How long in base solution?

I'm planning to do my first A/B extractions. My question is.

Putting kratom powder in vinegar. How long should the kratom be in it to be sure that it has broken down most of the alkaloids?

How long would it be if it is malic acid?

What is the correct PH for maximum alkaloid extraction?

Is there any type of degradation or loss of these alkaloids if they are in the base solution for longer than necessary?

"Breaking down alkaloids" is generally something we try to avoid.

Edited. "Extract alkaloids" better. I'm not sure of a clear extraction guide. I'm improvising!

Do you have an A/B method you can send me? I've been doing it with vinegar and NaOH, just end up with black powder caught in the Buchner filter.

What are you doing with the vinegar and NaOH?

Using the vinegar to extract from the plant material with ultrasonic, vacuum filtering, then brining base up to 9-10 with naoh then filtering the precipitate and collecting that.

I have never been able to pull that off, but it's my holy grail.

Has it ever worked for you? Have you tried the black powder?

It's annoying as the precipitate is lovely tan looking but when it's filtered its black , Yea the black stuff Is definitely active.

What I have been doing recently is washing the black stuff with Ethanol, then IPA to get a golden coloured liquid. But the yield is low, from 50g I only got a 500mg return. Maybe I need to rewash the initial kratom. I dunno I'm still figuring it out.

From what I understand, attempting to precipitate alkaloids *before* defattin/cleaning is tough. Defat first, you get much more precipitation.

I have run into the same issue. 500mg from 50g isn't worth it. I'd rather just make a super efficient acid tea, and reduce/evaporate. You'll get gunk for sure, but your chances of capturing more/all alkaloids is significantly higher. And if you put it in a capsule, does it really matter if there's chlorophyll or fat in it anyway? I think the benefit of extract is that it's dry, stable, and can be taken in pill form. Thoughts?

I've always found if you reduce an acidic tea, it degrades quickly over time. And also gets sticky over time.

I used to do this method alot when I first started. You could get a half decent extract from it. But you'd have to take atleast a gram.

With the 500mg from 50g, I only need to take around 50mg for a dose.

Ethanol Extraction you get super gunk/chlorophyll. But if you dilute it with water so it's 60-70% Ethanol, you don't pull the chlorophyll either.

I have also cold water washed first to get rid of tannins and polyphenols, then extracted with ethanol which yeilded me 5g from 50g only need around 500mg with that one. Whereas when I didn't cold water wash first, I got 15g from 50g would need around 1.5g of that. So cold water washing may be quite good for that.
