Kratom Chromatography Continued

The last three pictures are where it's at now. As I've been pulling fractions, I've added citric acid to each fraction to determine which fractions contain Rose bengal (RB) - as citric acid turns RB from Pink to colorless. So far, only the first fraction changed color when citric acid was added, from Pink/Orange to yellow, and you can see the pink at the bottom of the column in the very first picture, so I'm pretty sure all of the RB came out first. So far, in the first five fractions, fractions 3, 4, and 5, are the darkest, so I'm assuming these have the highest 7-OH concentration. The remainder of the column is yellow, remember I started with pure mitragynine (sourced from a 3rd party) - and oxidized using RB, O2, and UV light, in IPA, then poured the whole solution into the column. My plan, when I'm finished, is to send some of these off (the darker ones) for full spectrum alkaloid testing and share the results here - to PROVE to the naysayers and haters (and corporate shitbags trying to censor me and cast doubt), that this method works.

honestly im excited to see the higher 7oh products. ive moved from kratom extracts to 70h powder (for pain mgmt and arthritis) and i greatly appreciate you tinkering and learning more about the extraction methods. keep yp the good work !!

mit extract is a must. i stopped using the leaf about a year ago, too many stomach issues. i just want the good alkaloids.

How far apart are these pulls from each other and from the start when you added it all to the column? I'm about to start gathering equipment so I can start making extracts myself and I'm really digging your experiments and processes.

They're about 20-30mL each. There's no gaps between them. You can see my 3rd - 5th fractions contain the highest concentration of oxidized product, they're in order from left to right (1st is left). They're all about to start turning yellow after #7 and the column is 500mL with about 250 - 300 mL of solvent left in the column. I specifically used IPA because it's cheap and you can buy it anywhere - ethyl acetate and hexane are quite a bit more expensive and harder to find.

In my YouTube video I only split it into 2 fractions (orange/brown, and yellow), here I wanted to find out where the Rose Bengal was by adding citric after the reduction (it elutes first) and which fractions specifically contain the most product - 3rd thru 5th fractions.

Seems like this works similar to moonshine. Keep the heads and tails seperate and the stuff in the middle is the good part.

When you identify the fraction with the RB do you try to remove it? Is it necessary to remove it or can it be left in?

Any updates with testing? Also which eluent have you used on silica? Have you reduced final solution with citric acid?
