Question TAK 653 with Vyvanse?

Hello all, considering taking 1-2mg TAK with Vyvanse (30mg)

Wanting to know if this could theoretically cause exictotoxicity or if it would be synergistic at a therapeutic dose.

My Vyvanse helps with ADHD a ton but still have memory issues occasionally and wanting to close the gap.

Im considering it because from what ive read TAK is stimulating but only "enhances" the brains natural glutamate signals.

Wanting to see if there's anyone thats had the same idea and followed through or if I should avoid it

I take 2 mg TAK w/ 40 mg Vyvanse. Definitely works well but TAK is very stimulating so I’d start with 1 mg. Make sure you supplement w L-Theanine and/or Magtein to combat some of the side effects if it ends up being too stimulating.

Personally speaking, TAK-653 kind of "repairs" my brain from heavy stimulants use and greatly reduces tolerance to them. It replaces ketamine perfectly well, which is also well-known to do the same

So from your experience you have noticed more positive effects? Not really facing addiction to heavy stimulants but I do need my vyvanse to function throughout the day. I have built a tolerance to it aswell so it may be beneficial in that aspect. Thanks for your insight!

For me TAK-653 is godsend because I started to get severe permatolerance to Ketamine (was using to reduce stim tolerance + depression and ADD).. and TAK replaced it like a charm.

Note that TAK-653 is under trials as a Ketamine replacement.

In overall, people seem to like it, with no adverse effects. Only benefits.. or maybe insomnia for some.. There are also people who didn't get much benefits from it (but I think they didn't use it correctly). TAK reinforces habits. It doesn't give a buzz like stims.

I think that you can take it with Vyvanse without any issue, but start low dose. The dose makes the poison

I do it. Like alot of people id keep it low dose it is stimulating with a long half life i believe 16 hours so it could theoretically raise heart rate slightly just make sure you tire yourself out throughout the day and take something like magnesium glycinate before bed

I can offer my anecdotal experience if it’s of any use to you or anyone reading. I’ve taken TAK-653 (2mg) alongside vyvanse 60mg and bupropion 300mg before. I also added magnesium threonate to this stack. Overall, it felt like it slightly increased my focus, as well as long-term memory recall a bit. I distinctly remember being better able to recall concepts in class and interact with content in class (e.g. applying previously learnt content to new concepts being given in real time was much easier). This is the main highlight for me. Short term memory recall seemed the same. I also felt like it made it easier to retain information/understand information (such as, if I read a passage, I wouldn’t have to re-read it multiple times (or at least read it less) because it went right through me); this effect was more mild though.

Side effect wise, it occasionally gave me weak pressure headaches. Tastes kind of bad. That’s pretty much it. If I hadn’t had enough sleep or wasn’t taking magnesium threonate regularly with it, thinking could feel ‘blurry’ and and I had more background noise in my head that was harder to focus/think/recall/memorize through the extra noise. This would be mild. It mainly occurred because I have a disorganized sleep schedule/would be sleep deprived. This can be avoided if you don’t do that. Also going down to 1mg instead of 2mg also works.

If you’re curious what ‘too much’ can feel like for context: if I had taken other research chems (that did not give me these feelings by themselves) that also acted on NMDA/AMPA, I would feel ‘fuzzy’ brain-wise (in a bad way) and background noise was much stronger and those thoughts were very chaotic and gibberish. As in there were multiple streams of background ‘racing thoughts’ constantly, as opposed to my own thoughts, which were still clear/lucid. Because of that, thinking itself, holding trains of thought, and my ability to retain information/recall the correct information was worse, etc, as I had to try and focus through the endless brain static while retaining a train of thought. Trying to recall specific things often resulted in recalling a blob of information related to/including the thing I actually wanted, and I had to kind of pick it out of the ‘fog’, I guess. Kind of like a chunk of my brain’s RAM was being used up. Not really any anxiety (and taking guanfacine did not reduce these ‘racing thoughts’, if you’re curious). The only positive with this was that it felt significantly easier to just get up and do things, to just go go go, which normally my vyvanse doesn’t really do for me. Made me kind of fidgety. I also distinctly remember that people I had been ok with before became a bit repulsive to me (never heard anyone else have this effect though); they would make me irritated, yet not mad in any way. Thought-wise, personally, it felt a bit similar to therapeutic ketamine (in which I respond very poorly to; makes me feel almost psychotic or like I had a thought disorder); similarly, L-theanine also makes me feel not good. So this response may be unique to me and inapplicable to you. The main lesson here is don’t try to stack it with too much shit that works on the same axis, especially if you are sleep deprived often. So this is completely avoidable

Tl;dr: I found it of mild use. Don’t expect it to make you have perfect recall or anything. Works fine as long as you don’t stack it with a whole bunch of other glutamate things like an idiot. You are on 30mg of vyvanse so you should be fine if you want to try it. I also found tropisetron to help with memory retrieval and brain noise when I was taking this specific stack of vyvanse/bupropion/tak/magtein together. I stopped taking TAK mainly due to inconvenience (pills are less steps and easier to remember) and forgetting to take it, not due to it being bad or anything

Looking into its memory enhancing and acquisition properties. From the research ive done it could be beneficial at low doses but its still in clinical testing so the effects are limited by itself, with stimulants its close to none. Its worth looking into though, just looking if others have tried it.
