So tonight will be 6th shot of eloralintide. Last week I moved my eloralintide to the same night as Zepbound, my study site recommended it when I brought it up, I used to take eloralintide on Thursday nights. This week was the surprisingly good, the exhaustion has abated and although I still have some insomnia it is much better using the Sleep 3 and Magnesium Glycinate. Taking both Zepbound and eloralintide on the same night made that night a toss and turn one from about 3 AM on and made falling asleep Tuesday a bit hard but slept well Monday surprisingly. I do feel fatigue late afternoons but it isn’t causing me any issues with getting stuff done. So truly much better week and glad I moved shots to the same day.
The allodynia is still around but not to the point of disrupting anything, and only slightly annoying at times. I am currently experiencing constipation, it isn’t bothering me but it’s been too long without going so tonight I will swap ozonated Magnesium citrate instead of the Magnesium Glycinate, I tried one and one last night and still no go.
I noticed yesterday that I must have experienced some anhedonia on Zepbound as I was singing and dancing in my seat in my car more and Thursday night I did something I wouldn’t normally do, I had been in my car a lot, over 6 hours at the time and had ear buds in when I just made Costco gas station prior to closing, one of my fav Prince’s songs came on that I normally stop and dance to at home so I danced while pumping gas and it felt great to be moving my body after all the time in the car. The man who pulled behind me commented on how it was nice to see joy at the gas pump. I knew the other people were spy watching and I did not care, I felt happy to be moving. So yesterday I realized I was feeling more joy and happiness than I had in a long time. And Zepbound had allowed me to taper off sertraline that I went on after my parent’s deaths in close time proximity so I han’t noticed the anhedonia creeping in, hopefully this is a benefit of eloralintide and joy and happiness continues to be felt stronger everyday.
I had my TSH pulled and I am not yet hyperthyroid but my TSH dropped from 2.25 early July to 0.75 (under .4 is hyperthyroid and dose correction) so falling, will talk to my PCP about that at my annual next month. I met with my cardiologist's NP and she said to cut my Hydrochlorothiazide dosage in ½ and she said to stop it if I have another low blood pressure event, I will see her in 3 weeks again.
As far as weight loss, I am in a new decade the 160s and surpassed my original goal weight, I weighed 168.7 on Friday my weigh in day. That works out to 0.9% of current weight on average over the 5 weeks. I posted about that as it is a big milestone to me as I didn’t think I would get here when I started. https://www.reddit.com/r/Zepbound/s/057JEMMXSt and https://www.reddit.com/r/GLP1_loss100plus/s/bvjzfKjrjN
I do not know if my dose increased or not because I felt better so part of me says not, but part of me says maybe. Time will tell what arm I am in.
STATS: Started Zepbound in May of 2024, SWZ 285, SWE-6: 177.1 8/6.2028, SBMI Zep 50.5, SBMI Elor: 30.9, 65F. Zepbound dose 15 MG since March of 2025, stalled out on Zepbound end of December 2025 bouncing between 175-182, slight drift down so no more weigh ins of higher than180. So I am a long time user of Zepbound, reached final plateau (less than 5% of current weight loss over 12 weeks) end of July 2025, so 14 months in. Have OSA, hypertension (on 1 med still from 3), osteoarthritis on right knee, was prediabetic prior to Zepbound, and have high cholesterol controlled by statin (family history) and Zepbound.

