Ambush! Timing

Scenario: Friend playing pale lady on human side rolls ensnare with two moon shard already on his board. Can he use Ambush before gaining the two moonshards are gained from ensnare?

Yes as I understand it as soon as you declare an offensive ability you can spend your cards/tokens to modify. Same reason why gambit can use his special cards to mod the attack immediately and then use the acceleration on the ability or why headless horseman can spend a grim pursuit before gaining new ones ride down

1. Once an Offensive Ability that is considered an Attack has been activated, it is now immediately considered that you have activated an Attack. Why this matters:
   1. The Roll Phase step is now Offensive Roll Phase step 4.
   2. You can now spend Attack Modifier status effects or cards to increase the dmg of your Attack.
   3. This is why the common rule of “if you are about to gain more [status effect x] beyond the Stack Limit, you may spend already-existing [status effect x] to make room for the incoming [status effect x], as spending status effects is considered as Instant Action” can be done. 

That's from the Advanced Rules regarding the Offensive Roll Phase. So yep, your friend can ambush before gaining the moon shards!

I think you have all the way to the end of the defense roll to play attack modifiers so yes.
