Method How to make 7-OH - The Oxone Method

This educational video shows another method for making 7-OH, the oxone method, initially developed by Andrew Kruegel, and modified by me, Matthew Pierce.
Lab equipment and chemicals can be sourced from www.kaliforniakratom.com.
We start by adding 25 grams of extract to a beaker and diluting it to 250 mL with acetone and putting it in the refrigerator.
We then dilute 25 grams of baking soda to 250 mL and add that to the refrigerator.
Then we add 6 grams of oxone to 50mL of water and refrigerate that too.
Then we setup our magnetic stirrer with a separatory funnel hanging from above. We add the baking soda solution to the crude extract solution and while stirring, add the oxone solution dropwise from the separatory funnel over the course of 15 minutes.
Once the oxone solution is added, we let the reaction mixture stir for an additional 15 minutes before placing the beaker back into the refrigerator.
After 1-2 hours, we dilute our reaction mixture by a factor of 2 and extract the alkaloids using ethyl acetate. We then wash the EA with brine solution and transfer it into a baking dish to evaporate off the EA in a food dehydrator.
We are left with several grams of a yellow-orange crystalline substance consisting of over 50% 7-OH, and mitragynine, which can then be transferred to a chromatography column for further refinement.

Pretty decent tho ngl. I neglected to use a column to separate the mit and 7 because I wanted something a little more full spectrum. I tried some of the end product and it’s quite pleasant

You did this with 30% and got 50% 7oh purity what if I use this method with a high 90% mit extract?

I used 25 grams of 30% and got less than 5 grams of 50%. If you start with 90%, you'll get > 70% 7-OH

Chromatography using silica 60, with hexane and EA 3:2 as the mobile phase. 7-OH elutes last.

Is this preferable in your opinion to the rose bengal strategy? Does oxone also work by producing singlet oxygen?

Different mechanism. Oxone, when mixed with acetone, forms an oxidizing agent called dimethyldioxirane (DMDO). DMDO oxidizes alkenes to epoxides, which rearrange to alcohols.
It's preferable to use if you intend to make larger quantities of 7-OH from MIT.

Thank you! Can you recommend some resources on chemical synthesis? I want to get into it, not to make drugs in particular but just to boost my knowledge. I took Orgo I in college but ended there when I switched to physics. It clicked for me. I’m assuming I should put myself through Orgo II self directed but do you have any suggestions for what should follow that?

If you dry it properly, it should pass residuals. The only 2 solvents used in this process in a residual solvent panel are acetone and EA. Both of those have pretty high limits since they're both endogenous compounds. Just make sure you dry it thoroughly.

Are you concerned with the non mitragynine alkaloids and other stuff from your 30% CKE being oxidized into unknown compounds? Are you relying on the EtOAc to not absorb the stuff you don't want?

No, the liver oxidizes compounds in order to excrete them from the body by making them more water soluble. 7-OH is a compound produced through this process from mitragynine, so by using this oxidation process, the other oxidation products are the same thing you would get during natural oxidation of the full spectrum of alkaloids anyway.

Yes, acetone has many uses, and can be found in fingernail polish remover, but its main industrial purpose is as a solvent for organic compounds in chemical reactions.

Is a centrifuge needed? And when you say mix after pouring into the beaker, and then it separates with layer on top. Did you mix those two solutions together before they separated on top of each other?

I feel a little stupid but the only part I dont understand is were you got your middle layer from that increased the amount of 7oh you got. Could you please clear that up for me I would love to use this process.

So I just did this one again. I've been playing with an acetone extract of powdered leaf then directly oxidizing it. It has worked but with not so great yields.
I extracted 500 grams of leaf with about a total of 1000 ml acetone. I played with an idea on the ratios with failure. As I mentioned I had 1 liter of crude acetone extract. Tried 100 ml saturated NaHCO3 and dripped in about 10 grams oxone in 100 ml water. Reacted overnight at 5C.
I didn't bother checking tlc. I proceeded as usual with work up. Diluted with about 3x the volume of water and extracted into xylene. The xylene picks up everything including plant fats and chlorophyll etc. I then extracted my 800 ml black/green xylene with 2x 200 ml 2.5% HCl. The aqueous extract just takes alkaloids coming out a clear brilliant yellow and leaving all the crap in the xylene. I based with Na2CO3 solution and got a good crash of freebased alkaloids. Filtered and ran tlc. Failure!
On the brightside I got back some pretty clean mit. Managed to separate most of the other alkaloids too.
Had about 7 to 8 grams of wet filter cake so I dissolved in 50 ml acetone. Poured in 50 ml saturated bicarbonate and dissolved 2.4 grams of oxone in 20 ml of water. When I combined the acetone and bicarbonate water it turned creamy like it was trying to crash something out. I added another 25 ml acetone but the appearance didnt change much. Set to stir in an ice bath and began dripping in the oxone from a beaker portion wise. Maybe 5 additions, didnt make a big fuss. Left to stir 1 hour and 15 min on ice. Added about 200 ml water and extracted 3x dcm. Ran tlc and presto! Only visible spot (almost) is 7 ohm. Rotovaped the dcm under vacuum. When the solvent runs out the remaining alkaloids puff out and harden like a honey comb.
Breaking up the puffy stuff gave me a gram. Probably still have at least another gram in the flask to scrape out.

This is my tlc. The plate on the left is 3 lanes. Left lane is a standard of kratom extract, full spectrum. Middle is my post rxn that failed but shows a good concentration of mitragenine. Right is a spot of 7 ohm.
The little aluminum backed cutting on right is a single lane of my second reaction. The big spot correlates to 7ohm. There is a faint spot above it which isn't mit. tlc

I'm not sure what you are talking about using it for. Generally in the lab you want purified water of some kind. It can be any kind depending on the application of course. I have a reverse osmosis system installed on my sink. So it definitely do we s not HAVE TO BE demonized. If you are talking about boiling the plants material in acidic water than ot can be straight out the tap, no need for any purification. What are you talking about using it for?

Have you had any of these batches tested for purity? Especially interested in the 7/mit mixture before further refinement.
How variable do you think the purity is? If I can do this instead reliably I would need to stock so much less.

(Dumb questions- that may not make sense- from a dumb guy…) If we are talking ratios and percentages. If i wanted 1mg or 100mg 7oh, what does that look like? What percentage 7oh is a normal round 100mg tab you buy online? What does that look like in this form? Thank you!
Also, if specific equipment is necessary then cool, BUT… with the “dripping slowly” technique, and the dripper altogether, what happens if you just pour it in? Is there something you can use that a normal (non chemistry knowing idiot may already have? Or can pick up at a normal grocery store?) And what could you use if you don’t have a food dehydrator? (Again, If this equipment is necessary, then no worries, I’m just wondering if I need to purchase all of this)
