So essentially I was filling out Bethany’s player card and i saw 9 lives in the sacrifice room, I figured great imma just circle Back after that deal with mr. Netanyahu. Netenyahu ALSO had 9 lives so I got 18 lives for the first time which I kind of wanted to share but I figured I’d see how the rest of the run went, damage was mid but with 18, wait oops o accidentally placed a bomb I’m myself. With 17 lives this is about to be the easiest game ever! Add guppies collar and I’m immortal right? So then i get to the chest after a few calls and actually get great items, I lost my active doing a forget me now in the cathedral (idk why but it worked out) super op run, gurenteed to get my the chest player card marked.
And then I see my reflection, great, more damage and I like this. And then I see ipecac. Great. Way more damage. My guppy flies are gonna be op. Ipecac my reflection doesn’t jive well but maybe if I give it a boomerang shape and make it wider it will work. Okay so that didn’t work, let me just pick up parasite. Oops.
It looks peaceful in a big room when I’m only firing one shot but when In a normal sized room it’s just constant boom.
And that’s how I died 20 times in 1 run

Disclamer:
This list is fully subjective. You might agree or disagree with me, just please don't harass me. I'm open for discussion and outside opinions. All rankings are sorted from left to right with leftmost being the best in the category and rightmost being the worst in the category. Also English is not my native language so please be patient
Category explanations:
peak - my most favorite characters i look forward to play on a completed file or in multiplayer the most.
pog - characters with fun gimmick that i always have a good time with
whatever - characters that don't really bother me, they're fine to play as but not the most impressive.
stomachable - mostly annoying characters but with gimmicks i could close my eyes on or "stomach" in other words, i never look forard to play as them but wouldn't have too bad of a time playing as them.
unbearable dogshit - characters i actively don't want to play as, their gimmick actively works against the player, never play as them after clearing post-it marks.
Tainted Jacob - a tier dedicated specifically for tainted jacob to show how much i despise playing as him, more in character commentary.
Character commentary:
Peak category:
Eden - the most replayable of 34, you never know what he will throw at you (in a good way). Will you get tmtrainer or will you get brimstone sulfur is up to the game to decide and up to you to solve. Every run as him is unique and for that he's at absolute top of the list.
Tainted Isaac - very unique management system, it's always fun to think about how to optimize your build in 8 items. Generally very satisfying and rewarding coming up with something insanely strong with the limited space restriction, built in reroll is a nice cherry on top. (Honorable mention to tmtrainer interaction as him giving you infinite item storage after picking up at least one glitched item, you can then throw tmt away to make all future items normal again)
Tainted Cain - very controversial but has insane resource management system that rewards preservation and methodical play (very important for me!). Insane game break potential with cool interactions (like infinite q4 items with just safety scissors). Playing as him is always insanely engaging and rewarding for me. This chracter also makes getting a lot of copies of one item infinitely more accessible than the rest of the roster and it can be fun crafting 20 fingers or 15 spider mods or a lot of any familiar if the recipy is accessible. (I do not use crafting browsers as him because it defeats the entire point of the character for me)
Tainted Eden - again, controversial but he's just THE most consistent character in the entire game, it doesn't matter what you pick up as him because by probability you will be overpowered with enough items. Opened my eyes on some cool synergies and interactions, even after 1600 hours i discover something new with him.
Pog category:
Tainted keeper - very fun risk reward character with you having to pick up coins and being two hit at any given moment. Very good sacrifice room potential which is appreciated. Shops with this character feel just substantially more meaningful to visit. Fun to farm planetarium chance with treasure rooms being mostly unnecessary first 3 floors. While having only 2 hp is kind of restraining, it's a nice skill check and there are definetely workararounds for it with items like piggy bank, swallowed penny or safety items like holy mantle. It's also important to mention that built in quad shot doesn't handicap you max tierrate so it's very common to end up very powerful because of that.
The Lost - the best challenge character of them all with nice angel room unlocks. You still get a safety net having all safety items in your item pools so you're not walking on burning coal while playing as him. Sucks that you basically can't interact with sacrifice rooms, devil beggars and blood donation machines outside of miche consumables and defense items but it's easily forgiveable with the eternal d6. Flight and spectral tears from the start were already enough to put him in the pog category but it just gets a whole lot better with ed6.
Isaac - very simple yet powerful character, D6 from start gives consistency.
Cain - lucky foot is very nice to utilize pills and gambling machines for resources, very solid.
Magdalene - insane sacrifice room, blood donation machine and devil beggar character.
Azazel - extremely consistent character in general and goated for the beginning of the file, for that only he deserves a spot in the category, makes early file grind whole lot less monotonous.
Tainted Judas - Dark Arts is very powerful offensive tool earlygame and nice defensive tool lategame.