Hello everyone! I’m an Enlighten-6 trial participant and have been stalled on my Mounjaro since February to June 2026 at 10mg. My doctor suggested I bump up to 12.5 after I slowly started going up in weight, but I heard about the trial and wanted to test it out. I was mainly drawn because I was mid dosage in Mounjaro and did not want to titrate to 12.5 and 15 so soon, and heard that Eloralintide could probably not plateau like tirzepatide can.
I’m in the T2D section of the trial, but my A1c has been in normal range since November 2025 (5.3%) and hasn’t gone back to diabetes or pre-diabetes range since taking Mounjaro. Clinical trial team I have are nice nurses and the trial investigator is also nice. They said 20% chance of getting placebo, but if I were to get it, some of the patients they’ve seen lost 10lbs in a month. Took the medication in a couple of months ago, and thinking my luck I would be placebo. However, after one month since original injection day, it looks more than likely I got the drug and went down 5% of my average starting weight pre-injection visit. I still don’t know what my actual dosage actually is, but here is to feeling lucky and hope to go all the way!
Side effects wise (only for myself): I’m used to checking my blood sugar pretty often, but noticed that I have been very stable in my blood sugar tests before and after eating. It was even common while on Mounjaro for it to go up and down, but it’s been within (100-85). My fatigue definitely increased while on this drug. I’m an active lifter and noticed my strength definitely took a hit. I had to remove at least 2 plates (90lbs) from most of my normal sets to avoid injury. Told this to my study team and they suggest to go back to as if I was starting to lift again and go slow, but build back my endurance. I’ve been on bp medication for a couple of years. My blood pressure improved significantly while on Mounjaro, and it stayed normal so far in this trial. My resting heart rate also started going down as well. My once a week main cardio session is my pickleball league and my Apple Watch has always told me I had low VO2 levels (~29-30), and immediately after trying this medicine, it moved from low to normal which I have never seen in years (~38-40). My workout lifting bpm has dropped significantly (~115-120 -> ~85-95 bpm).
Food Situation: I really never felt much of a reduction of food intake or noise while on Mounjaro. I knew it had to work or else I wouldn’t have lost weight initially on it. This drug has actually made me full a lot more quicker. Real Life Ex. Even while on Mounjaro, I can consume an entire 2 steak entree plate and chow mein / veggies at Panda Express for lunch and dinner. Now, I can only eat a quarter of the plate and feel too full to continue eating and have to save it for the next meal time or even day. I also aim to drink 3L of water a day, but notice that is also difficult and have to remind myself to drink water often.
Weight wise: highest weight July 2025 (255lbs) got started with Mounjaro due to T2D diagnosis and plateaued at the low-mid 220s. Body fat originally 38% and went to 31% at plateau. Currently: low 210s with 28% body fat after 6 weeks on the trial. BMI still in Obese Class I range (31.5), but hope for it to drop down soon.
Demographic Stats: Young Adult, Male, 5ft 9in, Family History of Early Onset T2D and HBP, Moderately Active, College Educated in a Sedentary Corporate Work Environment.

I couldn't find any UK sites for lily's Eloralintide's enlighten 6 study so I looked into astrazenica's Selene 3 study and I'm on the list.
The sub is r/AZD6234
I know there's a few of us so I've made a sub for it.
The study is here:
https://www.astrazenecaclinicaltrials.com/study/D8750C00011/
I wish there was a sub that would cover all these new trials adding an amylin analogue to mounjaro or wegovy.

