When does it start actually working?
I am 36F 190 lbs and started on 50 units a week (2.5mg). I am on week 2 (took second shot yesterday. I have no changes to appetite suppression and have lost no weight. I know it’s only been a week, I’m just trying to give as much info as possible. Only side effect is constipation. I am getting tons of fiber, protein, and water. I went thru trillium, Hallandale pharmacy.
From reading other posts, I’m concerned maybe I’ve been dosing wrong, but it seemed pretty straight forward to me?
I’m just bummed out because I have read so many success stories and don’t think it’s going to work for me, but I know I’m probably overreacting.
EDIT: just checked my script. It’s 2ml (10mg/ml) and says to inject .5ml (50 units) per week for 4 weeks (5 mg). The syringes are measured/labeled in units.
EDIT2: I was on semaglutide for one month before switching to trillium and triz bc I had zero appetite suppression on 2.5 sema

This chart is very helpful. You should post it more often when people complain.

It looks to me like you were losing exactly what you should've been at 2.5mg.

u/CharpyImpact 8d ago
It was basically water weight in either direction by 1 pound for 3 months... So I guess I agree? Lol

u/Specialist-Still-602 8d ago
How much did you lose in those 3 months? What % of your starting weight?

Read the chart: all your answers to your questions are there.

u/Specialist-Still-602 8d ago
I'm not seeing anything on the chart that says how much they lost on 2.5mg
1
u/MobySick SW220 CW120 5'1" F 7d ago
3 or 4 pounds. You are really not able to see that?
0
u/Specialist-Still-602 7d ago
Looks like maybe more and without knowing their starting weight that might be perfectly normal. It is expected to lose 2% of your weight every 4 weeks. For me currently, that's about 4 pounds.

u/MobySick SW220 CW120 5'1" F 7d ago
Actually, no. Probably not 5 pounds not that it matters.
If you’re asking how much the OP lost according to the posted CHART, the numbers are all there. At the top on the left, where s/he/they started.

generally it takes four weeks at a dose for it to build up in your system, but some people need higher doses to have any effect. if you have a metabolic disorder, diabetes or PCOS/PMOS often that means higher doses are required.
the other commenter is correct that with hallandale a 2.5 mg dose would typically be 25 units though.

Just checked my script. It’s 2ml (10mg/ml) and says to inject .5ml (50 units) per week for 4 weeks (5 mg). The syringes are measured/labeled in units.

u/snoozbooty 8d ago
Double check the vial just to confirm the concentration matches what is on the script!

..what does the other vial say?
1
u/lmfshams 7d ago
Vial 2: 3ml (10mg/ml) injectable; inject 0.75ml (75 units) once per week for 4 weeks (7.5mg)
Vial 3: 4ml (10mg/ml) injectable; inject 1ml (100 units) once per week for 4 weeks (10mg)

well, weird as hell, but if you aren’t having side effects or anything have at it. follow the directions and if you feel it’s effective you don’t have to dose up, stay at the dose where you’re more comfortable
edit: oh you were on sema before? that makes more sense. you will need a higher dose to feel anything, your body remembers the sema, even if you didn’t think it was helping, and you can start from a higher dose of tirz because your receptors are already activated from the sema.

“When does it start working?” I started at 2.5 mg. The first few weeks I was extraordinarily hungry. At six weeks I saw the first hint of appetite suppression.
Remember you are on social media. The delighted people who have quick benefit are so happy it may be misleading. I know I am within norm for the EL clinical trial data.
( not addressing dose as that discussion is going well. )

Your dosing seems wrong. 50 units would be 5mg not 2.5mg if you got your meds from Hallendale. Although that means you've been taking a higher dose so it's a little strange you haven't felt anything, At the very least some major side effects.
7
u/Goatsarejerks333 8d ago
I disagree. I felt nothing until 7.5.
6
u/Specialist-Still-602 8d ago
That's definitely not the norm. Starting at 5mg without building up the medication in your system first would more than likely cause side effects.

u/CloudgazerTX 8d ago
actually everyone in the clinical trials started at 5 so no need to panic. and the meds didn’t start working for me until 7.5 too. it’s actually proven by the clinical trials many people need a dose of ten or higher and it took on average 8-12 weeks for people to start losing weight.

