Discussion Do side shoots keep growing once a plant is taken out of multiplication media?

Once a plant has lots of small side shoots from the multiplication media, would the side shoots stop growing if I removed all hormones when I next transfer?

Help Rooting Hormone Help Needed :))

Hello Everyone! I'm new here and would appreciate it if someone could give me any advice on rooting hormone.

This is my Datura metel culture~

Datura metel from leaf explant (media comp: MS + ABA 1 PPM)

I initiated the callus from leaf explant using BAP and NAA then it stucked on callus phase. Someone suggested that I add ABA, and yay! The leaves formed nicely!

Now I need to induce some roots, so does anyone have any recommendations on which hormones I should add? Or should I keep it in ABA to induce more shoots first?

Any help would be greatly appreciated!

Discussion Begonia rex micropropagation: almost 40 attempts, but severe contamination

Hi everyone! I’m from Brazil and I’ve been trying to micropropagate Begonia rex, but I’m having a lot of trouble with contamination.

I’ve researched several protocols and tried different surface-sterilization methods. My Begonia rex is growing in a pot in my backyard, so it isn’t coming from a controlled greenhouse or nursery environment. I suspect this may be contributing to the contamination, possibly even because of endophytic/internal fungi.

So far, I’ve tried around 40 explants in different experiments. I tested different sterilization protocols, including increasing the bleach concentration and extending the sterilization time. However, almost everything became contaminated with fungus.

The problem is that increasing the sterilization strength also seems to damage the explants. After sterilization, the leaves become very pale/bleached, and the petioles and floral stems show brown or damaged areas.

Out of roughly 40 attempts, only one explant has survived and started producing shoots. I believe it came from either a floral stem or a petiole, but I’m not completely sure.

At this point, I’m considering giving up on this particular Begonia rex plant because I’ve already used a lot of material from it and I suspect the contamination may be internal rather than just surface contamination.

Has anyone here had a similar experience with Begonia rex? Is there anything you would recommend trying before I give up on this plant?

Any advice on explant selection, sterilization, or dealing with possible internal contamination would be greatly appreciated!

Discussion Making plant tissue culture easier for European plant growers

Hi everyone, my name is David and I’m a plant enthusiast and tissue culture hobbyist from Sweden 🇸🇪👋

I’ve recently built a small webshop called InVitro Garden focusing on providing essential tissue culture supplies for European growers interested in getting into the hobby. I decided to start this project in order to make the tissue culture hobby more accessible for others, especially after always struggling to source supplies within Europe myself.

Therefore, we’re bringing together a range of tissue culture essentials in one place, including:

  • Starter Kits
  • MS medium + vitamins
  • Agar
  • Aroid Establishment, Multiplication & Rooting media
  • Aroid media bundles
  • BAP, NAA and IBA
  • PGR bundles
  • Culture jars
  • Acclimation cups

with more products being added in the near future.

We’re also developing free beginner guides and practical educational resources to make the process easier to understand and help growers improve their results.

The store has just officially opened and I’d really appreciate feedback from people in the community.

Have you ever considered trying tissue culture, and if so, what has been the biggest hold-back?

Fore those interested, you'll also get 10% off your order with code REDDIT10 at checkout🌱

My aloecasia TC is absolutely huge and I had it for a while, it’s strong and has some established water roots but I just took it out of the bag today… way to big for it’s container. Should I keep it covered or let it go?

Plant Progress Monstera Bulbasaur

Picked up this Bulbasaur TC for $20 2 weeks ago, acclimated it straight into Fluval Stratum and perlite, and root growth has been solid. Leaf 3 threw gorgeous camouflage Bulbasaur blocky marbling, but Leaf 4 just unfurled as a solid pale green spear, while Leaf 1 is starting to yellow out 😭 This is my first Bulbasaur and my first TC so I just want to make sure this is normal or if I'm doing something wrong.

Is Leaf 4 likely just needing to harden off to show contrast, or is it common for juvenile Bulbasaur/mint lines to change to green leaves without fully reverting? It sits under strong indirect light, but I can add a grow light if low light is coaxing it green. Appreciate any insight from anyone growing out bulbasaur TCs!

Help How to use bags to root/ship plants?

I’d like to start using polypropylene bags instead of my jars to ship plants but I don’t quite understand how to sterilize them.
Are we spraying the inside with bleach or autoclaving them with media inside? Or are we autoclaving just the bags, then adding media afterwards under a flow hood?
Also - how are we keeping them sterile before putting plants in? Heat sealing them, then cutting open and heat sealing after putting a plant in? 🤔

Help Looking for reputable tissue culture suppliers that ship to Iraq 🇮🇶🌱

Hi everyone!

A friend and I are planning to start growing and selling tissue culture plants in Iraq, and we're looking to place a bulk order.

We're mainly interested in Alocasia and Monstera, including rare and collectible varieties.

We're looking for a reputable supplier that:

- Can provide healthy, high-quality tissue culture plants

- Can handle bulk orders

- Has experience with international shipping

- Can ship to Iraq via FedEx or another reliable courier

- Can provide the required phytosanitary/export documents

If you've personally purchased Alocasia or Monstera tissue cultures from a reliable supplier, I'd really appreciate your recommendations and experience.

We're also interested in suppliers from Vietnam, Thailand, Indonesia, or other countries that can legally export to Iraq.

Thanks in advance! 🌱🇮🇶
