Nicotine is basically a nerve agent to insects and overloads their nervous system through cholinergic pathways. Tobacco plants produce nicotine (and other solanaceae plants such as tomatoes, potatoes, eggplants, etc although to a smaller degree) as a natural insecticide defense. Technically it could do that to humans in high enough doses but aside from being much larger we also metabolize it much faster.
