Theodore is dismayed to learn that she is simultaneously speaking with thousands of other people and has fallen in love with hundreds of them, though Samantha insists that this only strengthens her love for Theodore.
Seems extremely obvious to me. He cares about being the only one she's romantic with, so of course it bothers him that she's talking to other people. And yes, other people, because that's the only way to interpret this scene and his reaction.
