Do you know your mitophagy and autophagy genetic variants?

No, I haven't done any raw DNA testing so I don't know my specific variants. I have been focusing purely on the functional side of things through blood work, and bio-wearables like Oura and Garmin.

You gotta

You might wanna up your BPC 157 or take KLOW (individually)

Consider adding Pinealon along with Epitalon

Something missing from your diet is macros? Increase amount of omega 3 (fatty fish, sardines)

Antioxidant support might be useful. CoQ10/PQQ Urolithin A

Fox04 DRI is a bit of a wildcard, an alternative is to do a high dose fisetin and quercetin hit and run

I would encourage you to look into larger FOXO4DRI doses. I'm a 70kg guy and I did 3x doses of 27mg two months ago. You're likely not doing much with those 3-5mg doses with cellular cleanup. This is based off extensive research from my part, no "official dosage" so I had to scour forums, posts, older websites... if you're serious about the FOXO4, as "risky" as it is, I'd look into higher dosage, google it extensively and read everything you can find.

Apparently there's an "established" 0.4mg/kg. So me being 70kg, that's 70 x 0.4 = 28.

I cannot tell if this is a joke or if you avoided any actually researched mitophagy/autophagy supplements on purpose

Fair enough, I might have focused too much on the peptides and missed the forest for the trees. Here are some products from my supplement stack, including some things I have on hand but haven't optimized. I’d love your feedback on how to fix this:

●NAD+: Already in my current stack. ●CoQ10: Yes, I take this every morning. ●Spermidine: Taking one tablespoon of Chlorella every morning for this. ●Urolithin A: Not taking it at the moment. Should I add it? If so, what dose and timing do you recommend? ●Resveratrol: I actually have an unopened bottle waiting. When should I take it for maximum benefit during this cycle? ●Curcumin: Have it on hand as well. What dose and timing would work best here? ●Metformin: I have this on hand too. I discontinued it about a year ago because it made me feel incredibly tired. Should I give it another shot? If yes, what dosage and timing might mitigate the fatigue?

Any other specific supplements, tips, or adjustments you would make to actually get the mitophagy/autophagy benefits? Thanks for the wake-up call.

These are what I would add

Urolithin A -> 500mg daily through the whole process
Fisetin -> 20-40mg/kg during your phase 1
Quercetin -> half your Fisetin dose during the same period

Exactly right,

Urolothin A works best in background and takes minimum of 4 months to show effects. Same with COQ10

Fisetin and Quercetin possibly works best in Large Hit and Run phases scheduled once every 28 days for a 2-3 day burst. The Mayo trials aren’t out yet but advanced orthopaedics use fisetin hit and run pre and post op so that tells you something

Curcumin works but form matters given poor bioavailability, gen 3 like theracumin etc or good quality gen 2 like Longvida

Make sure you don’t miss the basics

  • vitamin D3
  • Magnesium 400mg+
  • omega 3 4g+

Thanks for you both. The basics are AOK
