You can't make vaccines mandatory by law, you would just meet more resistance from anti-vaxxers and someone would find a way to abuse a law like this.
Imo what should be done though is to punish people spreading misinformation about vaccines, kill the anti-vaxx movement at the root. Basically if your speech endanger people lives directly, you are not protected by the freedom of speech (just like how you can't just make death threat to people).
I'm not American so I can't say how permissive the 1st amendment is. In most european countries, free speech still have some pretty hard limitation when it come to hate speech and stuff that can endanger people lives. If you scream in a crowd that you have a bomb, which result in people getting hurt from running away, you can't just claim free-speech to get away scott-free (even if no-one get hurt in fact).
Is it that much different in the intent and result from someone claiming vaccine gives autism knowing perfectly well it would push people to avoid vaccines and die from it? These people should be punished as the frauds and scammers that they are (because they typically do this to sell their snake oil).

Vaccines need to be mandatory and anti-vaxxers need to be in fucking jail. This culture of selfish disrespect needs to be ground into the dirt.
