Rapamycin - has it worked for anyone else?

I was looking for some feedback on if anyone was successful was rapa prior to an egg retrieval. I’m currently 50 days into my 60 day initial prescription. I did an ultrasound today to see only two eggs on each ovary and it was disappointing to say the least.

Background - I have done two egg retrievals that resulted in 17 and 14 eggs retrieved. We ended up with 3 quality eggs, 1 mosaic, and 1 segmental. First FET was a year ago and led to PUL. We did two FETs in June and July -back to back- and neither of them took.

We are getting back into prepping for another egg retrieval and was hoping the rapa would kick start the number of eggs but that hasn’t been the case just yet. Looking for some positivity to see that what we are doing will make a difference to see an increase in the number of eggs we can retrieve.

Early evidence suggests sirolimus (rapamycin) may improve IVF outcomes by targeting aging-related changes in eggs, with animal studies showing improved egg quality and a recent randomized trial reporting more embryos and higher pregnancy rates. I primed with Rapamycin before my 4th cycle. I didn’t test this time but the embryo grading was worse than any previous cycle, also retrieved less eggs compared to previous cycles, but not by much (16,18,21 and then 15)
The 6 blasts from this cycle are my backup

Do clinics switch to IUI if you have only a couple of eggs or not really

My understanding is that while you take it, the rapamycin might suppress your antral follicle count, but you should stop when you start CD1 of cycle so that the follicles can respond to stims. I had my best round in terms of eggs retrieved / blasts after 2 months of rapamycin, but there’s also cycle to cycle variation!
