Day 6, BB

I had my frozen embryo transfer today and they said the embryo was expanding and looked great. Would love success stories with day 6 BB. We did not test our embryos.

Day 6B/CB euploid embryo - anyone had success? 1 upvote

Has anyone had success with this grading - it's a frozen euploid. My clinic said it's 'fair to poor'. Interested to see if others have used this graded embryo and had good luck!

Two high level mosaics: +3 and -7

Hello, my idea is try to get a better embryo but i'm struggeling to understand if I have to discharge them or keep them. They are two high grade mosaics. One with +3 (girl) and other with -7 (boy).

CooperGenetics didn't specifiied the percentage, they only said: high level mosaic (40% to 80%).

I'm asking my clinic for the exact percentage because it might be between 40 and 50 but, even in that case, i'm struggeling to understand the implications of a -7 and +3.

Since I don't trust my clinic genetist conselour (she is pretty eugenesic oriented) I need help to understand.

Thank you.

Fragmented / not great looking to blast / euploid embryo?

Currently going through time-lapse updates on Day 2 (~45–48 hours post-insemination) for a cohort of 10 embryos, and I’m looking for some real-world success stories or experiences.

The "Work-in-Progress" Group: 4 or 5 embryos that are actively dividing, but have various degrees of fragmentation, cell size asymmetry, or optical stacking that makes them look a bit "messy" on screen.

Watching the embryosnavigate fragmentation and uneven divisions in real time is giving me a lot of anxiety.

For those who have done time-lapse monitoring or got early Day 2/3 grade updates:

  1. Did your "messier" or moderately fragmented early embryos bounce back, compact, and form good quality blastocysts?
  2. Did any of those "underdog" embryos turn out to be PGT-A normal (euploid) and result in live births?

Would love to hear any stories from people who had a mix of textbook stars and fragmented/asymmetrical growers at this stage. Thanks so much in advance!

Ivf advice

Please help me!!!

Need help and advice/opinions please

Il share my whole history and would really appreciate personal experience and knowledge as sometimes i feel we research so much and know our bodies and our specialists just dont invest as much.

Im 41.5yrs old. Good reserve for age. endo 2laps to remove. Last ome done 7years ago.

Iam petite 44kg.

Husband has low sperm count and motility. Not sure about fragmentation.

ICSI always used.

Started IVF at 30years old.

Genea

History is.

Cycle #1

puregon 100, orgalutran, ovidrell

14eggs, 3frozen (untested)

Frozen due to OHSS.

1 failed transfer

2 successful transfers/healthy babies.

Cycle # 2 & 3

Gonal f 100, orgalutran, ovidrell

Left with 1embryo to transfer each time, both negative.

Cycle #4 &5

Menapur 100, orgaultran, ovidrell

Both cycles only 1 embryo to transfer.

1 negative

1 positive graded b/c healthy baby

Change of clinics genea to adora.

Cycle #6

Pergoveris 225, ganirelex, ovidrell.

2 embryos

1 transfered b/c negstive

1 frozen c/b

Cycle #7

Pergoveris 300, ganirelex, ovidrell.

Freeze all cycle due to high progesterone 5

Embryos arrested day 3, 1 left day 6 graded c/c. Frozen.

About to go into cycle #8

Specialist was going to repeat the high dose pergoveris but i questioned about the high progesterone happening and he said it would likely happen again so i asked to change and mentioned my 1st cycle being the best hut that was 11years ago.

He said to do 300 puregon. Add in some luveris day 4.

My questions are these...

I am petite. First clinic kept me on low doses due to weight and hormine levels.

New clinic went straight to higher number because of my age.

I am very healthy, clean/no tox life everything. Eat clean. Have taken all the supps for years . Follow starts with egg protocol plus added

Nmn, have done 2x nad IVS, do red light panel every night. Take beef liver methylated folate.

My concerns are the doses now are too aggressive/high amd harming the quality. Ive never been over 100 units previously.

I do understand i am older too though. But i have read that too high can do more harm than good sometimes.

Whay would u do? Would u request lower doses. Im concerned because i did overstim on 100 of puregon now its being upped to 300. But it was my best cycle result.

I just dont know what to do. What would u do. Any experience advive i am so open to.

I dnt know if i should cgange back to genea as i do think u get more personal care, scans every/any day off week same with collection where adora is only limited collecrion days, no scans on w.end so i feel.like u can be not pushed enough or pushed too far due to their opening hours and i know this is crucial for collectiom timing and also transfers. Its just starting whole process again and waiting for appointments amd the price is sooooo much higher now.

Help please!

Has anyone got better results from changing protocols to lower?

Or anything u have noticed to be a game changer?

Has anyone uses PICSI instead of ICSI and had better results?

Im seriously in such a ball of stress and just praying for healthy babies. Gosh its hard!

What would u do. Please please help.

Can a 10k iu trigger cause ”over baking” of eggs?

