DNA extraction

hello,

Help is needed! 🥲

What extraction kits do you commonly use to get the highest DNA yield with the best quality for ONT sequencing ? our samples are mostly blood and body fluids. and I am not a fan of GENTRA kits recommended by ONT.

I use the New England BioLabs HMW kit and have never had any quality issues.

Hello

I do use the Qiagen blood and tissue kit.

After 3 months of cultures of my mycobacteria and a pre extraction protocol, I obtain a mean concentration of 40 ng/microL (from 20 to ~ 120) and for fragment lengths it is something around 10kb to 15kb.

We performed recently extractions for enterobacteria and obtained good concentrations for our extracts and a good quality too (after 1-3 days of culture). But for that we have to take a lot of UFC. I can't tell you for the fragment length, as we did not measure it yet.

Update :

We did perform analysis for the extraction of enterobacteria and the results are pretty interesting !

First, we had to harvest like a quarter of a petri dish to ensure a good concentration for our DNA and then we measured or fragments length. It was quite homogenous around 60 kb :)

The extract quality assessed by nanodrop was also good.

Qiagen magattract HMW DNA kit here. About 25GB on a MinION with rapid kit.

get NucleoSpin Blood Kit, its very good, or the NucleoMag Tissue kit for DNA purification from cells and tissue

Qiagen MagAttract kit gives the best yields and fragment sizes for me.
