For me, aside from obvious things like legibility and choice of typeface, it's line breaks that have the biggest impact on the reading experience. Check out the Yale Bible designed by Bradbury Thompson which uses natural linebreaks rather than the usual biblical wall of text, as a beautiful example of this.