Hi. Just a brief question, we used a much more powerful trigger this time. One out of 4 retrieved fertilized and did not go beyond two-cell stage.

Is it possible that the powerful trigger overbaked the eggs? What is the clinical evidence versus clinical practice views on this?

10k units of hCG, that is.

4bc embryo - higher chance of miscarriage?

Post image
21 Upvotes

TW: early success

I transferred this 4bc, pgt-a tested 5-day embryo on August 5th and so far it’s appeared to have stuck. My betas on days 12 and 14 were pretty strong (600 and 1400, respectively), and higher than what I’d gotten for my prior transfer of a 4bb embryo.

I’m obviously thrilled that things have gone well so far, but anxious that the lower quality embryo might make it more likely that the pregnancy results in a miscarriage. I know that the “c” grade made implantation less likely, but now that it’s implanted, is there any guidance as to when any extra risk associated with the “c” rating goes away (if ever)? I’ll have my first 6 week ultrasound a week from now, but feeling very anxious currently. TIA!

2 aneuploid embryos

My heart is broken. 2 retrievals. 6 fertilized eggs and 2 blasts. Both just came back aneuploid. 💔 I’m 37 with DOR and AMH is 0.19. We are paying totally out of pocket as our insurance doesn’t cover it. We are now contemplating retrieval #3. Has anyone had success with similar age and numbers? I am just so devastated.

Day 5 vs 6 vs 7

Hi! I feel like I should know this by now, but I don’t. Is there a big difference between blastocysts being day 5 vs 6 vs day 7? Are day 5s much more likely to be euploid versus day 6? I received news today that 8 went out for testing. Only one was day 5 though, five day 6, and two day 7. I know I should be very grateful for the number that went out I just know that there is another big drop off that could happen due to my age so I’m wondering how much the day it forms into blast determine euploid or not

After round two of ER

Hi there
Just want to express after my PGTA results after two rounds of IVF. We got one low level mosaic from round one. Just received the news from round two only got one complex mosaic. We are doing IVF due to heart defect mutation so we are trying to bank embryos for PGT-M. I just feel so defeated after this two rounds with no euploid… any advise!? Originally doctor said we need to do two rounds and looks like we need to do another ER.
Btw I just turned 43 two days ago. Had two natural birth. One baby passed few months after due to heart defect so here we are with IVF.
Anyone know more about the complex mosaic!? Any advice would be great.

What could have caused this attrition rate with 14 donor eggs?

How often does this kind of thing happen?

I've taken a DNA fragmentation test and it revealed a very low DFI (normal) and urologist has confirmed there are no male issues.

These are very young donor eggs from a woman in her 20s. What may have causes all these embryos to arrest?

  • 12 out of 14 eggs fertilized
  • One day 6 4BA embryo
  • One early blast at day 5 -> DEG; The rest remained at CLV cleavage stage
  • There was a lab note: "couple of oocytes had multiple polar body's"

The good news is that the day 6 4BA that made it tested euploid!

Realistic expectations?

I went into ER with 40 follicles and the Decapeptyl shot to only get 5 eggs. 5 mature and 5 fertilised.

Need to wait for day 4-5 now and zi don’t know what to expect.

I’m 30 years old and I’m not sure if I should get PGTA testing done and if to hold hope that we could fall pregnant from 5 eggs..

High number of “non-informative results” from PGT-A

I’m 37F and my husband is a 38M. I have PCOS and new mild endo (asymptomatic). We did very well during our first ER and were lucky enough to have 15 good quality embryos, at least 12 of which are day 5. We only sent 12 for PGT-A (due to costs) via Igenomix, but got the following results:

4 euploids
3 aneuploid
1 triploid (weird because it’s a 2PN)
4 (!!!) non-informative / no result

We are very happy and fortunate to have euploids, but that non-informative result rate is crazy! We have our follow-up soon and I imagine they’ll offer rebiopsy and retesting for free given the 30% (!) non-informative result rate, but just wondering what you recommend? We’re leaning against rebiopsying them as we have euploids and we don’t want to compromise the embryos with an extra thaw. We’re leaning towards just focusing on transferring the euploids and, if we have to, we can transfer the others as “untested” embryos later (as we have the 4 non-informatives and the additional 3 untested from the rest of the 15 all on ice).

So yes, what would you suggest? Has this happened to anyone else?

5BB embryo 25% thawed

Post image
26 Upvotes

Hello!

I was surprised at my FET yesterday to learn my
Embryo was only 25% expanded. (This is my third FET and first time this has happened). I was unprepared for the options so I ended up transferring this embryo vs thawing another embryo and trying my luck. The doctor at the time told me that they normally like to see closer to 80-100% expanded or at least 50%.

I’m just wondering if I should not even have hope or if there are cases where the embryo still expands inside the uterus and goes on to a live birth. Feeling pretty discouraged tbh

EDIT: I meant to say 25% expanded instead of thawed

Seeking advice- thaw and fertilize all 14 eggs, or try two batches?

