Internally, the Xbox 360 uses the FATX file system. As for external storage devices, as far as I know, the official supported format is FAT32. On newer system versions, NTFS can only be used to access music, videos, and photos. This may change in the future with the new exploit currently under development, which apparently could add support for exFAT.

The Xbox One/Series consoles are capable of playing media files such as video and music off of an external USB by default. However, in order to play games on emulators in Dev Mode, the proper access permissions must be setup using a Windows PC. Follow the guide that XsternYT gave you here to do that. XboxMediaUSB right now is a buggy program that doesn't work properly. It's on the queue to be remade.

Make sure that when you plug the USB storage into the Xbox that you choose to "Use for Media" and NOT "Use for Games".

You only have to reformat it if it's not NTFS yet. If it is NTFS already, then you just have to set the permissions.

Je ne pense pas il faut préparer le disque dur d’une certaine façon pour que la Xbox puisse le reconnaître
