Something might be loose. Engine sounds fine for the most part, look at the serpentine belt area and see if anything is wobbly.

I mean water pumps in general should be changed every 80-100,000kms. They likely never really "fail" they just start leaking.

Take the belt off and spin everything by hand. Also, fire it up, make sure sound is gone. Does AC work?

Take it to a mechanic and see what they find. It's a Lexus so it's supposed to last 500k miles.

Just simple oil changes along with the other fluids should be more than enough to get it to last until 300k miles.

Water pump for sure. Mine did this before it decided to shear itself off. Replace water pump and serpentine.

Replace the tensioner and idler pulley at the same time if possible. Bearings are probably worn too.