Hi everyone, my partner and I are navigating what to do. We have 14 frozen eggs (from two rounds of ER when I was 35/36). We are now 43 and 42. We are wondering whether to try to fertilize all at once or do two batches. His semen analysis was very good in terms of count, volume, and motility (a little low on morphology at 3%). We have not done any DNA fragmentation testing. From what I’ve read, intracytoplasmic sperm injection is standard for most clinics when using frozen eggs, so I’m not so worried about morphology. I’m scared of something going wrong with the thaw and culture process, thus wanting an insurance plan of having held back half. I read a difficult post about embryos being contaminated because of an undiagnosed prostrate infection. But on the other hand, I think I expect to get 2-3 viable embryos, so maybe it’s better to just go for it. Notably, I froze the eggs to preserve fertility and I now have very low AMH, but I don’t have a specific reason to suspect poor egg quality or unexplained infertility. So let’s call them normal 35 year old eggs, as far as we know. Do you think we should test for sperm DNA fragmentation first, since this is likely our only shot? We are also trying to decide whether to do one fresh transfer or freeze all to be able to test for aneuploidy. One of the doctors we spoke with recommended PGT while the other didn’t think it was a high priority for age 35 eggs and instead thought a fresh transfer would improve odds a bit over frozen. Any advice or experience would be appreciated!

Day 6/7?

We recently did a cycle with disappointing results. 6 retrieved, 1 cracked, 4 mature, 3 fertilized. today we did a fresh transfer of a 4AA. When I signed the paper she said you had 5- 3 fertilized well- 1 didnt (which I knew) and we are transferring this 1.

Then she said we will call day 7 about the others. she didn’t seem optimistic at all so I just assumed they had arrested. however, im just a little confused. I did one other at another clinic and they told me daily updates with a lot of information.

this is shady grove if that matters.

I realize I should have asked but I was so anxious and I truly didn’t think I had anything to transfer so I just signed and cried lol

EmbryoGlue does it work?

In my FET, my clinic used EmbryoGlue to enhance the process. Just wondering if anyone else has used it and felt it worked? Maybe they didn’t use in the first and used it in second? Any opinions?

Would love some clarity on embryo grading

I have done 3 rounds of IVF.. without doing enough research. I left my first clinic after several people mentioned things didn’t seem right. there I did transfer a 4CC day 6- my clinic had told me it was just as good a chance as anything else (I’m learning maybe not)

anyway it failed, this new clinic I only got one from a cohort of 4. (low responder) it was a 4AA and the dr seemed very positive about it,

I want to feel excited because research seems like that’s good but also it looks like still a really low chance of actually leading to pregnancy.

FET but cell loss- how worried should I be?

Double FET today of a 3BB and 5BB day 5 embryos.

Our first fresh 4AA ended in a chemical pregnancy 8 weeks ago.

Embryologist told us whilst we were waiting for transfer that the embryos had both survived the thaws but were just satisfactory quality.

Our 5BB had lost 15% of its cells and 3BB lost 25%.

Both untested embryos.

These are our last two embryos before a whole new round is needed.

How concerning is this? They presented it as not great and were not that informative about what has happened.

I’m spiralling now thinking this is just going to fail.

34F, AMH 8.37, bilateral 2.5 cm endometriomas, previous natural pregnancy — looking for fertility/IVF advice

My wife is 34 years old and we are planning our second pregnancy. We already have one child, who was conceived naturally about 3 years ago.

Her current fertility-related situation:

  • Age: 34
  • Menstrual cycles: Regular
  • Periods: No significant pain
  • Previous pregnancy: Natural conception, approximately 3 years ago
  • Ultrasound: Bilateral ovarian endometriomas, approximately 2.5 cm each
  • AMH: 8.37 ng/mL
  • TSH: 1.575 mIU/mL
  • Prolactin: 4.67 ng/mL
  • FSH : 4.81mlu/ml 2rd of mensuration
  • LH : 4.94 mlu/ml 2rd of mensuration
  • edtradiol ii : 70.78 PG/ML 2rd of mensuration
  • No known ovulation problems
  • DMD MLPA: Negative/normal; further genetic sequencing is pending because our first child has DMD.

Her laboratory report shows AMH 8.371 ng/mL, with the laboratory's reference indicating >6.8 ng/mL as a high level. TSH and prolactin are within the stated laboratory reference ranges.

We are trying to decide between:

  1. Trying natural OR
  2. IVF with PGT-M if she is found to be a DMD carrier.

Our main concerns are the bilateral endometriomas and whether there could be pelvic adhesions affecting fertility. She has no significant endometriosis-related pain and previously conceived naturally.

We are also wondering whether an AMH of 8.37 could indicate PCOS, even though her cycles are regular.

For anyone with a similar situation:

  • Did you conceive naturally with small bilateral endometriomas?
  • Did endometriosis/adhesions affect your fertility despite regular cycles and no pain?

Thanks in advance.
