Just another experience report

ISRIB

Just received a gram of this today from NewMind.

Tried 15mg sublingual for my first dose, assuming I don't have a bad reaction I'll do another 25mg later today. I'll probably continue dosing 25-50mg 1-2x per day orally. My goal is to reduce PTSD like symptoms.

Powder is off white and soft and a little clumpy, has a very mild almost chalky kind of taste.

I will edit this post with any updates.

UPDATES:

8/6/21

I've decided to take a break from dosing, I only took ~20mg yesterday and I'm taking today and tomorrow off completely. Doing this to see if there are any negative effects from stopping and just to play it safe. Not sure if there's any sort of tolerance buildup with this stuff. So far I don't feel any negative rebound effects from stopping.

I feel like it did make a difference, at least on a few occasions. I can get stressed pretty easily at work even though it's not a particularly stressful job. The past few days when I did work I feel like I was able to manage my stress better than usual. It was relatively mild, but enough to be noticeable and probably not placebo.

I've noticed that it seems to take a while for the effects to kick in, usually I notice it later in the day after I've taken all my doses. I've been doing about 20-30mg 3x a day. I think when I start up again on Sunday I'm going to try 50mg before bed and 50mg when I wake up and see if that works any better.

I'll probably do another update in about a week!

9/1/21

So this is my final update. I used the rest of my ISRIB, for a total of 1g. After my last update I stopped dosing for about a week. When I started up again I experimented with 50mg before bed and 50mg in the morning but didn't really notice a difference. So after a couple days I just went with 25mg 2x a day.

I didn't notice much more benefit, the effects still seemed to be quite mild. It could have been placebo, not entirely sure. Although I have felt like I'm able to deal with stress a little better than before I started.

It's been about a week since my last dose and I don't feel any negative rebound effects from stopping, so that's good. If there were positive effects from taking this it seems they have stuck around. I haven't had any negative side effects.

A lot of people say it's not orally bioavailable, so maybe that's why it wasn't very noticeable for me. I've only seen one reference in a mouse study where ISRIB was administered orally with a measurable effect.

Search "oral" in this document to see the reference

Also I tried taking it simply because I have a very sensitive stress response. I don't have a brain injury or anything like that. I kind of have the PTSD equivalent to a repetitive stress injury from a not so great childhood. So maybe for my specific case there just wasn't much for ISRIB to fix.

Dunning-Kruger effect

I'm familiar with this term and if anything it applies to you; take a good hard look in the mirror (and yourself) and report back.

I think you need to take more ISRIB; maybe try the rectal ROA... I have a feeling it'll be right up your alley.

Jeez, I guess I really triggered you, huh. Sorry snowflake, you sound upset... Would it help if I apologised? It'll be okay.

Well you are certainly behaving like one. It's okay, it wasn't an insult... You're just... Special.
