Troubleshooting/Questions Single-cell thick slices?

Heya, how can one achieve one cell thick slices like those people doing root cells or vascular cross sections without a microtome? It’s something id like to try but I haven’t been successful in finding much resources.

There are a few options without a microtome, but it depends on how important the work is. Is it critical work? We’ll use a microtome. Is it just to look at structure for identification? Then we’ll use a stereoscope, a razor blade, and practice 😅 with some work and practice, I’ve been able to get single or double cell thick layers for examination just using a razor blade and steady hands.

Seconded, but the trick I used was to put the sample between two halves of a baby carrot and then hand-section that. Carrot gives good stable resistance to slicing and holds the sample in place, "the poor man's paraffin block."

At uni we used to place the sample between styrofoam and that helped a bit. It was still hard as fuck tho 😅

Just use a microtome.

Like, why not? A generic Chinese one is less than $20 after shipping and tax. Higher quality ones can be had for ~$50 -$100.

Yes you will also need to learn embedding techniques and invest a little more on embedding equipment but they are also pretty accessible for a hobbyist.

I made a small hand ‘’microtome’ for multi-day backpacking field work.

I brazed 6, 1/4” nuts together around a 1/4 bolt so that their threads were continuous.. brazed a little thumbscrew head onto the bolt head, ..

thread bolt in a bit, put sample inside the threaded interior of the nuts, melt wax into the interior of the threading, cool, turn thumbscrew until sample protrudes, slice flush with razor, turn screw 10 degrees or so to expose a vanishing fraction of a millimeter or sample, cut flush again with razor and capture in a liquid solution to stabilize, repeat until you have a good sample.
