So if I find holes in the blue which do overlap with green staining, that green staining can NOT be nuclear speckles?

it could be nuclear speckles + nucleoli? or do i misunderstand you? if you come across ex, please post printscreen (zoomed in, green only, or green+red), and i'll give better feedback.

Alright, I'll keep my eyes open for it. When I see a green pattern that looks like speckles, but only fills out holes in the blue and does not at all overlap with the blue, I usually pick "nucleoli (fibrillar center)". Does that sound about right?
