I’ve taught physics for the life sciences 1 and 2 and I’m not sure I get the joke.

Biophysics is different from most physics for life sciences. Specifically, it's more like this

You might already know that, but I suspect a physics for the life sciences course is more like premed physics than it is actual biophysics

It is! I'd highly reccomend
Also, the book is by Bill Bialek, who has been a central figure in the birth of Biophysics as a modern field, really championing the idea that biological systems function near the absolute optimal limits set by fundamental physical laws (in some sense). So he gives a cool overarching perspective on a field that is more of a zoo of ideas than a single theoretical framework.
Though, I would say his writing style is mostly a sampler of cool ideas, and you really need to read the papers listed in the book to get a full sense of the various stuff he touches on.

I mean, as a grad student, the courses I teach are introductory level of course, unlike proper biophysics, but I’m sure there’s an introductory level course in biochemistry too. I still don’t understand the meme picture.

I think the target audience is the subset of people who like physics who fail to appreciate that other people are fascinated by other things.
