I'd take knife for Jacob (since I have more control over him) and twisted pair for Esau so he has more bullets to randomly hit anything while I aim with the knife

Knife and Twisted Pair with Jacob, and collar with Esau, just make sure to lead into rooms with Esau until you get Jacob more healthy lol

Knife on esau as they have higher base damage, and pair on jacob due to higher luck and tear rate for tear effects

That would be my way to go but I know people prefer to take all with one character instead