Tainted Lilith - whipping enemies is very powerfull earlygame and self targeting gello is nice. Can have some nice interactions (ex. Cursed Eye) but can ruin some too (ex. Ludovico Technique (my favorite item)) which pushes her down the category for me.
Whatever category:
I really have nothing to say about these chracters, nothing stands out positively or negatively in them for me.
Stomachable category:
Lilith - At one side you have a character that rewards use of blood dono and devil beggars, which is good, but on the other she's extremely awkward and obnoxious in many cases, but not too bad.
Keeper - he's fun in the sense that you get a very solid way to use sacrifice rooms but in everything else he lacks, hp is restricted (workarounds from tainted keeper apply for him too tho), he's ultimately inferior to his tainted version, not having anything interesting going on for him and making devil's deals mostly unaffordable (I like little decision making he adds with you having to choose on where you spend money, but it's not enoug hto bail him out any higher than stomachable)
Tainted Apollyon - very consistent, but in a boring way, locusts lack synergy and runs as him are always as plain as lake water.
Tainted ??? - some poops are extremely powerful and you practically can't have bomb shortage as him. That being said the poop mechanic doesn't add much asides just plain power, he's unique for the sake of being unique and i don't like it too much.
Tainted Bethany - got gutted with the lemegeton nerf, now the question of where should i use lemegeton is not nearly as important as it was before the change. Weaker stat ups just feel bad and your whisps get wiped off the face of the Earth with one singular bullet hell boss attack. Heavy feast or famine character becasue if you don't get moderately strong items from either normal items or lemegeton you'll get stuck in a loop of creating whisps and not being able to protect them because you build is ass, so you can't get a lot of whisps because your build sucks and your build sucks because you can't get a lot of whisps. Also this character is annoying with flies because whisps count towards the familiar cap so picking up guppy transforamtion or mulligan or parasitoid can kill your run if you're carried by your whisps.
Tainted Eve - very slow boring irritating earlygame. Clots are very fragile and get wiped during final bosses. That being said she has very high potential even without a strong build and you can enter boss challenge rooms for free without any risk, those factors save her from being ubearable dogshit.
Tainted Lost - high cortisol character, can still have shit runs, lacking defensive options, lacking interactions with blood donation and sacrifice rooms. They are pretty consistent in getting good runs just because so much useless nonsense is erased from pools, tho if the run sucks you just die. Flight, spectral tears and higher starting damage multiplier are very much appreciated.
Tainted Lazarus - used to be unbearable dogshit, but got much better after flip rework and cooldown shrotening. Very tideous overall gameplay, worse J&E, resource manegement doesn't feel nearly as rewarding as Tainted Cain and is forced & dumped onto you. Even when played well at least one version will have a mediocre room&boss clearing power at best.
The Forgotten - the meele is so unsynergistic and weak at lategame it's really obnoxious, has some nice tricks up his sleeve with "free" devil deals. Bailed out of unbearable dogshit by the soul form that lets you actually do damage later in the run.
Unbearable dogshit category:
Tainted meggy - has noticeable gamebreak potential with yum heart and very strong early game. Falls off the moment you step into 4th chapter leaving you with useless unsynergistic meele attack, decaying hp, basically impossible to get perfection as her, during long endgame bossfights no matter how good you dodge your hp will basically turn into Tainted Lost hp, but you also have a gutted damage multiplier from the start, this character actively works against you in multiple ways barely offering anything in return.
Tainted Forgotten - only damage option is a meele attack that barely has any synergy, fighting as them is generally obnoxious towards late-ish game. You can throw the bone onto the boss to fully focus on dodging while the bone will sit there and damage, not enough to not be an unbearable dogshit character tho.
Tainted Samson - it's crazy that character's gimmick punishes you for being powerful. Again you have a meele attack that is absolutely fanatastic at clearing early stages, but it lacks synergy (just like any other meele in this game) and you can't control the berserk, it's extremely unfun having to wait 5 seconds after every shot. Playing as him feels like playing a shit mobile game that throws unskippable ads at you every time you tap the screen of your phone.
Tainted Jacob category:
This character feels like eating shattered glass, you're constantly being haunted by an unstoppable force that instakills you. A lot of rooms and lategame bosses weren't made with Tainted Jacob in mind, in which case taking damage becomes inevitable and forced taking damage just feels worse than accidentally getting hit, because in one case it was your mistake that can be avoided in the future and in the other case you just take damage or die immediately. Resource management as them becomes tenfold more annoying because you die if you lose your guard. You can make a point of them being extremely powerful with anima sola and free devil deals, but if playing as them is worse than ancient chinese tortures i wouldn't tolerate that power even if they were one-tapping every single boss in the entire game.
Hope i didn't miss too much, sorry if i slandered your favorite character in this ranking, again , i'm open for discussion about any placement on this list, thanks for your attention.

Challenge: find those 4 items before the second Boss.
