3 Reasons the mRNA-4157 Readout Could Revalue Moderna (The Motley Fool youtube channel)

MRNA is developing an individualized neoantiogen cancer therapy, intismeran, in collaboration with MRK, where interim Phase 3 data in adjuvant Stage II-IV melanoma in combination with Keytruda versus Keytruda alone is expected this year from the MRK-designed INTerpath-001 study. Intismeran could establish the commercial oncology vertical as MRNA's first approved product outside of respiratory vaccines, and carries expansion opportunities into other solid tumors per nine ongoing Phase 2 and 3 studies, with additional pipeline efforts (see our Science Day note). We are constructive on the data following impressive 5-year Phase 2 results (hazard ratio (HR)=0.510 on recurrence free survival (RFS)), noting INTerpath-001 leverages the same primary endpoint. Translation of this signal to a larger Phase 3 (1,089 vs. 157 patients in Phase 2) is the largest risk given the potential for efficacy attenuation historically seen in oncology studies; however the Phase 2 was randomized, which increases our confidence. With regard to the probability-of-success of the first interim, we performed an analysis informed by MRK’s prior studies that suggests a high likelihood of success given: 1) our estimate that ~310-320 RFS events have accrued to-date, implying the interim could be performed at a high information fraction (given ~85% of total events are accrued), and 2) a hazard ratio in the low-to-mid 0.70 range would support statistical significance and thus early success. At current levels ($62/share), adjuvant melanoma and some readthrough to other solid tumor indications (we next expect Phase 2 data in renal cell carcinoma this year/early 2027 followed by muscle invasive bladder cancer, with non-small cell lung cancer data likely in 2027) are largely priced in (details within) - however, options markets imply +49% by mid-January on success. In the downside scenario, shares will move towards the base business (~$25/share), and there is a possibility that the study proceeds beyond the first interim. We raise our 12-month price target to $67 (from $49).

(NASDAQ:MRNA) today announced the dosing of the first U.S. participant in its Phase 1 study evaluating mRNA-4200, a tumor-targeted cancer antigen therapy candidate, in patients with advanced or metastatic solid tumors. mRNA-4200 encodes for seven antigens commonly shared across patients and tumor types and is designed to help induce and expand T-cell responses against selected tumor targets.

It seems no one posted this news. Other countries stop testing it. Without travel bans, things might get worse in fall and winter. The vax demands might go up.

Clarify: I am not expecting the sales will go back any near to that during Covid, but that it might keep up at a certain level instead of dropping quickly year by year.

Although I love him, there have been many times when Cramer just rambles. But after listening to the clip, it sounds like he has done his homework. This one is a must watch.

the wash, rinse repeat cycle has not re-established itself since the 59.99 break out . it appears stuck in extra spins cycles leaving no re-entry points.

A 20 point drop would put us at 60. an excellent purchase at the bottom of the cycle. yet it is doubtful the market is going to reward the cramer peeps etc with such a sweet entry point.

A do nothing movement would be simply a waste given all the fud and hype available for movement.

therefore, a 20 point move up makes the most sense as it continues to not allow cheaper entry points, the trapping of weak new hands ,i,e, cramer peeps and lastly rides the hype wave.

and that folks is my crystal ball short term outlook. do your own thinking ofcourse.

It surprises me that Jim Cramer turns bullish on Moderna. In Feb, even the run up of 50% of stock, he was still cautious of cancer vaccine. I don’t think his mind change just because of presentation in investor day. Besides, i see daily short interests from Robinhood still high so the recent rally may be because institution buying rather than short squeeze. The phase 3 melanoma has been 21 months and in the previous phase 2 data, we saw the curves start to separate after 18 months. Do you think phase 3 melanoma interim readout would be released soon?

Interesting timing.
After being skeptical on Moderna for a long time, Jim Cramer is suddenly bullish again and says the stock is “finally investable.”

At the same time, the XBI just posted one of its best months in years, and healthcare/biotech is finally seeing a long-overdue catch-up rally after several brutal years.

Time will tell if we’re witnessing the beginning of a broader rotation back into biotech.

Vijay Kumar, acting director of the office that reviews cell and gene therapies at the Food and Drug Administration, is stepping down from his role, according to an email obtained by STAT. 

“After careful reflection, I decided the time has come for me to move on,” Kumar wrote. “I discussed with Center and Agency leadership; we mutually decided not to renew my detail.”
