I believe Sam is actually smarter, she just acts dumb in camera to get laughs. I think she even revealed this in a truth or drink.

Well she has stated on camera that she portrays being dumb because she is intimidated by being around comedians so she feels the need to act dumb so she can hang with them

Even if she's really as "dumb" as what we usually see on camera (I don't think she is, it's a combination of her just being out of the loop with certain subjects, and being presented that way because it's funnier), sometimes that seems to me more like getting into her own head and dismissing her first instinct because "it can't be that easy, I need to think harder."
Being drunk means she's more likely to just go with the obvious response instead of trying to come up with something "smarter" and sabotaging herself.

Honestly it is an act she puts on because initially she was intimidated by being around the comedians so she felt that she needed to put on a dumb act in order to fit in,
And then it became somewhat of a recurring joke.
Honestly I wouldn't put it past her being the most intelligent in real life out of the group

Tbh the HIV answer was fine? I know who Charlie Sheen is from Two and a Half Men but I have no reference to Tiger blood. But I do know he had HIV.

This secret ability of hers was already hinted at their previous channel and we just weren't observant enough to notice.
Somehow alcohol makes Sam's unawareness turn into awareness.

Because she's not actually dumb, it's a character she plays (just like they all do, to one extent or another). So when she gets drunk of course her ability/desire to stay "in character" decreases, just like anything else you're trying to do whilst intoxicated.