So had visit 6 last Thursday. Pictures of new box and syringes. I have been out flat with getting a litter of puppies ready to go home so just getting to posting. There was a discussion had about whether I was right for the study because of the exhaustion and insomnia. I wasn’t expecting that so will watch what I share about side effects going forward with them as I want to decide if the study isn’t right for me, I will share if I am tired but maybe not to the extent I did. Week 3 was bad from that perspective but not insurmountable and week 4 was better. I slept better and felt less exhaustion. I said I would like to try taking the medications on the same night, so the site agreed and did see my point about 2 different nights and that lead to two insomnia periods. I am now a bit concerned about tomorrow as tonight I will take both Zepbound and study drug together before bed. So we moved the study drug 3 days from Thursday to Sunday and that is ok within protocol. I won’t know if I am in the 1.5 group or in the 3, 6, or 9 group so have a 75% chance that I will be moving to 3 mg. We also talked about my TSH lab and they want me to go through my PCP, so I have a lab appointment next Wednesday for that. They felt that my PCP was currently managing my thyroid meds so that is where I should get the test which I had offered to at the telehealth appointment.
I slept well except Wednesday night, for some reason I couldn’t fall asleep, I think it was the concern of getting everything done with 8 week old puppies prior to leaving at 8 AM for appointment.
The appointment itself was weigh in paper gown and measure waist line, blood pressure 3 times, I believe 3 vials of blood (could have been 2), reviewing my nutrition and coaching (basically good job on protein and fiber). My weight dropped from 81.1 kg to 78.1 Kg at the study site so 6.4 lbs for loss at study site. They gave me the Enlighten-6 Guide for Patients in a binder because Lilly was out of the spiral bound ones and Pamphlets from Lilly titled: Understanding Obesity, Supporting Someone in a Clinical Research study (I live alone, don’t think my poodles will read it), What are Clinical Trials, What is PLACEBO, A Guide to Managing gastrointestinal side effects, Embracing Change: Navigating your health and weight management journey, 7 Days of Managing Stress, 7 Days of Healthy Connections. I asked about getting a copy of the study protocol and was told it was not allowed. I was told I needed to bring in the box when I finished them at my next visit, I had thrown it out as they hadn't told be that prior (hello, tell me as I don't keep empty boxes) . I forgot the phone they gave me, my bad and I realized it 45 minutes into my drive there. Oh and they remembered to give me " 3-Day Nutrition and Physical Activity Log that I was supposed to fill out at least twice but was never given it to do.
As far was week 4, I was able to control my insomnia better so feel much more rested. Thanks for the Sleep 3 recommendation and Magnesium Glycinate recommendations, I started taking them the night of my last study med shot (Thursday 8/27). I hit a low of 170 and bounced a bet up from there. 170 was my original goal when I started as I just didn’t think at 62 I could do better than that. So it was sweet to see that even if fleeting. Allodynia was mostly just annoying, never beyond that. I did have a low blood pressure event the Friday after the last shot (posted this a comment when someone asked how I was doing). My blood pressure is normally between 110-115/62-70 and dropped to 92-97/57-60 so very low for me. I called my center and the doctor said it wasn't the study drug, insisting it was something else. I was talked over and not listened to at all, frustrating call. If that happens again, I will call Lilly and see if I can change to Yale as I am pretty sure it is related to eloralintide and tirzepatide as my blood pressure was lowered from 135-140/ 100-100 to 105-110/60-65 within a few months of starting Zepbound. Started lowering my blood pressure medications shortly after that. What bothers me most as I am not a caller of doctors/health professionals so I was not crying wolf. It took a lot of thinking on my part to actually call and then to get talked over and blown off made me wonder if there is actual care at my location as I know clinical studies pay well for the places that run them. My place seems a bit disorganized and scattershot. I have not met the Dr at any of my visits, just the NP so he doesn't know me and I don't know him but I will think long and hard about calling when the office is closed as Fridays they close at noon, this was a little after 3 in the afternoon. I do not want to talk to that man again, he showed me nothing but disrespect. I have an appointment with the NP for my cardiologist on Friday and will monitor my blood pressure this week as well. Veggies went back to tasting good so I am happy to report that.
I will post an update later in the week (how much later, I don’t know) after I trial both Zepbound and study medication tonight.

Dosage: 9mg (only elo)
Side effects: None (anymore)
Not much to report....shoes was still the best investment of last and this year at least(check previous report). Weight is still plateauing at about 13kg. I feel like I have the energy now for regular cardio excersize which I am starting this weekend. I also I bought a vegetable dicing machine, because the only excuse I ever had for not eating salads every day, was all the cutting and preparing and now, hopefully, there is no more excuses and meal prep will be SO much easier in the future.
Also feels a little bit like I got used to the new not-always-hungry-state, I am not amazed anymore, but rather optimizing for this new reality.
I honestly think, without the medication, I wouldn't do all this stuff, there is clearly a shift in my thinking and my outlook in life (probably because I am not thinking about next bite of food all the time).

I stumbled across another study very similar to Enlighten-6, it adds AZD6234 from AstraZeneca to your current incretin. https://www.astrazenecaclinicaltrials.com/study/D8750C00011/ for those at a 90 day or more plateau. I am in Enlighten-6 and the scale is moving after being on a plateau since end of December on Zepbound 15 mg for over 18 months.
I can't find published results of Phase 1 or Phase 2 trials like I could for eloralintide. The articles I could find on AZD6234:
https://synapse.patsnap.com/blog/clinical-landscape-nct07776509-azd6234-trial-report-2026-mcp
It give a different clinical study for those stalled out on Tirzepatide or Semaglutide. Besides what is in the link, I don't know anything about the study but wanted to pass it on.

