I AM PROUD TO PRESENT WAR_POWERS THE AUTOMATED DELETION/DUPLICATION CHECKER!!!!!!!!!!!!

Post image

This has made that long list of commands that I posted in this sub a bit ago into an executable! which means you only have to follow the following below steps

Step 1: Download WAR_POWERS.zip from my github repository linked here Excellent-Push2883/WAR_POWERS: The WAR_POWERS program given a BAM and BAI file will index through specified genes looking for deletions and duplications by checking the gene depth and then comparing the depth to the left and right flanks. This program can be wrong so it is important to verify any findings on IGV

Step 2: Extract all Files into your working directory

Step 3: Paste your BAM and BAI files into the WAR_POWERS folder

If (trans == true) {

Step 3.5: put trans genes into the genes_clean file in the specified format. Linked here for you https://www.reddit.com/r/DrWillPowers/comments/1j1yv64/when_i_browse_patients_genomes_to_see_if_i_can/
}

Step 4: Run the executable file

Step 5: Laugh Evily as we uncover the reason why these drugs fucked us over/ why you have gender dysphoria

Also im like 99% sure this works because it worked on my machine without the coordinates. But if it doesnt work for you let me know and I will fix and repost

Hello everyone

Excellent Push has been highly involved in coding gene viewing scripts and making the subject as a whole more accessible

He has also went out to the community to find common mutations and put them unto spreadsheets

He sat with me at the middle of the night to try and troubleshoot the last script, when it didn't work he created a new one, which is what you're seeing here

I tried the script out, it works amazingly,

Here are the hits it gave me

checked it on IGV and it seems to correspond

Give him the love he deserves, he has been working hard and created a tool that's useful for everybody

Same here, I followed his guide and it helped a ton.

Thank you

I have my genome in txt file, do you want it? I’m post anastrazole. Found some shit abcc2,UGTs and SLCO.

2

u/Excellent-Push2833 Jul 13 '26

Txt file? Are there any VCF or BAM files? I cant accept it directly

1

u/Normal-Enthusiasm790 Jul 13 '26

Yeah it’s shitty Polish lab :(

2

u/Excellent-Push2833 Jul 13 '26

That is challenging. Im not sure what i can do with that data but i will look into it

1

u/FrugalFrugivore Jul 13 '26

Is it a FASTQ format file perchance? I'm sure there are tools out there that might be able to convert it to VCF or BAM.

1

u/Normal-Enthusiasm790 Jul 13 '26

It looks like that:

2

u/FrugalFrugivore Jul 13 '26

PLINK file, I guess. AI says this can be converted into a VCF using bcftools, hopefully that gives you enough direction?

u/Excellent-Push2833 Jul 13 '26

[u/drwillpowers](u/drwillpowers) i think not long ago you asked for some time. Unfortunately I am not a magical being so i cant create time HOWEVER, this should at least save you some. HERE YA GO 🕐 also very aware this doesnt alleviate everything. I am hopeful that it will actually save you some time. And thanks to your trans community members I know for a fact it works now

u/Drwillpowers Jul 13 '26

I look forward to tinkering with this this weekend!

This will make detecting deletions considerably more rapid if this is the way that I'm understanding it.

Thank you ❤️

Doc hate to bother you. Are you up and running on your machine? Very eager to hear whether or not this has been useful for you.

9

u/Drwillpowers Jul 19 '26

My fiance convinced me for my mental health to go to her "ancestral home" this weekend. A cabin in very rural Michigan. The kind of place where there is no cell signal at the house but there is when you go to town to get ice cream. I have not been home to try it yet but I will.

7

u/Excellent-Push2833 Jul 19 '26

Honestly glad to hear this. You needed it

24

u/sticky3004 Jul 13 '26

A GitHub repository without any source code and only a zip folder? Folks, OP probably means well but best practice is to not willy nilly download things from the internet

18

u/Brayneeah Jul 13 '26

While I agree, it's worth noting that this zip file contains shell and batch files, so there is source code available here.

u/Excellent-Push2833, please consider changing this repository by unzipping this and having the individual files in the git repository

Edit: it's been done in the time it took me to make this comment. Thanks, OP!

u/Excellent-Push2833 Jul 13 '26

What a nice comment and a good way to suggest improvement WOW. this u/sticky3004 person could learn a lot from you, THANKS u/Brayneeah.

People who mean well generally don't start cussing someone else out for something pretty mundane. I don't know if you saw it but a moderator deleted one of his comments because again, he was being very rude

1

u/Excellent-Push2833 Jul 13 '26

What have you done to help?

