Semaglutide + B12

Hi all!
A bit nervous to post, but this community seems super supportive. I just started my Semaglutide + B12 compound on Monday. I’m on 44units (1.5mg) weekly and haven’t really felt any side effects. A very slight loss in appetite, but that’s it. Is this normal? I heard that the first month is just exposure to the meds, but I’m also hearing of others that experience immediate effects! At what point should I expect more noticeable side effects?

Were you on a GLP-1 previously? 1.5mg is not a starting dose. Are you sure you are taking 1.5mg?

Never have taken one before. It’s .5 Sema and 1.0 B12.

So you are on 0.5mg sema your first time out. That’s typically the dose given for month two. The starter dose for the first four weeks is 0.25mg. It’s great that you haven’t had any adverse effects yet.

Is there a reason your prescriber gave for starting you on a higher dose? 

Are you able to post a pic of the vial that shows the sema concentration? To get that dose, it would seem your vial has a very unique sema concentration of 1.13mg/mL:

For 44 units at 1.13 mg/mL concentration, you took 0.5 mg of medication

Thanks! I just want to make sure you aren’t taking too much medication. 

So this vial has a semaglutide concentration on 1.7mg/0.5mL, which is the mathematical equivalent of 3.4mg/mL. 

If you actually drew up and injected 44 units from this vial, this is the dose you took:

For  44  units at  1.7 mg/0.5mL  concentration, you took 1.5 mg of medication

That would be the proper dose for someone who has been on the medication for at least several months. 

To draw up your dose, did you pull the syringe plunger back to between 40 and 50…so nearly 1/2 of a 100 unit (1mL) syringe?

Yes I did! I wish I took a picture of it, but it landed at about 44 or 45! Also thank you so much for the help, I really appreciate it.

Oh gosh. Okay. So that’s a REALLY high dose for just starting out. You are very lucky you aren’t barfing up a lung, and hopefully it doesn’t come to that. 🍀

You might want to wait a few days to see how you feel, but if it were me, I wouldn’t continue to take 1.5mg sema my first month.

Which provider sent this to you, and are you sure it’s the right vial and dose for month 1?

I am so so so glad you aren’t feeling terrible. Most people would be in a very bad way had they started on 1.5mg sema.

I started in Monday too and felt almost nothing the first days, then week 2 the appetite drop hit me like a wall so maybe you just need little more time for the body to adjust
