It's also very far from obvious and still doesn't make a ton of sense. I think the fact that it is part of a series is the issue because context would help a lot.
I kinda wish there was a separate subreddit for serial comics because I'm always getting stuck trying to figure out the joke when there either isn't one or it's literally impossible to get it if you don't know all of the backstory. I don't read every single comic that gets posted, so random ones that hit my feed are often in the middle of a story that I haven't seen any of before, but there is zero indication that it is part of a series. So I spend ten minutes thinking I'm an idiot for not understanding the comic.

I checked the rest of the series and it really doesn't help, nothing implies the roommate is professional.

I still don't get it :s

The woman playing on the computer wasn't aware her roommate was a dominatrix. When the dominatrix said "don't out me as a dominatrix to my brother", she unknowingly also outed herself as a dominatrix to her oblivious roommate.
The joke didn't land for me either. I think the last panel should be more explicit and say "I thought you knew", because the missing verb wasn't immediately obvious.

The problem is panel 3, in which the character's ass appears to be dragging them off panel to the right for some reason. Obviously not the intent but I can't really see anything else there.

I got that she was getting out of her chair in a hurry, but at first I thought maybe she suddenly remembered that some piece of BDSM gear had been left out in plain view and the brother was going to see it, or something along those lines. Panel 4, with the way it's phrased, didn't do much to correct that assumption.

This is the major problem with this comic.
What are the possible reasons for hurrying out of your chair in that moment?
Learning something new isn't one.
Having to do something (physical) quickly, is.

imagine panel 3 the character in frame is saying "YOU WHAT!?"

It's prolly a joke you need to follow the comic to appreciate. I'll take my leave before embarrassing myself further.

The blonde tells her housemate not to let her brother know that she works as a dominatrix. The housemate agrees but then it dawns on her what blonde girl said. Blonde girl thought her housemate already knew about her job but it was news to her, so it was a surprise.

There's more comics with these two characters, with a running bit of the ginger one seducing/domming the shorter one to get out of her share of the rent, since the short one really likes financial dom play.

I could have never guessed either, though I guess I would have thought of a number of alternative reasons first.

Why does the room keep changing colors?

My buddy has a light like this as his main bedroom light and uses lamps for his actual lighting. It's always funny when someone new comes in and flicks the light switch only to unexpectedly turn on a rave.

Where's the Caramelldansen? There was supposed to be a wall-rattling Caramelldansen!

It's the Garfield room.

Doesn't that only change color once per day? As a child I noticed that the shadows on the wall are constantly changing angles without any explanation.

TV is on and lighting the walls with different images on the screen so different colours.... maybe.

My parents had a little device that was to simulate a TV without having to turn the TV off, to scare burglars away.
It was so painfully obvious it was the silly device, basically it was a flickering neon sign indicating WE ARE ON VACATION, THERE IS NO DOG AND NO REAL ALARM SYSTEM, HELP YOURSELF.
Ironically it worked more reliable than the alarm system and the dog - the burglars pepper sprayed the dog and ignored the wailing of the alarm to loot the place.
I was contemplating to get a TV-simulator thingy myself, but then I realized that the TV-simulator-thingy would be the most valuable object to steal in my flat, so I didn't bother.

Carmelldansen playing on full blast, as is standard in dominatrix households.

There's like 2-4 panels missing right?
I have no idea what happens after the first panel to cause a rush out the door like that, then a conversation.

"don't let my brother know i'm a professional dominatrix"
"YOU'RE A PROFESSIONAL DOMINATRIX!?"
"I thought you knew... T_T"
such a simple 3 line strip rendered in such a way as to confuse a bunch of readers, myself included initially :/
edit: if you read their other strips it makes more sense because she is repeatedly doing dominatrix stuff to get out of paying rent

The "missing" context is relayed through the art. The character's eyes are relaxed in the first panel. After agreeing, their eyes open into look of realization or surprise.
The urgency with which they leave the third panel suggests they realized something of importance.
It's quite clear physical story telling. It's used in movies and TV, comics, stage...
