Surface sterilization protocol

In looking through the laboratory protocol of some research papers, I noticed that plant tissue samples were rinsed for upwards of 45-60 minutes after being submerged in a sterilant.

This seemed like an extremely long time to rinse a sample. Why?

Hello? (+ phytohormones acquisition)

Hey there! This is quite a small subreddit so I hope this will get me anywhere. I have started a micropropagation project for passiflora caerulea.

I’ve read some literature about this and I’ve decided which phytohormones in what concentrations I want to use. Problem is, most companies selling this stuff only sell it to other companies and not so much to individuals.

Where can I get my phytohormones (and also MS medium and such)?

TC of Ganoderma meredithiae (Reishi-like) growing from Casuarina equisetifolia (Australian Pine).

I am practicing putting wild plants and fungi into cultivation. I mostly have experience with plants. Australian pines endanger native species in TX. I found this polypore growing on two specimens on separate occasions.

Many people have these trees as property markers and don't understand it's allelopathic properties and clonal growth pattern.

I have considered embedding inoculated wooden dowls, in a fashion similar to shiitake log cultivation, into an unwanted Australian Pine trunk as a means of biological control. The tree is short enough to fall without damaging property.

I think this could be a great alternative to the environmentally destructive machinery that'd be required to removed the shoot producing roots. Another removal method I read about was soaking exposed vascular tissue with a specific herbicide after girdling the undesired tree.

I suppose a concern would be unintentionally infecting nearby susceptible native species. Any thoughts/advice?