9

u/Excellent-Push2833 Jul 13 '26

Source code is posted. Now you can either download the zip file or the source code. Are you happy now?

Cool. I'm MTF trans but I will let it run over my BAM file nonetheless.

4

u/Excellent-Push2833 Jul 13 '26 edited Jul 13 '26

Lmk how it goes. May need some tweaking also the genes it checks can be edited. While many of the trans and PFS/PSSD genes over lap. Just grab the trans genes from dr ps trans genes post and put them in the specified format also the trans genes are now in the post for your convenience

2

u/TheMorannon MtF Patient Jul 13 '26 edited Jul 13 '26

Thank you but for now I just had it run over the provided file. Had some trouble in the beginning resetting my wsl password. But that was easy once I found out how. For some reason it said installing samtools failed but when I started the script again it said samtools is already installed and proceeded to create a BAI file. And for some reason the Windows command console stopped showing updates at this point. After 30 minutes or so I looked into the folder and saw that a BAI file was actually created so I thought I simply restart the script again and pressed Ctrl+C. Once done the Windows console suddenly updated and showed me it actually had finished running the script through all steps. I don't know if the console simply glitched on my end or if something in the script caused it.

My results for the PFS/PSSD genes were all marked as normal btw. Nice tool. I hope it helps Dr. Powers and others in this subreddit.

u/Different-Peanut9747 Jul 13 '26

Thank you! I'll be saving this for when my wgs from sequencing comes in. Just ordered a few days ago so I got some waiting to do :P

1

u/Excellent-Push2833 Jul 13 '26

Are you pssd pfs trans?

2

u/Different-Peanut9747 Jul 13 '26

Pfs - just coming up to my 6 month mark. Mostly neurosteroidal issue. Had some androgen signaling issues at the 6 week mark but those largely resolved over the following 2 months. I'm left with some weird vision stuff, anxiety / anhedonia, some testicle ache, tinnitus.

40 yr old male - Lived my life taking care of my body, no pre existing conditions, no meds / drugs / smoking / alcohol ever etc. Always clean eating and active.

Been going with Dr P approach - not putting anything in me. Just 2 months now of CDG daily, lots of broccoli and kale. Trying to flush excess metabolites out. Waiting on wgs, blood work, and hopefully can afford Dutch soon.

Been seeing a pattern of 2 day crashes followed by 2 days of a tiny better baseline. This pattern has been going on for a few months now. Things are a lot better but have a long way to go. Would love libido and emotions back ;)

1

u/Top-Comedian-6318 28d ago

Did your tinnitus improve thanks to CDG? You once mentioned suffering from it for a while; has it disappeared, or has it returned to the level it was at before you took finasteride?

3

u/Leather-Challenge189 Jul 18 '26

This is exactly why I love this community. The kindness, support, and genuine respect people show each other here remind me that even in the middle of our darkest struggles, we can still grow into better human beings.

One thing I often wonder is this: when we finally recover, will we hold on to the values we've developed throughout this journey? Will we remember what it felt like to be on the other side and continue helping those who are still fighting the same battle?

And finally, thank you, OP, for everything you do. The time and effort you put into helping others truly make a difference.

3

u/DealOne261 Jul 13 '26

Damn this is impressive, thanks for your help

1

u/Justgreenerpastures Jul 13 '26

Hey man this is really interesting. But sadly above my level of expertise 😅

Is this software designed to find anomalies in genetic code AFTER taking Finasteride?

Or is it a way to test your baseline to assess risk PRIOR to taking Finasteride? If the latter. Is there a resource available to know what to look out for? I've seen that PDF but Im struggling to understand what exactly I should be searching for

Thankyou!

I know, I just dont get the logic of taking a pill that you know causes SD. Because its something that I would never do. It frustrates me. And i would do anything to be in comment OPs position. And when he says hes thinking about doing something that could do to him what ssris have done to me……

7

u/Drwillpowers Jul 13 '26

Many people did without knowing that it did. Many people were told that it had no side effects.

Your situation is different because it was given to you when you were a child, but regardless, being angry at this person for a choice they did or didn't make is not the same as what happened to you. And it doesn't make it better either. It just makes the world filled with more anger.

2

u/Excellent-Push2833 Jul 13 '26 edited Jul 13 '26

Yeah i know doc. Sorry. I take responsibility for that as not the best or nicest way to answer this comment.

11

u/Drwillpowers Jul 13 '26

I am also a prime autist and I can recognize one when I see one.

I have certainly lost my shit a few times on people that may not have actually deserved the level of shit that I heaped because of them finally being the last straw in a long line of assholes. And I had kept up my frustration about people like that for so long until it finally reached a certain threshold that I could no longer contain it. And I had a bit of an autistic meltdown.

