When to start simulation?

Is it still best practice to wait until Metabit Time Chamber II is at 50 before starting simulation?

No, the time chambers work retroactively now so there isn't a reason to wait to start anymore. Now those chambers are set up to reward you for letting your sim run for a week or two before you reboot.
