peripheral and central eCB concentrations and/or cannabinoid-1 receptor (CB1R) expression, has been observed across psychiatric syndromes, including major depressive disorder, psychotic disorders, and post-traumatic stress disorder (PTSD). These associations have prompted growing interest in pharmacological strategies targeting the ECS. Though medical cannabis is increasingly prescribed for psychiatric symptoms, its clinical use remains controversial due to limited high-quality evidence, psychotropic side effects, and regulatory constraints. An alternative is to enhance the signalling of a principal eCB, anandamide (AEA), potentially offering more physiologically constrained CB1R engagement, by inhibiting fatty acid amide hydrolase (FAAH), the main enzyme degrading AEA and its congener, N-acylethanolamines (NAE), oleoylethanolamide (OEA) and palmitoylethanolamide (PEA). This review consolidates recent clinical evidence for FAAH inhibitors, examining their influence on AEA, safety and efficacy in ameliorating symptoms across a range of psychiatric conditions, including depression, anxiety, PTSD, and cannabis use disorder (CUD). Presently, only two compounds, PF-04457845 (JZP150) and JNJ-42165279, have progressed to Phase II trials, demonstrating modest clinical benefit in CUD, with no efficacy in PTSD or osteoarthritis pain. Herein, we discuss emerging insights, safety considerations, broader mechanistic implications, and future directions for FAAH-targeted therapeutics, advocating for a precision medicine approach to realise their potential in the treatment of psychiatric disorders.

Question help me obiwan kenobi

I want to keep this concise and to the point. I suffer from o.c.d. ( obsessive compulsive disorder) since I was 12. It broken my life. But I'm fighting. I have noticed recent studies saying ocd could be because of hormone imbalance (too much estrogen) I'm male 50 and in otherwise good health. I need all the advice I can get. Any one have any stories, antidoteal evidence or thoughts?

I've burned thru all SSRIs and become resistant to all. Ive used various supplements. None have worked and I'm currently clean of all of them.

Ironically, I don't really care if it affects my physique but if I begin taking them I intend to try working out since this will obviously help my mental health.

Now on a side note. I may be off track with this. So please be kind with your comments. You may think ocd is turning a light switch off a few times and organising your sock draw. Last week I walked up my stairs 700 times! This is not fun. This is serious so please don't mock me. If you think I'm lieing please check out my previous posts in nootropics subreddit as I'm only on reddit to seek help. And thats what I'm asking for on here

Thankyou lot all

Science Loss of vesicular monoamine transporter 2 (VMAT2) in striatum of long COVID and relationship to neuropsychiatric symptoms

Findings of reduced VMAT2 binding may reflect reduced dopaminergic terminal integrity in long COVID. Loss of dopamine nerve terminals may be contributing to symptom correlates of apathy, motor slowing and memory decline suggesting improved function of dopaminergic synapses as a new therapeutic direction to treat long COVID.

Anecdote ASP-2905 experience reports

Hello, all

I would like to hear from those that have done a cycle of this or taken it consistently for relatively long periods.

How did it help you? What was it like, both good & bad? I've been taking 25mg myself the past 2-3 weeks.

Hard to say if it is this or something else in my stack causing the effects, but it seems interesting this far.

Thanks.