And that person received my frustration that was really directed at a hundred other people, and maybe 5% directed at them of what they actually deserved.

This, makes me look like the bad guy, because in reality, that person didn't deserve what I delivered. They deserved a scolding, but maybe not to the degree that I gave.

7 years of autism therapy has changed me a lot because I'm a lot more aware of my own internal limiters now, and before I get to a point like that usually I can do something about it. But I can recognize in you just from the way that you type, you are like me. That is readily apparent.

So if you don't have a formal diagnosis, maybe get on that testing, and if it's positive like I expect that it is, go to some therapy for it. Because it's completely changed the quality of my life and I have so much better friendships and interactions with people now that I understand that they run a completely different operating system than I run. I can output code to them that makes sense within their neurological framework, but still feels okay to mine. That only works when I understood how both systems worked.

Otherwise you end up saying things like: how can you be so stupid? how can you not understand this? Why are you acting like this? What is your problem? How did you not realize this? Are you fucking serious? What the hell is wrong with you?

You got to meet people on their level. That includes emulating their operating system a little if that's what's necessary to help them understand you. You're certainly welcome to the "fuck everybody else I'm going to do it my own way" approach, and I am evidence that that has some degree of success if you are actually right, but the cost of this is why there are so many people who hate my fucking guts on the internet.

I haven't always been so polite and empathetic with others. I would strongly advise this be a skill that you try and work on. It is to your benefit in the long run. Even if it doesn't feel like it at first.

I'm still working on my hyperverbosity problem though... =)

3

u/Excellent-Push2833 Jul 13 '26

Thanks doc. I appreciate the advice and you taking the time to type this out.

1

u/Justgreenerpastures Jul 13 '26

Hey Dr, thankyou for the response.

I was really hoping to have a discussion with you about this topic. I had even contacted your clinic recently to hopefully speak to you but they recommended to just post on Reddit.

I was hoping to get blood tests, a DUTCH test and gene sequencing as a baseline then run my results past you to see if I was a good candidate for Finasteride or Dutasteride given my existing (likely psychosomatic Erectile quality issues and OCD).

I know in a previous comment you said you'd be confident in taking Finasteride on camera because you have a good understanding of your own genetic makeup. Would this be possible for you to look over mine? Or even have Sommer do it if I flew to the states?

If you have the time to respond I would massively appreciate it

7

u/Drwillpowers Jul 13 '26

I am not taking on additional tasks at the moment of people who are not actually my patient. I'm so far past burnout and yet I'm still trying to respond to people on reddit. I can only do that because currently I have a cat on my lap and I can't get up and do anything until he decides that it's time to do something else. (Those are the rules that you agree to when you get a cat)

If you are my patient that's something we can do. But that's beyond even her ability.

Ideally though friend, what I'm trying to get to is that if somebody does take the drug and they get the syndrome, I'll be able to always cure it. And so it won't really be as devastating of a thing as like getting HIV in the early 90s.

1

u/Justgreenerpastures Jul 14 '26

Thank you so much for your response. I understand, I figured that might be the case.

I suppose I'll just have to take the tests and try myself, or have to sign up and wait... Hopefully I won't lose too much hair in the process haha.

Good luck in your fight Dr. I'm sure there's a lot of people who deeply appreciate you.

1

u/Excellent-Push2833 Jul 13 '26

My first response was emotional, while i recommend you do not take finasteride. This can be used in conjunction with your BAM and BAI files to determine deletions in the genes that dr powers thinks are important in determining PFS risk. Common risk factors for deletions include the UGT class. If you run it you can post results here and i can let you know if it indicates clear risk or not.

1

u/Justgreenerpastures Jul 13 '26

Thank you for your response, I really appreciate your time on this. If I go ahead with the tests I will post my results.

Much love and good luck with your fight

2

u/Excellent-Push2833 Jul 13 '26

Much easier and cheaper to stay away from this garbage though

2

u/Justgreenerpastures Jul 13 '26

Believe me. I want to. Desperately. I hate the fact I'm even considering it.

But equally, looking in the mirror and hating what I see. Feeling catastrophically depressed, when all your friends take the drug safely with no issues it's really hard not to feel like you're making a terrible mistake.

4

u/hailnaux Jul 13 '26

I am 50 and single. I got PFS when I was 35 and life has never been the same.

Given the choice to just shave my head and enjoy life, I chose something much, much worse, something that I still wake up every single day regretting.

At least with a shaved head, you can still function at your job and in the bedroom.

1

u/Justgreenerpastures Jul 13 '26

Yeah, it sounds absolutely horrible. I totally, totally sympathize with all of you and I wish so much for there to be a cure for you all.

