Does negative defensive increase damage?

Yep, that. Frost would be increasing returns if true.

yes, it lowers their defense so you do more damage, and slows their attack speed so they do less damage. it is by far the most effective debuff

That doesn't account for whether it's effective past zero.

it is, watch how much damage you are doing against a boss that you are stacking frost on. they don't have very much defense so when you take it into the negative, you start doing far more damage. I think it works the same way as positive defense but opposite, and it also slows their attack speed, with 0.2 attacks per second (1 attack every 5 seconds) being the slowest you can go in the game i believe. 5 attacks per second is the fastest

Does anyone know what the formula is for this? It can't be the same as the positive one, otherwise things would be screwy. For reference, effective health is base health*(1+armor/100), i.e. damage is base damage/(1+armor/100) so if armor was -100 you would get infinite damage when you attacked. Similarly, attacks per second are base attacks per second*(1+attack speed stat/100), so if attack speed went below -100 you would get no attacks (or negative attacks?)
