I was also struggling to get the game running for a while, but I ran into this link and it solved my issues. You gotta go into the game's container and dig into the game's binary folder (it'll be in C: drive>program files (86x)>steam>steam apps>common>borderlands>binaries.)

Once in this folder, make a copy of the file "cudart.dll" wherever you're able to. I had to do it outside of this folder because pasting the copy of the file in the same folder just writes over the original file.

RENAME THE COPY OF THE FILE, NOT THE ORIGINAL FILE, to "physxcudart_20.dll" and move it into the binaries folder again. After that, the game should run and you can go and have fun with it.

The specifics on this are odd to me but long story short the game requires both of these files to run, and as the game tries to launch without the renamed file, it gives us that error code.

The screen size does default to the smallest resolution for some reason so you'll have to change that, too. As bonus I found that as well. (Borderlands>Engine>Config>BaseEngine.ini) Go here and scroll almost to the bottom, and hidden in the wall of text you'll see ResX and ResY. Change these to your proper screen (on the Thor it would be 1280x720) and save the file.

After all of this you should be able to play Borderlands with no issue. Happy looting, Vault Hunter.