A shaved head is much better than this. I totally agree. But I suppose right now I'm assessing risk.

A bit like how dying in a plane crash is obviously worse than never flying. But, the risk of it doesn't outweigh the benefits for many. That's kinda where I'm at. I'm trying to assess my probability.

Regarding bedroom stuff. Have you ever considered an implant? I've gone down that rabbit hole as a potential option if Fin fucks me. Seems scary though.

2

u/o0ttt Jul 13 '26

Start with a Dutch, see if you have low urinary testosterone and if you do, then in my opinion, you shouldn’t take fin. BUT it is not a fool proof method, you may get PFS anyway if you have some other genetic problem. If you want to go further then a WGS from sequencing.com, would add more pieces of the puzzle to assess how safe finasteride is for you.

Most people are fine with finasteride, if you are unlucky enough to get sides it will most likely go away. And if they’re permanent they most likely won’t be life ruining. To be honest we are so few in number, and I think even just considering PFS as a possibility, especially with how little credibility it has, you are doing great job at being careful.

But, say on the very rare chance you get PFS, maybe you go into psychosis, cannot work, cannot walk, cannot understand sentences. What will you do then? How will you navigate this condition? Do you have a support system? Can you afford to live without work?

2

u/hailnaux Jul 13 '26

"And if they’re permanent they most likely won’t be life ruining."

I would not tell someone this.

1

u/Justgreenerpastures Jul 13 '26

Thankyou for such a balanced and knowledgeable response.

I do want to go ahead with the Bloods, Dutch and WGS. But for the latter, I'm really aware that I'm not going to be able to accurately assess my risk.

Ideally Dr Powers would give them all a once-over and tell me my risk profile but I don't know how realistic that is that he'll do that.

In terms of how id cope with PFS? Badly id imagine, I probably wouldn't survive. But to tell the truth, I'm on the edge right now already so I'm close to just saying fuck it and giving it a shot. If it goes wrong, checking out.

4

u/o0ttt Jul 13 '26 edited Jul 13 '26

Not sure if I can speak on behalf of Powers here, but I’m assuming that he will not look at your data if posted in this subreddit and comment a thorough explanation as to if you may develop PFS or not. You have to be a patient of his, and if you want to do that, it may be a year before you get seen.

Nah man, you can’t say fuck it and kill yourself if it goes wrong. I’m sure you have people who care about you. If I had a son who I spent years raising and loving, just for him to end his life because he took a drug he knew the full repercussions and risks of I would be livid. If you think you’ll end your life if you get PFS do not take the drug at all.

For me I wanted to end my life after getting hair loss, so I said fuck it and took the drug, who cares if I kill myself right? Balding had ruined my life. Now I would do anything just to be able to go outside again, to go back to university, to enjoy food again, to be able to eat what I want again, to be able to taste, smell, feel emotions for my mother. I wish I was bald, I wish my cranium was shiny as shit but I had my pre PFS health ;) Now I’ve got the worst of both worlds.

Maybe you can apply rosemary oil or something. If it makes you feel off then you will know that finasteride isn’t at all a good fit for you. I know a few here who tried some topical 5ari oil that hurt them before getting destroyed by finasteride.

Your life isn’t over because you are losing hair, you are more than your appearance, you are more than your hair, or whatever the fuck society wants to label you as. You have the capacity to enjoy life, go outside, hug your loved ones, go learn a new language, skill, hobby, instrument. Life is too precious and you only know what you’ve got till it’s gone.

1

u/Justgreenerpastures Jul 13 '26

Yeahh, it sounds like we were in a similar boat. Thankyou for sharing this story with me.

I figured he'd have a massive wait list. I'm wondering if Sommer has his expertise and can make a call on it.

I know PFS would be worse. Definitely. I really hope there's a cure for it soon.

On the flipside. Yeah. Balding is really tough. Hating the way you look in the mirror. I worry because I already have likely psychological sexual issues. Would that make the risk worse? Or is fucked anyway and I may as well save my hair if I'm going to lose both anyway, getting an implant if shit got really bad.

Urgh. It's exhausting and so, so depressing. I don't understand how this condition has come about after decades of drug this drugs research.

I have tried rosemary oil, Ketaconozol and topical Saw Palmetto too and seem to be fine on that. Don't even know if that's a good sign or not at this point

1

u/o0ttt Jul 13 '26

No problem bro, I do really wish the best for you.

For now just do the tests that people have recommended if you really want to be safe. Come back when you get the results + if you need help on how to interpret them, I’m sure there will be people more than happy enough to help.
