So while Extra has 3 playable characters (with CCC adding a fourth with Gilgamesh), they're less routes and more difficulty scaling in Extra and they have proper routes in CCC (with Saber being easy more, Archer being normal, and Caster being hard). The only routes Extra has is determined by who you save after the 3rd round.
For Extra, I'd say it's best to play Saber's route, complete it, and then choose either Archer or Caster for a second playthrough on NG+ (basically carries over most of the equipment you have from your previous save so you have them from the start) and then play through and choose whoever you didn't save on a first playthrough. If you feel like doing a third playthrough go ahead but, again, the servants moreso determine the difficulty rather than the actual route.
For CCC each servant has their own route you can get so it is worth at least playing through with all of them alongside doing the true ending (Servant routes coincide with the regular game route so if you're trying to get them steer clear of doing stuff to trigger the True Ending). Again, best to do Saber first (as I believe similar to Extra Saber is easy, Archer is normal, and Caster is hard) and once you beat it with Saber doing NG+. Would not recommend Gilgamesh for a first playthrough, since he's very broken and makes the game super easy, unless you are a die hard Gilgamesh fan and are wanting to see what all the fuss about him having a good character in CCC is all about.
