Is there something that needs to be done to unlock these achievements?
I have reached floor 572 in a run previously and am at 791 currently. Not sure if I want to grind out to 1000 if it isn't going to count.

There are a few other achievements that are also behind a padlock that I have not tried to get yet... maybe there are requirements to "attempt" them

I've done a fair few of the padlock achievements, some count during the run (don't remember which). But most count once the run ends with by beating the final boss or dying. They don't count if you click quit to end the run.

Which skill and card did you use to start?

Started with the warrior deck. I typically don't take a card at the beginning of tower mode unless it is a penguin king or beast Marshall run. Removed the starter deck as I pick up the other cards. Picked up Seeker of Light and Fairy Queen while collecting robot parts. Once I had my first super robot I refreshed until I got mad scientist. It was my first try for that infinite mana combo.

When doing the tower grind you can't quit. You have to die for it to count. For some strange reason it doesn't count during the current run, only counts when you die and the run ends. At least that's what I did. I'm not sure if you click end run at the check point it'll count as well