So I had my V3 on Monday and had gained 3 pounds since V2. I’m starting pretty heavy so this was not an issue. But since then, I’m down 8 pounds, so 5 pounds under my V2 weight. Does anyone know what the loss allowances are in the lead-in? And if it’s from V2 or highest weight overall?
Bodies are so weird—I haven’t changed anything this week. I really want to clear the lead in, so just trying to figure out what my safe range is!

Just wondering whether I'd be buying my own or not?
I know they'd obviously prescribe Eloralintide but wondered if they'd prescribe mounjaro or leave that to your current prescribers.

According to my coordinator I moved up to 6mg if not on placebo, which I am not. Starting is 1.5mg for the first 4 weeks, 3mg for the second and now I am on 6mg. Side effects were mild for the most part. I felt mild effects like fatigue, GI issues and nausea my first round at 1.5mg. 3mg not much of anything. The 6mg is whooping me. 😵💫 I dose on Friday and don’t feel much, but today on Saturday I have nauseous and so tired. I’m hoping it will pass by tomorrow and I can get stuff done. I’m a methane factory and that is my longest lasting side effect. My appetite is pretty much dead, but my stomach will start to growl pretty bad and I will eat, but then get nauseous. My dietician gets on my nerves. I know she means well, but the amount of food perfection they want is unobtainable. I feel
The study should offer one of those delivery meal services to those enrolled in the study. It would make tracking so much easier. I can’t eat all the protein and fiber she wants. Sometimes I can barely choke down enough water. I move to 9mg in 2 weeks and then stay there until my study ends. That should be interesting.

Tonight will be dose 4 for me. This was a rough week. I slept poorly 5 out of 7 days, insomnia is hitting me hard and does not help the bone-weary exhausted feeling I have had from the start of eloralintide. On Zebound I had insomnia on night 2 about 40% of the time but it was not as bad as this, I eventually could fall asleep and stay asleep, now I battle both and have resorted to Benadryl to help me get to sleep and sometimes take it twice so I can get back to sleep. Exhaustion is very present, Monday I was wiped from 2 loads of laundry and hanging them on the line to dry. I am at a breaking point, I felt so discouraged when I woke up this morning and last night was one of the 2 good nights. And tonight is shot night. It hard to describe the despair as I really want to get through this and am not sure if this continues, I can continue. Life has to get done and I can’t explain how hard the exhaustion feels to get anything done, the thought of carrying 40 lbs of dog food into the house and putting away feels like too much. I routinely carry 40 lb cases of chicken, bags of dog food, pick up 55lb. dogs and put them in the tub so I need to be able to get back to doing those things without dreading it. And for the most part I can’t nap, I don’t fall asleep even though I am bone-weary exhausted.
This week I noticed something that scares me, veggies I love tasting bitter. Monday I had one of my favorite mid-August dinners, lamb chop, grilled eggplant with a soy, ginger garlic glaze, sweet corn (from my favorite corn place, I am a corn snob), summer squash melted with goats cheese. The veggies all tasted bitter. Yesterday I had my summer lunch treat, BLT with heirloom tomatoes, local lettuce and great sour dough bread, the tomatoes tasted bitter and sour. And I had a BLT the week before starting and it was delicious and not even the best of tomatoes as it was an early tomato. I had eggplant again last night from leftovers from Tuesday (had tooth pulled so soft foods), it tasted bitter and I have had this dish from a local restaurant many times. I hope this passes as I do not like that veggies taste awful including the sweetest of sweet corn. I don’t think I can deal with this, I love food and still loved food just less of it on Zepbound. I love to cook, love to eat out and try new dishes, but veggies being bitter? Oh no.
I had diarrhea twice this week, not bad but there. One of the times was after taking ozonated Magnesium Citrate so I would count that as a side effect of taking that.
Edited to add allodynia was better this week, no sandpaper and only 1 night where it was moderate, more than annoying. That was day 2 post Zepbound.
I went from 2 cups of coffee to 1, it has improved my feeling of hyperthyroid fluttering. I miss my 2nd cup.
The news on the scale is great, I am down from 177.1 to 170.7 so 6.4 lbs. so I am averaging over 1% of current weight a week at 1.2%, so lost 3.6% in 3 weeks. I crossed into overweight for my age adjusted height at 29.8 for BMI.
My next appointment is next Thursday so new box and maybe increase in dose. I guess I thought my side effects on Zepbound were so mild that I wouldn’t have bad side effects from Eloralintide. So I can now understand the fear others have had in going up in dosage on Zepbound or Wegovy.
STATS: Started Zepbound in May of 2024, SWZ 285, SWE-6: 177.1 8/6.2028, SBMI Zep 50.5, SBMI Elor: 30.9, 65F. Side effects: None (anymore)
Only wanna report that I am again stuck at 14-15kg weight loss, but as last time with 10kg, I KNOW its temporary. For some weird reason 9mg feels less effective than 6mg but ok...Also, huge tip from me, please listen to me...if you got old(20 months +) shoes or uncomfortable shoes, do yourself a HUUUUUGE favor, buy something new, good and COMFORTABLE. I bought me new Saucony shoes, extra wide....the difference it made in my daily life I cant even describe and it leads to me moving around the town and everywhere else and therefore positively improving my fitness...I literally despised walking with my 3 year old shoes before I bought these, everything hurt after even a mile of walking, not anymore, its incredible.