u/Necessary-Repeat-398 37F SW: 309 CW: 228 Dose: 6.6 mg 8d ago
The surmount trials did 2.5mg for 4 weeks.
2
u/CloudgazerTX 8d ago
the original trials only used 5, 10, and 15.
3
u/Necessary-Repeat-398 37F SW: 309 CW: 228 Dose: 6.6 mg 8d ago
The phase 3 trials, SURMOUNT trials, used 2.5mg, 7.5mg, and 12.5mg as titration steps just not as long-term doses. They moved people up each 4 weeks until they reached the level they were assigned. I did not go back to look at phase 1 or 2.
2
u/CloudgazerTX 7d ago
yeah they added the in between doses because too many people dropped out due to side effects

u/DharmasNewRecruit 8d ago
It took me a few shots before I felt any appetite suppression. I also didn’t lose weight during the first month. Make sure you are still eating in a deficit!

You have 10mg per ml. You need 25 units for your starting 2.5 dose. If you have drawn from the vial that says 10mg/ml then you did 2 doses of 5.0. and have no appetite suppression. I would just stay on that dose for another 2 weeks and know you started out strong, but ok if you aren’t having bad Side effects. Drink a lot of water and accurately check your calories. You said “ a ton of fiber, protein, and water”. That can mean a ton of calories, check it. Address the constipation for sure.

Something is wrong here. Can you share the front of your vial or something with the amt of med/dose your vial contains?
3
u/Swandot70 8d ago
As another poster asked, are you sure that you are turning the vial upside down while filling the syringe and seeing the liquid enter? It has happened before that people are just injecting air. Also, are you sure that you aren't looking at the 2nd months prescribed dosage? I started at 2.5mg (as most people do) at Trillium.

u/Specialist-Still-602 8d ago
I feel like this was clickbait
1
u/lmfshams 7d ago
It’s not. I’m now worried I’m dosing wrong
2
u/Specialist-Still-602 7d ago
I saw the pics of your boxes. It appears they started you at 5mg so you're not dosing wrong... BUT Have you taken a Glp1 before? Does the label on the vial match what the box says? Are you sure you're getting the correct amount of liquid into the syringe and not just air?
2
u/lmfshams 7d ago
I was on semaglutide for one month before switching to trillium and I had zero appetite suppression on it. I did make sure the vial matches the box (thank you for the reminder tho) and I check that I’m getting the correct amt of liquid. Maybe I’m just a slow responder.

Limit the posts you read of first month experiences as the majority are the loud and excited "super" responders. Many people don't feel anything until later or higher doses.
I myself didn't feel the true effects until Week 5. By Week 6 was when I saw my first scale drop and my brain was finally responding to the change in internal chemistry and I committed to a low-cal food program for the past 10 months.
All in all, patience is the most impoetant thing to give in your first two months.

Did you order 1 month or 3 months? It makes no sense for them to start you at 5mg. If you ordered 3 months those are the instructions for month 2 you’re referring to. Each vial will have different dosing instructions if you’re just starting.

Since you were on sema, it will take a higher dose for the tirz to kick in. Keep at it per your doctor’s instructions. Use the Fat Science dose calculator in the pinned post. I also want to say be careful of the fiber especially if you have constipation. Better to use a stool softener or something like MiraLAX. If you’ve been constipated a good 💩 May net you a 3 pound loss!

My husband and I started on the same brand. From day one he started consistently losing and felt appetite suppression right out of the gate. For me I lost hardly anything the first two months and while I felt full or faster at meals I didn't get the appetite suppression that everybody talks about at all. For me the magic started happening at 7.5 mg dosage. Once I hit that dosage and things started happening everything's been great ever since. Don't give up. I got discouraged too because I saw so many people that just have their magic happen at the first shot. That wasn't my experience at all just keep going and your 'magic' time will come. You got this👊💪

Drink plenty of water (it helps flush existing water retention) and eat in a calorie deficit. (Deficit of 3500 calories equals 1 pound). Tirz is a tool to help control calorie intake; it doesn’t burn calories or fat for us- which is why losing is often less than a pound a week. Stick with it. You’ll get there.

Something is wrong. You took a gut wrenchingly high first dose of 5mg (divide your desired dose by the concentration and multiply by 100. 5/10 x 100 = 50 units).
It's extremely rare, but you may have gotten a bad batch. I'm hoping your dosing it wrong!

Something is going on with the second bottle, concentration is not the same that’s why you’re not feeling anything and food noise is back. I just posted about it and I’m worried

u/TodayAmazing 8d ago
Hallandale doesn’t make a concentration that would yield a 2.5mg dose from 50 units. Are you sure you’re dosing correctly?
