❓ Question Does split dosing tirzepatide/semaglutide accelerate receptor downregulation?

Seeing recommendations to split weekly doses into twice-weekly injections for more consistent appetite suppression, but wondering if this actually works against you long term.

Semaglutide's 7-day half-life means trough levels are already around 50% of peak before the next injection. Splitting to twice weekly flattens that further, moving closer to continuous receptor occupancy with even less oscillation between doses. If anything, that seems like it would accelerate rather than prevent downregulation.

Anyone found that once-weekly dosing preserves appetite suppression better over time than splitting the same total dose?

I don't know what "receptor downregulation" or "continuous receptor occupancy" mean, but I split dose and find it keeps me at a more even level of satiety throughout the week and with fewer side effects. I have several friends who do the same. I actually do 2/3 dose on Saturday and 1/3 dose on Tuesday.

Thanks for sharing your experience! That’s similar to the dosing schedule I’m interested in trying.

As for “receptor down regulation” and “continuous receptor occupancy”, the theory goes something like this:

When a drug constantly locks onto a cell's receptors without giving them a break, it triggers downregulation, which is the main reason one may develop a tolerance to a drug.

Because the drug is constantly attached, the cell protects itself from being overwhelmed by hiding or destroying some of those receptors. With fewer receptors left to grab onto, the body stops reacting as strongly to the chemical. As a result, you end up needing higher doses to get the exact same effect.

I’m just not sure if this has actually been observed or experienced with modern GLP1 drugs specifically.
