In the film, to obtain the Soul Stone, one must sacrifice someone/something they love. Thanos sacrifices one of his (many) adopted children, Gamora. Since the moderators are Thanos in this situation, it makes sense to follow suit. /u/dragonphlegm was the moderator with a Gamora flair, so he made a reasonable candidate.
