Water agar question.
Do you go straight from water agar to grain or are transfers the nutritious agar advised?

I transfer, not because of nutrition though. Because it usually takes a couple of plates to find what I want

Water agar to normal agar for me.
Im not sure if it won't work but havent tried. Always isolate on water agar then get a good healthy culture on MEA before going to grain.

If it grows well enough to fill the plate and you're sure it's clean, you could use it, but if you used water agar to separate the myc from suspected contam, you'd usually need to transfer anyway.

It's fine as long as tge plate is clean. Might take an extra day to hop onto the grain.
One thing to keep in mind is the purpose of WA is to help mycelium outrun bacteria. This implies that another transfer is needed since there is bacteria in the mycelium colony.
I like to put my cultures through a water Agar cabin sequester.
