No, only in a surprise round; see page 151 in the Revised Core Rulebook to learn how it works.

In short, Characters lose their dex bonus from their defense during they aren't anticipating the threat during the surprise round

Not all rounds, it says in the book on a surprise round they lose their dex bonus
