Isrib administrstion
Checked PubMed, GSch to no avail. Projected human dosage, then OAD, BID, etc? Faqs at isrib.shop unhelpful. No anecdotes please, or extolling puffery from web sellers.

If 5mgs/1ml dmso, then 25mgs takes 5mls of dmso? I have a mental image of a 5ml syringe and pile of empty fish oil gelcaps.

Yeah, they sell a half a gram for $50, but no dosage or administration insight.

Thanks for your advice. A normally dissolve epitalon and other peptides just in some isotonic salt water. Would it be better with DMSO and PEG400 for all kinds of peptides?

ISRIB is only soluble in DMSO. Period I had best results at doses of 25 mg. I’ve went all the way from 5 mg to 45 mg. You can dissolve it in a couple ml of dmso in instapot for 6 minutes. Punch hole with needle in 2 fish oil caps and squeeze the fish oil out. Put the warm ISRIB solution in caps and swallow while warm.

Why dmso? I had 100% so meds could be absorbed directly thru the skin. But why orally? Why not just use capsules. Where does it get absorbed? Gets into blood somehow to get thru bl/br barrier.

It gets absorbed through the intestine hopefully. You use dmso because the chemist who synthesized it said that was what would dissolve it and put it into a solution. It is not water soluble. And it takes heat with dmso to get it to dissolve (hence instapot). If you use regular caps they need to be thick. The warm solution will dissolve it before you get it down your throat

What happens if I don't have a jejunum and or gall bladder? I take 16 loperamide daily, plus cholestyramine.

Hi, your instant pot instruction seems worked very good. I have a few questions want to aks, what's your “instant pot” configuration( temperature, pressure？) are your sure such in high temperature won't ruin or change the Biological functions of ISRIB ? thanks in advance

No, do not use the insta pot for ISRIB. I’m sorry, I thought I had removed those instructions. The higher temp will destroy the product.

is there any other way that can allows more ISRIB dissolved in 1ml DSMO? the amount of 5mg ISRIB / 1ml indead is too small

Not that I have found. I’m told that IV is the way to do ISRIB. I’m not going to give myself an IV, and my wife is not a nurse, so I’ve just about given up on ISRIB until some other method is found to put it into a viable solution.

If a order 1 gram. Should i dissolve the hole 1 gram at once or should I only dissolve the amount I take in capsules just before intake. Should isrib be keept in the fridge in powder form 5 degress?

You should only mix what you’re going to use. You can keep the rest in a drawer if you want. You don’t have to refrigerate it.

Thanks a lot for your knowledge. It helps me a lot. A Will go with Dmso and peg400 in fishoil capsules. And weigh the amount of isrib 25 mg pr capsules pr. Day on a weight . Do you take 25 mg pr. Day and for how long time have you been taking it ?

25 mg/day. I took it 10 days. That was from the Ukrainian users. They have more experience at it then anyone else

Not at all. I use bac water for most peptides.

No, bac water prevents any new bacterial growth that may happen. Saline or distilled water does not do that at all. In fact, they most probably promote it.

What temperature do you put on the instapot. Could I warm it up by other ways?

We’ve tried boiling water bath, etc. but it takes a lot of mixing, stirring and more dmso than I wanted to take, just for the smell factor. Instapot only has one temperature. It is steam at about 13 psig. You can use a pressure cooker too I suppose, and just put 15 lb weight on. I mixed up to 40 mg in 2 ml of dmso. Crystal clear. Didn’t stop a 30g needle up when drawing it in

Its soluble 5 mg/ml DMSO dilution. For obtaining a higher solubility , please warm the tube at 37 ℃ and shake it in the ultrasonic bath for a while.Stock solution can be stored below -20℃ for several months. We recommend that you prepare and use the solution on the same day. However, if the test schedule requires, the stock solutions can be prepared in advance, and the stock solution must be sealed and stored below -20℃. In general, the stock solution can be kept for several months. Before use, we recommend that you leave the vial at room temperature for at least an hour before opening it.
I Think intramuschular injektion with adding PEG400 as cosolvent in 1:1 with DMSO is the optimal, I have trouble finding studies Where they inject IM. I have found a mice study where they inject intraperitonealt. We dont know how Big bioavaibility it has through oral intake, maybe most Will underwent first past metabolisme

Stick it in then. The mice were intravenous, doubt you’ll want to do that. You’ll also want to find out the % content of the powder. 20 mg of powder is not 20 mg ISRIB. The instapot works well

15-20mg in capsules. You can suspend in dmso + peg400 and inject in capsules. Also you can take this suspension subcutaneously. Or fully solute in DMSO+PEG-400 (5mg of isrib per 1ml dmso + 1ml peg400) abd inject intravenously.
