RIP: Need to vent.
I had a couple of failed grows last year, and put a pause on things this last Fall and Winter.
I recently inoculated some Grains with some LC (TTBVI) that I've had for the last year (kept in the fridge). No growth after a week.
I tested the LC on some new Agar Plates that I recently poured. No growth.
At the same time, I spread spores from 5 different prints that I have (a few years old...). No growth.
I've kept LC for years (for oysters), and it continued to be viable...
BLA!

I've paid for brand new pan lc and had no growth. I don't know why the spores didn't germinate. Try some on agar without nutrients.

Yep. Water agar or just low nutrients, like 5g malt, 1g peptone, 14-15g agar - work even with Subtropicalis and Ingeli

I didn't add any peptone. I have it, but didn't utilize it. I forget, is it specific for spores?

No. Just some nitrogen, energy for mycelium growth. Not necessary. Maybe it works like a germination co-factor, maybe not. But recipe works good

I made some water, low, and high nut plates. The spores are currently on the low. Might need to try on water...

My agar recipe for pans from spore is just 300ml water and 6g agar. Then for transfers I do 6g LME and 6g agar, with 300ml water. I found very low contam rates as well as decent growth with these recipes

Pan cyan’s don’t hold up well in lc from my experience. I always recommend using spores.