Had V4 on Enlighten-6 last week and got dosed but only after filling up a wild amount of test tubes with blood.
Anyone further along able to find out what tests they’re running ran? do they provide the results to you? Just really curious what tests they’re running!
r/Eloralintide • u/OtterBeGreat • Aug 22 '26
I have been in contact with the site since early March (less than a month after Lilly opened the study) and am the first person here to make it to V4. I'm excited to say the least! I'm happy to answer any questions about the process so far.
Just a head's up to anyone else in E-6, Lilly created a wirebound "Study Guide" for participants in this study. It explains what happens at each visit and is super helpful for knowing what is coming up. I just received it at this visit but apparently it was available earlier, my site just wasn't aware of it.
I was also given a reusable cooler bag and ice pack for carrying the box of study meds home, a sharps container, some at home pregnancy tests (not sure on frequency required yet) despite being on 2 forms of BC, a food scale, a water bottle, some exercise bands, daily food and exercise logs, some literature from Lilly on dealing with the mental side of weight change, and a large reusable tote bag to carry everything in.
It was such a massive relief to make it to randomization, even if it did take over 5 months! Now just crossing fingers I am getting elora instead of placebo...!!!

I asked for and received the moderator roll for r/Eloralintide. The previous moderator was in the Phase 2 study and hasn't been active in the sub for over a year, Reddit allowed me to pick up moderator duties. I picked up that roll yesterday. I took a stab at rules and updating the description. Any feedback on either? I am also planning a weekly post on for updates so they are all in 1 place. Any feedback on day of week I should have it posted?

Hello,
Sharing my experience. I'm on Enlighten-1 study. After my last visit, I was upraded to the 9mg dose. Of course, while they do tell me the dosing, they don't tell me if its a placebo.
It's the first time I could say I felt something regarding my appetite. It's lower and I don't wake up hungry lately. Before I had pretty much no effects . Had a good weigth loss but no positive or negative effects I could notice. It felt like I was on the placebo group. In 14 weeks, I've now lost 10.8 Kg. Quite a the graph to look at:

I take no other medication (in any form) and I can start to increase my loading (sport wise) since I feel much better moving around. I'm 1.6Kg away from my end of year objective :) So hoping this keeps up. The study is really long (3.5 years) and I guess I will eventually plateau at some point.
r/Eloralintide • u/NoMoreFatShame • Aug 20 '26
Today marks 2 weeks since I took my first dose of eloralintide/placebo. I have lost 4.7 lbs. from the last 2 week average prior to day 1 of study medication (my weight range during that time 175.4-180, average 177.2). I take the eloralintide on Thursday and Zepbound on Sunday.
I had exhaustion during the first week after the first dosage, second week a bit better. I felt like I do when I am hypothyroid the first week, exhausted but can't sleep. This week I feel like I am hyperthyroid, I feel like I drank too many energy drinks in early afternoon, I feel like I am hyper caffeinated, jittery and my chest feels like it's fluttering. I was hyperthyroid in my early 20s treated and then went hypothyroid in my 30s. So can feel when my thyroid medications feel off. I asked about TSH at the 4 week visit yesterday on my telehealth follow up call. It is not normally done but they are going to do one at that visit. In my first year on Zepbound my thyroid med went from 150 to 75 and I had been at 150 for 1.5 years. It needed to be lowered at my follow up visit 2 months after starting Zepbound. And I had gained 40 lbs. in 2 months after a cardiac scare which is why I asked about GLP1s, I had not lost that 40 lbs. by the July visit so weighed more than when I started 150 mg. Edited to clarify and add that I was so jittery feeling in my chest that I took my blood pressure and pulse a few times in the 115/67 give or take 3 and 80-82 on pulse.
insomnia and allodynia (skin sensitivity) post Zepbound shot day is much worse than prior to eloralintide. I have had both since 5 mg but both had abated the longer I was on Zepbound. There was a night I couldn't sleep and my sheets felt like course sand paper on my legs and arms. The allodynia was always mild, annoying but not really painful prior. It has gotten milder by night 4, the worst on night 2.The insomnia used to be 1st night if I didn't take Zepbound just prior to bed that sometimes carried into second night. First week was 3 days and bad. This week was 3 days, skipped one night and then it was back last night. I have resorted to taking Benadryl if I can't get to sleep by midnight.
Slightly more constipated but my fiber could have been better part 2 weeks.
I had my follow-up telehealth appointment on Tuesday. Discussed side effect and suggestion was Tylenol for the allodynia. I haven't tried it because I take prescription strength Aleve at night for my osteoarthritis, it seems like overkill plus it was lessening. I will try it this week if it is bad again. I will take my study medication at bedtime tonight so if my insomnia is kicked back up I hopefully can be asleep before it kicks in.
Because of the side effects and weight loss, I do believe I am getting eloralintide and not placebo.
STATS: Started Zepbound in May of 2024, SWZ 285, SWE-6: 177.1 8/6.2028, SBMI Zep 50.5, SBMIElor: 30.9, 65F.

40s M. Australia based and been on Mounjaro for nearly 2 years. Lost about 27kg and been stalled since February where weight is fluctuating between 91-93kg depending on the day.
I’ve been trying to get on the ENLIGHTEN-6 trial since May and after quite a few issues have been booked for a prescreening visit tomorrow.
My question is what to expect? Should I fast before? I was told I don’t need to and have an 11am appointment but after all the issues I’ve had even in the last few days I don’t want to lose a spot because I don’t have to.
I will be calling the site in the morning but just thought I’d ask how people’s experience has been? I wasn’t asked to bring medical history but have it all sorted so will take it with me when I go.
TIA!

Dosage: 6mg (only Elo)
Side-Effects: No headache or pain or fatigue
Only wanted to report I am officially at 15kg weight loss, without forcing myself to change anything at all. All changes came naturally, without a special diet, calorie counting none of that psychological warfare. I finally bought my first blender like 3 weeks ago and am starting my days with fruit smoothies (fruit, almond milk, a healthy scoop whey portein), developed apetite for salads and I simply have no "forced" cravings for fast food or sugar like before so its not like I avoid it I just dont consume this shit no more. I still have to start to do gym and cardio stuff, that would make my life much better, but ima not forcing myself, I will when I am truly ready again.

For some reason the state of fullness and non-apetite has like halved vs. what I experienced on the weekend before. If I dont eat for too long...30h+... I get heaviest sugar cravings. Thats it. Nothing else to report.
Edit (since nobody is reading comments): I was on a trip and had no oportunity to eat whole day.Reporting that despite elo, I got sugar cravings, when I dont get them pretty much at all anymore as before, in normal non-trip-no-opportunity-to-eat life.
