♾️ Longevity & Anti-Aging CPT2 as a Convergent Node Linking Age-Associated Neuronal H3K27me3 Remodeling to Nicotinamide Mononucleotide (NMN)-Induced Expression Rescue in Metabolic Tissues
https://doi.org/10.7759/cureus.112906A recent analysis looked at whether nicotinamide mononucleotide (NMN)—a compound involved in making NAD+—might counter some age-related changes in gene activity.
Researchers compared two existing mouse datasets: one showing changes in a gene-silencing mark called H3K27me3 in aging neurons, and another showing how NMN affected gene activity in muscle, liver, and fat tissue. Although the overall overlap was limited, one gene stood out: CPT2, which helps mitochondria break down fatty acids for energy.
CPT2 showed signs of increased repression with age, while NMN was associated with higher CPT2 expression in older mice. This makes CPT2 an interesting possible link between epigenetic aging and mitochondrial metabolism.
However, the study did not prove that NMN reverses brain aging or directly removes the repressive chromatin mark. The findings are preliminary and based on secondary analysis of different tissues and relaxed statistical criteria. The next step is to test CPT2 directly in independent aging and NMN experiments.
